The 7000 g pellet of homogenized mature sponge-induced granulation tissue was fractionated by centrifugation in a stepwise sucrose gradient in order to study the synthesis and secretion of collagen and other components of this tissue on the subcellular level. As indicated by chemical and enzymatic assays, by electron microscopy and by incorporation experiments, the collagen-synthethesizing rough endoplasmic reticulum fraction was isolated free from the secretory vesicles (smooth endoplasmic membranes and Golgi elements) and fibrous extracellular matrix. Collagen differed from other proteins in the distribution among the subcellular fractions. In pulse-chase experiment the translocation of 14C-labelled collagen was demonstrated from the rough endoplasmic reticulum through the secretory vesicles to extracellular fibrillar collagen. This fractionation method will be used to study the modulation of collagen synthesis and secretion in the reparative tissue.

The relative configuration of 3-hydroxy-3-isobutyl-2-pyrrolidone-5-carboxylic acid has been determined by an X-ray crystal structure analysis. The compound crystallized in space group P212121 with two molecules in the asymmetric unit. a=16.023(3) A, b=19.349(7) A, c=6.9053(16) A. The structure was solved by direct methods using MULTAN and refined by full-matrix least-squares technique to an R of 0.137 for 645 diffractometer-collected intensities. The absolute configurations of the title compounds were deduced; the configuration of the amino acid and the lactam are 2(S), 4(S) and 3(S), 5(S), respectively.

The crystal and molecular structure of the meso-ionic title compound, C13H10N4O, have been determined by X-ray methods using 2667 reflections above background level collected by counter methods at 105 K. The crystals are monoclinic, space group P2a/c, with cell dimensions: a=5.233(2) A; b=18.846(6) A; c=11.643(2) A, beta=105.71 degrees(2), with 4 molecules per unit cell. The structure model was refined to an R-factor of 0.047. In order to reduce the influence of the valence electrons all reflections with sin theta/lambda smaller than 0.5 A- minus 1 were excluded in the last stages of the refinement procedure (leaving 1658 Fo'S, R=0.058). The central five-membered ring is found to be planar and the bond lengths within this ring indicate a resonance stabilized system, as has been found for sydnones. The planes of the two benzene rings are tilted 11.7 and 8.0 degrees, respectively, with respect to the plane of the central oxatriazolio-ring.?
