Furthermore, a novel approach based on accuracy profile was used to validate the PLS model. The resulting accuracy profile clearly showed that PLS model was able to determine baicalin content, which the LLOQ was 2020 μg/mL. 摘要

本研究旨在建立一种手持式AOTF-NIR测定方法,以准确测定中草药的低剂量含量。以银黄口服液为例,采用实验方案,两名操作人员,独立生产批次进行数据采集。在此基础上,进行偏最小二乘回归(PLS)。该方法的校正标准误差(SEC)和测定系数(R2)分别为146.6 μg/mL和0.9945。在预测步骤中,预测集的预测标准误差(SEP)和R2分别为87.7 μg/mL和0.9923。在此基础上,提出了一种基于精度剖面的PLS模型验证方法。结果表明,PLS模型能够准确测定黄芩苷的含量,定量限为2020 μg/mL。结论:手持式AOTF-NIR测量方法可用于复杂的低含量CHM,为便携式环境下的AOTF-NIR测量铺平了道路。
