The best results were obtained by inflating pulmonary parenchyma with a 1 : 1 mixture of O.C.T. with phosphate buffered saline (PBS) frozen in supercooled n-heptane placed on dry ice. 摘要

通过免疫检测技术对肺组织进行适当的组织学评估和准确的抗原检测,需要对组织进行组织学处理,以保持组织反应性和打开肺泡间隙。在这项研究中,我们着重于测试和比较不同的组织冷冻保存方法。蔗糖或Tissue Tek与几种方法一起用过冷液体和液氮冷冻样品。记录样品冷冻过程中组织的变化以及冷冻保护剂对组织的影响。本实验采用大鼠肺组织和猪肺组织。本研究旨在优化肺冷冻保存的过程,重点是实现适当的解剖评价和保持高组织免疫反应性。用1:1的O.C.T.与磷酸盐缓冲盐水(PBS)混合在过冷的正庚烷中冷冻,放置在干冰上,充气肺实质获得最佳结果。用这种方法制备的肺组织能够进行适当的组织学评估,并通过免疫组织化学分析检测靶分子。
