Coagulant activity testing, prothrombin time (PT), and activated partial thromboplastin time (APTT) were used to determine procoagulant and anticoagulant properties. For measuring molecular weight, 15% SDS-PAGE electrophoresis with a molecular weight standard ranging from 6.5 to 200 kDa was used.</p><p><strong>Results: </strong>We obtained five fractions named F<sub>1</sub>, F<sub>2</sub>, F<sub>3</sub>, F<sub>4</sub>, and F<sub>5</sub>. The F<sub>1</sub> and F<sub>2</sub> fractions showed procoagulant activity, and the F<sub>5</sub> fraction had anticoagulant activity. The molecular weight of F<sub>2.4.2</sub> from fraction F<sub>2</sub> and F<sub>5.1</sub> from fraction F<sub>5</sub> were analyzed by SDS-PAGE electrophoresis under the reducing condition. These factors were identified as a single protein band at the end of purification. 摘要

背景:棘鱼毒液中含有促凝剂和抗凝剂成分,可促进或阻断血凝级联反应,其中部分成分以不同方式影响血小板功能。目的:建立蛇毒粗毒液的纯化工艺,设计相应的凝血试验,对蛇毒的促凝和抗凝成分进行鉴定。方法:采用凝胶过滤、离子交换色谱、反相高效液相色谱等色谱方法进行纯化。凝血活性测试、凝血酶原时间(PT)和活化部分凝血活素时间(APTT)用于测定促凝和抗凝性能。分子量测定采用15% SDS-PAGE电泳,分子量标准为6.5 ~ 200 kDa。结果:得到F1、F2、F3、F4、F5五个馏分。F1和F2部分具有促凝活性,F5部分具有抗凝活性。在还原条件下,用SDS-PAGE电泳分析了F2.4.2和F5.1的分子量。在纯化结束时,这些因子被鉴定为单个蛋白带。F5.1(b)和F2.4.2(b)纯化后的分子量估计分别为7.5 kDa和38 kDa。结论:本研究结果提示了一种高效、合适的方法,可用于鉴定和纯化伊朗蛇毒的促凝血因子和抗凝血因子。

Isolation of the Anticoagulant and Procoagulant Fractions of the Venom of Iranian Endemic Echis carinatus.

Background: The venom of Echis carinatus contains both procoagulant and anticoagulant components that can either promote or block the blood coagulation cascade, and some of these components affect platelet function in different ways.

Objectives: The present study focuses on setting up a procedure for the purification of crude venom and designing appropriate clotting tests in order to characterize the procoagulant and anticoagulant fractions of E. carinatus venom.
