Objective: To investigate the regulation of HPA axis and nephrotoxicity of different processed products of Psoraleae Fructus in rats with kidney-yang deficiency. Methods: Hydrocortisone was injected intramuscularly to create a model of kidney-yang deficiency, and the drug was administered for 15 d. ELISA was used to determine the content of 17-OHCS, CRH, ACTH, and CORT in serum, and HE staining was used to observe the adrenal tissue lesions and morphology; ELISA was used to determine the content of Kim-1 in serum and 24 h urine, and HE staining was used to observe the renal histopathology and morphology. Results: Compared with the model group, the body weight of rats in each group of different processed products of Psoraleae Fructus increased, but there was no significant difference ( P > 0.05); the liver index and serum levels of 17-OHCS and CORT of the rats in each group of different processed products of Psoraleae Fructus were higher than those of the model group ( P < 0.05), and the inflammatory lesions of the adrenal tissues were improved on different degrees; the renal index and adrenal index of the wine-made group and the renal index of salt-roasted group were significantly higher than those of the model group ( P < 0.05); the adrenal index of the salt-roasted group was higher than that of the model group, but there was no significant difference ( P > 0.05); compared with the blank group, the renal tubules of each group of different processed products of Psoraleae Fructus were slightly dilated, and the lesions of the wine product were more serious; the blood Kim-1 level of the tonic fat group was significantly higher than that of the blank group ( P < 0.05); blood and urine Kim-1 levels (except for blood Kim-1 in the salt-roasted group) were significantly higher than those of the blank group in the processed products of Psoraleae Fructus ( P < 0.05). Conclusion: Different processed products of Psoraleae Fructus can improve the symptoms of kidney-yang deficiency and regulate the HPA axis in rats, but the specific mechanism of action has some differences; different processed products of Psoraleae Fructus still retain a certain degree of nephrotoxicity, but the toxicity of salt-roasted tonic bone resin is lower.

Objective: To provide a reference for clinical administration of trametinib (TRA), we mine the TRA-related adverse event (ADE) of respiratory system based on the FAERS and explore the characteristics of ADE occurrence, the correlation strength between ADE and TRA and the risk factors of TRA-related ADE of nervous system. Methods: We use reporting odds ratio (ROR) methods and healthcare products regulatory agency (MHRA) methods to mine the TRA-related ADE of nervous system based on the adverse reaction report data of 24 quarters from the first quarter of 2017 to the fourth quarter of 2022 extracted from the FAERS. Results: Totally 4 392 TRA-related reports were obtained, 86ADE of nervous system were reported. There were 15 PT with signal, which could be divided into 7 high level group term (HLGT). The top 3 reported PT were seizure, syncope, sleepy. The top 3 PT with the highest signal intensity were central nervous system lesion, hemiparesis and hydrocephalus. There were 15 PT that were not included in the drug instructions. Conclusions: Need to attach importance to TRA related nervous system the risk of ADE, especially the peripheral neuropathy and hydrocephalus, patients with underlying diseases related to the nervous system need to be closely monitored to ensure the safety of medication.

Objective: To observe the effects of Quanzhen Yiqi Decoction on the expression of LASP1 and CXCR1 in lung tissue of chronic obstructive pulmonary disease (COPD) rats, and to explore the effects of Quanzhen Yiqi Decoction on inflammation and immune function of COPD. Methods: Eighteen SD rats were randomly divided into Chinese medicine group and model group, with 9 rats in each group. COPD rat model was established by smoking combined with lipopolysaccharide (LPS). The model group was given 0.9% normal saline daily. The traditional Chinese medicine group was given 40% Quanzhen Yiqi Decoction concentrated decoction every day. The mrna levels of LASP1 and CXCR1 in rat lung tissue were detected by fluorescence quantitative PCR, and the protein levels of LASP1 and CXCr1 in rat lung tissue were detected by Western blot. Results: The mRNA expression abundance of LASP1 in the Chinese medicine group was 0.145 times that in the model group. The mRNA expression abundance of CXCR1 in the TCM group was 0.614 times of that in the model group. Compared with model group, the protein expression levels of LASP1 and CXCR1 in TCM group were significantly decreased. Conclusions: Quanzhen Yiqi Decoction can inhibit airway inflammation in COPD rats by down-regulating the expressions of LASP1 and CXCR1 genes and proteins, which may be one of the potential mechanisms to reduce the risk of lung cancer caused by COPD.
