摘要

透明螨和鼻头螨(蜱螨亚纲:伊蚊科:鼻头螨科)是最重要的体外寄生虫和病原生物媒介。一些透明瘤和鼻头属物种的分类更有争议。利用聚合酶链反应,蜱虫的传统分类已经发生了革命性的变化。PCR的特异性、效率和准确性高度依赖于寡核苷酸引物。本研究设计了5种引物,分别用于扩增ITS2片段TAH-1、TAH-2、TRH(用于透明瘤)和TAH-3、TAH-4(用于鼻头虫)。利用这些引物成功对24和29份ITS2样品进行了测序。基于贝叶斯推理(BI)方法,利用BEAST构建了ITS2系统发育树。由于设计的引物在少数情况下不能扩增基因靶点,因此大多数Hyalomma和Rhipicephalus物种使用ITS2片段分化良好。
