巨噬细胞炎症蛋白-3α是一种诱导分泌蛋白,通过在多种免疫细胞上的相应受体产生趋化因子,在多种疾病的免疫反应和炎症损伤中发挥重要作用。我们通过RT-PCR获得MaCCL20的ORF,插入表达载体pET-32a中,转化大肠杆菌BL21。经IPTG诱导表达融合蛋白pET-32a-CCL20,制备多克隆抗体,并对其进行Western blot和抗菌活性研究。实验表明,在37℃、0.8 mmol/L IPTG条件下,融合蛋白在4小时内高表达。SDS-PAGE凝胶电泳图显示融合蛋白的相对分子量为30 ku,与之前预测的一致。经His Bind镍柱纯化得到一条单条带,表明MaCCL20重组蛋白纯度高。抑菌实验表明,MaCCL20重组蛋白对金黄色葡萄球菌、粘质沙雷菌、嗜水气单胞菌均有较强的抑菌作用。
