T cell acute lymphocytic leukemia (T-ALL) is an aggressive hematologic malignancy in terms of its pathology and populations. It is important to understand the phenomenon of autofluorescence in living cells because normal and cancer cells can be distinguished by this feature. However, the autofluorescence link to T-ALL is poorly understood. We provide direct evidence that EL4-based T-ALL model expresses both high autofluorescence (FL1+) and low autofluorescence (FL1-) in the current study. The FL1+ phenotype is not as stable as the FL1-phenotype. Interestingly, CD25+ cells of FL1+ can transform to CD25+ cells when injected into mice. Autofluorescence associated with differential expression of CD25 is related to the malignant degree of disease, which is presented by the tissue infiltration and growth speed of EL4 cells. Of note, the phenotypes of autofluorescence and CD25 expression of EL4 cells are related to the sensitivity to NK cells killing and the survival time of EL4-treated leukemic mice in vivo. Thus, autofluorescence monitoring might provide as new insight for investigating the malignant degree and prognosis index of T-ALL.

Background: Breast cancer is the most common cancer in women across the world with low survival rates in less developed countries. Low survival rates in less developed countries can be explained mainly by the lack of early detection programs and lack of adequate diagnosis and treatment facilities. Pakistan has high incidence of breast cancer. About one third of all cancer deaths in Pakistan are due to breast cancer. Given that Pakistan is a low resource setting, we aimed to assess the feasibility of breast self-examination followed by clinical examination as a population-based screening tool for breast cancer.

MicroRNAs (miRNA) are potent regulators, controlling multiple biological processes, including cell growth, differentiation, cell death, development and immune responses. With emerging data supporting that miRNAs play a central role in gene dysregulation in human malignancies, unraveling the miRNA genetic variations in cancer is essential and critical if we want to develop better diagnostic and prognostic system for our patients. Oral cancer may have a unique miRNA profiles in different cancer sub-types, which in turn may play a critical role in cancer development, progression, and can therefore be useful for cancer diagnosis and prognosis. Based on our findings and reviewing current literature, this article presents some of the miRNA expression profiles of different cancer tumors to understand its functional relevance and its genetic polymorphism with the oral risk of cancer.

Compartmentalization Modules of Inflammatory Response are Centered on the Epithelial-Mesenchymal Transition of Transforming Cells in Carcinogenesis

The epithelial-mesenchymal transition (EMT) event in carcinogenesis is dependent on multiple operant pathways of master transcription as proposed for NF-kappaB and in terms of the initiated progression of malignant transformation. Inflammation is a primarily compartmentalized series of distinct and overlapping systems that induce and enhance multifocal operabilities within both the nucleus and cytoplasm by systems of enhancer/inhibitory modes of modulation of multi-gene transcription. In terms therefore of a compensatory system of response, carcinogenesis includes a series of steps in characterization of nuclear/cytoplasmic duality targeting ultimately the emergence of tumor cell invasiveness as the epithelial-mesenchymal transition. Emergence of such transition is hallmark for carcinogenesis within contexts of aberrant cell proliferation and anti-apoptosis as exerted by NF-kappaB proinflammation. NF-kB is the main transcription factor that regulates the expression of inflammation-related genes and is in turn influenced by autophagy; also autophagy interacts with inflammation in numerous disease states. It is relevant; in addition, that NF-kB participates in the release of inflammatory cytokines in patients with sepsis with pathogenic implications of sepsis in carcinogenesis.
