从透析患者血清中提取的外泌体可促进内皮细胞的内皮-间质转化和钙化
背景:血管钙化(VC)在终末期肾病(ESKD)患者中普遍存在。外泌体是细胞积极分泌的细胞外小泡,含有蛋白质、核酸、脂质和其他生物活性物质,被认为是细胞与细胞相互作用的主要介质。内皮-间充质转化(EndMT)已在多种病理条件下被观察到,如异常剪切应力、血管损伤和慢性炎症。本研究的目的是评估ESKD伴VC患者血清来源的外泌体对内皮细胞中EndMT诱导的影响及其在加速VC中的潜在作用。方法:选取血液透析患者20例,健康志愿者10例。对透析治疗的ESKD患者进行心脏和脑VC评估。透析开始时采集血清样本分离外泌体。人脐静脉内皮细胞(HUVEC)用100µg/mL外泌体处理24-96小时。孵育结束时,收集细胞进行mRNA和蛋白分析。结果:从透析患者VC中分离的外泌体诱导HUVECs的末端mt。24h后,内皮标志物CD31和VE-cadherin分别下降31%和51%;结论:来自VC透析患者血清的外泌体可诱导EndMT并促进钙化。恶性循环突出了外泌体、EC和VC之间复杂的相互作用,强调了治疗策略的关键必要性,以破坏这一途径并减缓钙化进展。
