Molecular biological analysis of Bt-transgenic (Bt-9) rice and its effect on Daphnia magna feeding 转bt水稻(Bt-9)的分子生物学分析及其对大水蚤取食的影响

Insect-resistant transgenic (Bt-9) rice was generated by inserting mCry1Ac1, a modified gene from the soil bacterium Bacillus thuringiensis, into the genome of a conventional variety of rice (Ilmi). With regard to potential problems such as safety, an evaluation of non-target organisms is necessary as an essential element of an environmental risk assessment of genetically modified (GM) crops. We studied the effects of the Bt-9 rice on the survival of cantor Daphnia magna, a commonly used model organism in ecotoxicological studies. D. magna fed on the Bt-transgenic rice (Bt-9) and its near non-GM counterparts (Ilmi) grown in the same environment (a 100% ground rice suspension). The Bt-9 rice was confirmed to have the inserted T-DNA and protein expression evident by the PCR and ELISA analyses. The feeding study showed a similar cumulative immobility and abnormal response of the Daphnia magna between the Bt-9 rice and Ilmi. Additionally, the 48 h-EC50 values of the Bt-9 and Ilmi rice were 4,400 mg/L (95% confidence limits: 3861.01 5015.01 mg/L) and 5,564 mg/L (95% confidence limits: 4780.03 6476.93 mg/L), respectively. The rice NOEC (No observed effect concentration) value for D. magna was suggested to be 1,620 mg/L. We conclude that the tested Bt-9 and Ilmi have a similar cumulative immobility for D. magna, a widely used model organism, and the growth of Bt-9 did not affect non-target insects.

Variation in the functional compounds of molten salt Kimchi during fermentation 熔盐泡菜发酵过程中功能化合物的变化

To produce a high-quality Kimchi product, molten salt was used for the Kimchi. Changes in the physiochemical properties and functional compounds were analyzed during fermentation. The salinity of bay salt Kimchi was higher than that of the molten salt Kimchi. The fermentation speed of the lactic acid bacteria in the molten salt Kimchi was significantly faster. To evaluate the effects of the salts on the changes in the functional compounds during fermentation, the antioxidant activity, total phenolic compounds (TPC), flavonols, phenolic acids, and glucosinolates in Chinese cabbage were analyzed. In the first 9 days, antioxidants were decreased during this fermentation period and then, increased after that. TPC was slightly increased for all the conditions after 40 days fermentation. Kaempferol was a major flavonol but had a relatively larger decrease in the molten salt Kimchi than in the bay salt samples. Phenolic acid did not show any significant difference among the samples. The glucosinolate contents were significantly decreased in all the conditions of Kimchi during the fermentation period. Consequently, the molten salt greatly affected the fermentation speed of Kimchi and the total characteristics of the Kimchi lactic acid bacteria. Although the functional compounds of Chinese cabbage were decreased during the fermentation of Kimchi, this decrease did not profoundly deteriorate the food quality. Therefore, high-quality Kimchi with enhanced bioactivity will be available if appropriate Chinese cabbages that have enhanced functional compounds are used.

Effect of Optixcell and Triladyl extenders on frozen-thawed sperm motilities and calving rates following artificial insemination in Hanwoo Optixcell和Triladyl扩展剂对韩虎人工授精后冻融精子活力和产犊率的影响

In this study, we examined the effect of a liposome-based extender (Optixcell) and a tris-citric egg-yolk extender (Triladyl) on the frozen-thawed spermatozoa characteristics and the calving rate. The percentages for the total motility of the frozen-thawed spermatozoa were similar in the Optixcell and Triladyl groups. However, among the motile spermatozoa with a straight line velocity (VSL) ≥ 25 μm/sec, the curvilinear velocity (VCL, μm/sec), VSL (μm/sec), average path velocity (VAP, μm/sec), amplitude of lateral head displacement (ALH, μm), beat cross frequency (BCF, Hz), and plasma membrane integrity of the frozen-thawed spermatozoa for the Optixcell group were significantly higher than those for the Triladyl group. Furthermore, the calving rate in the Optixcell group (79.0%) was higher than that of the Triladyl group (62.8%). However, the acrosomal membrane integrity of the frozen-thawed spermatozoa in the Optixcell and Triladyl groups was not significantly different. These results indicate that semen freezing with Optixcell improved the motility and plasma membrane integrity of frozen-thawed spermatozoa and the calving rate of Hanwoo cows (native Korean cattle). In conclusion, our results suggest that semen freezing with the liposome-based extender Optixcell is more efficient than with the tris-citric egg-yolk extender Triladyl for improved offspring production.

