Somatic genomic rearrangements in human leucocyte antigens region in solid ovarian tumors 实体卵巢肿瘤中人白细胞抗原区体细胞基因组重排

The success of immunotherapy for ovarian cancer is determined by the effectiveness of the patient’s immune response, as well as the qualitative and quantitative representation of antigens of the main histocompatibility complex in tumor cells and microenvironment. The level of the HLA (Human Leukocyte Antigens) genes expression in tumor tissue does not always correctly reflect the expression of these genes in different populations of tumor cells. Aim. To analyze the frequency and range of somatic deletions and duplications in the HLA locus. Methods. LOH-analysis (Loss of Heterozyhosity) of the STR-markers (Short Tandem Repeats) in the ovarian tumors of affected patients. Results. We have shown that complex somatic genomic reorganizations, involving several HLA loci, are quite common in ovarian tumors. The detected genetic alterations vary by both the type (deletions and mutations in STR alleles) and proportion of the sub-population of cells with the alterations. Conclusion. The results obtained allow us to propose the LOH-analysis for highly effective detection of aneuploidy and different types of somatic genomic reorganizations in the HLA region even in minor cell populations. This analysis provides the additional prognostic information for predic ting the effectiveness of different types of ovarian cancer immunotherapy at the indi vidual level.

Antimicrobial and cytotoxic activities of thiazolo[4,5-b]pyridine derivatives 噻唑[4,5-b]吡啶衍生物的抗菌和细胞毒活性

Aim. The screening of antimicrobial and cytotoxic activities of thiazolo[4,5- b ]pyridine derivatives was accomplished. Methods. The antibacterial and antifungal activities of synthesized thiazolopyridines were evaluated in vitro with the agar diffusion and broth microdilution methods using clinical and reference strains of Gram-positive, Gram-negative bacteria and yeasts. The structure-antibacterial/antifungal activity relationships of the screened compounds were established. The target compounds were screened for their cytotoxicity effects on HaCaT and HEK293 cells using MTT assay. Results. The highest antimicrobial activity was observed for compound V 2-oxo-7-thiophen-2-yl-2,3-dihydrothiazolo[4,5- b ]pyridine-5-carboxylic acid with minimal inhibitory concentration (MIC) 12.5 μg/mL against Candida albicans . At the same time, the synthesized compounds were explored in the interaction with amoxicillin against multidrug resistant clinical isolates of ESβL + Klebsiella pneumonie and Staphylococcus haemolyticus (MRSH). The best synergistic activity with amoxicillin was exhibited by compound VI. HaCaT human keratinocytes and HEK293 human embryonic kidney cells demonstrated resistance to the thiazolopyridine derivatives treatment and did not reach the IC 50 value up to 100 µM. Conclusions. The tested thiazolopyridines constitute an interesting background for further development of new chemotherapeutic agents.
