赤血球膜上で補体反応を抑制する作用をもつシアル酸をニューラミニデース処理により除去すると異種血清補体の反応を受けて溶血反応を起こすようになるが,同種血清補体では溶血反応を起こすようにならない.細胞膜上には自己補体反応を種特異的に抑制する膜分子が存在し,これが働くために同種血清補体の反応を受けない.補体と種特異的補体制御因子の間で認識反応の合致が起これば補体反応が抑制されるという仕組である.そこで,このような膜分子の機能をブロックするモノクローナル抗体があれば,その抗体が作用した赤血球は同種補体によっても溶血反応をおこすようになるとの想定のもとに探索を行い,ヒト赤血球に対する1F5,ラット赤血球に対する512,モルモット赤血球に対するMCA 44等のモノクローナル抗体を得た.1F5に対する抗原分子としてHRF 20/CD 59を同定した.512の反応する抗原分子として512抗原を分離同定し,その機能を解析したところラット補体のC3コンベルターゼのステップを抑制する膜因子であることが判明した.512抗原の遺伝子解析から,マウスの補体制御膜因子として同定されたCrry/p 65と相同の分子であることが明らかとなった.一方,MCA 44の抗原として抽出した44抗原もC3コンベルターゼを抑制する機能を持ち,その遺伝子解析からヒトDAFと相同の分子であることが分かった.512やMCA 44を実験動物に投与してin vivoで補体制御膜因子の機能をブロックするとショック症状やII型アレルギー反応を引き起こすことが立証された.
