The reconstituted samples were chromatographed on Zorbax XDB C18,4.6*50mm column by using a mixture of acetonitrile -5 mM ammonium acetate buffer (80:20, v/v)as the mobile phase at a flow rate of 0.6 mL/min.The calibration curve obtained was linear (r  0.99) over the concentration range of 0.102-101.389 ng/mL for moexipril. Method validation was performed as per FDA guidelines and the results met the acceptance criteria. 摘要

本研究的目的是建立一种简便、灵敏、选择性的液相色谱串联质谱法定量测定人血浆中莫西普利的含量。利用固相萃取技术,在Waters Oasis®HLB 1 cc (30 mg)萃取筒的帮助下,从人血浆中提取分析物和内标物(稳定标记的同位素)。色谱柱为Zorbax XDB C18,4.6*50mm,流动相为乙腈- 5mm醋酸铵缓冲液(80:20,v/v),流速为0.6 mL/min。莫西普利在0.102 ~ 101.389 ng/mL的浓度范围内,标定曲线呈线性(r = 0.99)。方法验证按照FDA指南进行,结果符合验收标准。每个样品的运行时间为2.2分钟,每天可以分析350多个人类血浆样本。
