Abstract

Objective To study whether the different concentrations of hydrogen combined with natural killer (NK) cells can contribute to the inhibition of lung cancer cell growth. 目的研究不同浓度的氢与自然杀伤(NK)细胞联合作用是否有助于抑制肺癌细胞的生长。方法从非小细胞肺癌患者外周血中分离培养NK细胞。分别用20%、40%、60%不同浓度的氢气处理NK细胞。采用甲基噻唑四氮唑法测定NK细胞生长曲线,流式细胞术分析NK细胞周期,酶联免疫吸附法分析NK细胞中炎症因子的分泌,以评估氢气对NK细胞增殖的影响。NK细胞和A549细胞(肺癌细胞)以1∶1的比例加入共培养体系,分别用浓度为20%、40%和60%的氢气处理。通过分析A549细胞的克隆形成、迁移和侵袭能力,评价氢与NK细胞结合的杀伤能力。结果氢气浓度分别为20%、40%和60%,对NK细胞增殖影响不大。在A549细胞的克隆形成和迁移能力方面,60%的氢与NK细胞结合对A549细胞的克隆形成和迁移能力有一定的抑制作用,而20%和40%的氢则没有作用。然而,所有浓度的氢与NK细胞结合对A549细胞的侵袭能力没有影响。结论60%氢与NK细胞联合使用可抑制肺癌细胞的克隆形成和迁移能力,抑制肿瘤生长,为后续60%氢与NK细胞联合治疗肺癌的研究提供了框架。关键词:肺癌,非小细胞肺;氢;自然杀伤细胞;细胞增殖;抗肿瘤能力