The effect of progeny numbers and pedigree depth on the accuracy of the EBV with the BLUP method 用BLUP方法研究了子代数和谱系深度对EBV精度的影响

This study was done to estimate the effect of progeny numbers and pedigree depth on the accuracy of the estimated breeding value (EBV) using best linear unbiased prediction (BLUP) method in Hanwoo. The experiment groups (sire = 100, 200, and 300; progeny = 4 and 8) were made by random sampling and by genetic evaluation of the following traits: Body weight (BW), carcass weight (CW), eye muscle area (EMA), back fat thickness (BFT) and marbling score (MS9). As a result of the genetic evaluation, the accuracy of the EBV was roughly 30 60% with 4 progenies, and the accuracy of the EBV increased by about 50 75% with 8 progenies. In the other words, when the number of progenies increased from 4 to 8, the accuracy of the EBV simultaneously increased by about 15 20%. Moreover, when the number of sires was higher, variations in the accuracy of the EBV within the groups for each trait decreased. Therefore, this result indicates that not only the number of progeny but also the number of sires can affect the accuracy of the EBV. Consequently, collecting information on the progeny and careful management of that information are very important things in the Hanwoo breeding system. Therefore, the EBV can show more precise results when conducting genetic evaluations.

The experiment groups (sire = 100, 200, and 300; progeny = 4 and 8) were made by random sampling and by genetic evaluation of the following traits: Body weight (BW), carcass weight (CW), eye muscle area (EMA), back fat thickness (BFT) and marbling score (MS9). As a result of the genetic evaluation, the accuracy of the EBV was roughly 30 60% with 4 progenies, and the accuracy of the EBV increased by about 50 75% with 8 progenies. In the other words, when the number of progenies increased from 4 to 8, the accuracy of the EBV simultaneously increased by about 15 20%. Moreover, when the number of sires was higher, variations in the accuracy of the EBV within the groups for each trait decreased. Therefore, this result indicates that not only the number of progeny but also the number of sires can affect the accuracy of the EBV. Consequently, collecting information on the progeny and careful management of that information are very important things in the Hanwoo breeding system. Comparison of the change in quality indices during distribution period by import season in three grape cultivars 三个葡萄品种不同进口季节配销期品质指标变化的比较

As a result of analyzing the quality of imported grapes during the 2018 season, the hardness of the grape berry was found to be 10 N or less in total. In the case of Chilean grapes, the soluble solids tended to be lower as imports and distribution periods were delayed. The berry weight was the largest at 14.4 - 14.8 g for the 'Red Globe', 7.1 - 7.4 g for the 'Thompson Seedless' and 6.0 - 7.0 g for the 'Crimson Seedless'. The 'Crimson Seedless' grapes imported from Chile, which had a high berry shatter rate, had a shorter pedicel length, pad width and brush length than that of the other 2 varieties. Regardless of the grape varieties, the weight loss during the distribution at room temperature ( $25^{circ}C$ ) increased rapidly as the import season was delayed. Additionally, it was found that the later the distribution seasons, a higher stem browning index was observed regardless of the variety. The 'Thompson Seedless' was revealed to be more susceptible to browning than that of the other varieties. The shrinkage of the stem and the pedicel browning progressed within a short time during shelf-life as the distribution time was delayed. The incidence of berry decay of the imported grapes tended to increase with the progression of the distribution period. In the fruits distributed on April, except for the 'Crimson Seedless', the grapes were rapidly corrupted in the second half of the distribution. The 'Red Globe' grapes completely lost marketability due to a berry decay of 26.1% and 69.9% at 9 and 12 days after shelf-life, respectively.

The SNP of WBP1 is associated with heifer reproductive performance in the Korean native cattle Hanwoo WBP1的SNP与韩国本土牛韩雨的小母牛繁殖性能有关

It is well documented that intensive selection in dairy cattle for economic value such as increased milk yield led to a decline in reproductive performance. Recent studies using genome-wide association studies (GWASs) discovered candidate genes involved in the lower fertility including embryo development and conception rates. However, the information, which showed a lower reproductive performance, is limited to dairy cattle, especially Holstein, and the candidate genes were not examined in the Korean native cattle Hanwoo which has been intensively selected and bred for meat in the last few decades. We selected the candidate genes WBP1 and PARM1 reported to be associated with cow and/or heifer conception in dairy cattle and analyzed the genotype because those genes have nonsynonymous single nucleotide polymorphisms (SNPs). To determine the single base change, we used the high resolution melting (HRM) assay which is rapid and cost-effective for a small number of genes. We found that most heifers with higher conception (1: service per conception) have the AA genotype coding Threonine rather than Proline in the WBP1 gene. We did not detect an association for a SNP in PARM1 in our analysis. In conclusion, the genetic variation of WBP1 can be used as a selective marker gene to improve reproductive performance, and HRM assay can be used to identify common SNP genotypes rapidly and cost effectively.
