Aim To analyze the clinical characteristics of patients of corona virus disease 2019 (COVID-19), and summarize the treatment experience, aiming to provide diagnostic and treatment reference for the front-line clinicians Methods Seventy-nine patients with COVID-19 admitted to the Infectious Hospital of Anhui Provincial Hospital from January 22 to February 18, 2020 were selected as the research subjects There were 55 cases in general group and 24 cases in severe and critical group The clinical data of the two groups were collected and compared, including general conditions, clinical symptoms, signs, laboratory tests, computed tomography imaging of the lungs and complications Results The average age of 79 COVID-19 patients was 45 1 ± 16 6 years, and forty-five of them are males The severe and critical group was older than the general group Besides, there were more males and comorbidities In terms of laboratory tests, the lymphocyte (LYM) count and albumin (A L B) decreased more significantly in the severe and critical groups Moreover, the percentage of neutrophils (N E U), c-reactive protein (CRP), D-dimer, lactate dehydrogenase (L D H), troponin I (cTnl) and urea nitrogen (BUN) increased significantly Among all the patients, the types of antiviral drugs in severe and critical group were significantly more than those in general group;moreover, the glucocorticoids, gamma globulin and oxygen inhalation by nasal catheter were used more frequently in severe and critical patients By the time of February 18, one patient died of acute large area cerebral infarction, 34 patients were discharged from hospital, and the rest were still receiving treatment Conclusions The elderly patients with multiple cardiovascular diseases are more likely to get severe and critical COVID-19 They usually combine multiple organ or system abnormalities Clinicians should make timely judgment and adjust the treatment plan according to the clinical symptoms, signs and laboratory examination results
目的分析2019冠状病毒病(COVID-19)患者的临床特征,总结治疗经验,旨在为一线临床医生提供诊断和治疗参考方法1月22日至2月18日安徽省立医院感染性医院收治的79例COVID-19患者,2020年被选为研究对象一般有55例组,24例严重和重要组两组临床资料的收集和比较,包括一般情况、临床症状、体征、实验室测试、计算机断层扫描成像的肺部和并发症结果79 COVID-19患者的平均年龄是45 1±16 6年,其中45是男性严重和重要组比一般群体之外,在实验室检查方面,重症和危重症组淋巴细胞(LYM)计数和白蛋白(A L B)下降更为明显,中性粒细胞(N E U)、c反应蛋白(CRP)、D-二聚体、乳酸脱氢酶(L D H)、肌钙蛋白I (cTnl)和尿素氮(BUN)百分比均显著升高。重症、危重症患者使用抗病毒药物的种类明显多于普通组,且重症、危重症患者使用糖皮质激素、丙种球蛋白、鼻导管吸氧的频率更高。截至2月18日,1例患者死于急性大面积脑梗死,34例患者出院。结论老年合并多种心血管疾病的患者更易发生重症、危重型新冠肺炎,多合并多器官或系统异常,临床医生应根据临床症状、体征及实验室检查结果,及时判断并调整治疗方案

Intracellular levels of ubiquitous transduction signal cyclic adenosine 3',5'-monophosphate( cAMP) and cyclic guanosine 3',5'-monophosphate( cGMP) are partly determined by the hydrolysis by phosphodiesterases( PDEs). So far,the superfamily of PDE is composed of at least 11 isoforms and each family is characterized with unique cellular distribution,expression,regulation mode and inhibitor sensitivity,and PDEs are considered to be involved in a variety of pathological processes, such as inflammation,asthma,depression,and erectile dysfunction,etc. Currently,these characteristics draw PDEs more and more attention as great targets for drugs. In this review,the basic biochemical properties,cellular regulation,expression pattern,physiological functions and inhibitors of different PDE isoforms will be briefly discussed.

Aim To investigate the immune regulation of IL-23 fusion gene and the correlation between its biological function and the airway inflammation in asthma during pregnancy. Methods The eukaryotic expression plasmid,which was constructed to express the IL-23p40 + p19 double subunit,was used to transform E.coilBL21 bacteria,and the expression products,which were induced by IPTG,were purified and detected by SDS-PAGE and Western blot. The transfection,mediated by recombinant protein,was identified by fluorescence microscopy and plasmid's Western blot,while DNA binding activity of nuclear proteins was measured by gel electrophoresis. To evaluate the impact of foreign protein in vivo,mouse serum inflammatory cytokines levels and inflammation injury in lung tissue were measured by ELISA method and HE staining. The CD4+T cell subset's proportion of mononuclear cells in peripheral blood was verified by flow cytometry. Results The results showed that soluble exogenous recombinant protein,which was obtained successfully,had the ability of trans-membrane transport and could effectively mediate EGFP into the target cells. Its stable expression and transcriptional activity were higher than those of simple plasmid's effect. The specific binding between GST/pCEP4-IL-23 and plasmid DNA formed macromolecular complexes,which could decrease the mobility of gel. The combined effect with LPS significantly increased the IL-17A levels in serum(P 0. 05) and the inflammation reaction in lung tissue of sensitized mice. Mean while,it also reduced the IFN-γ,Foxp3 level in serum,promoted the activation of IL-4 and IL-17A,and then increased the ratio of Th2/Th1and Th17/Treg(P 0. 05). Conclusions The recombinant IL-23 protein is pathogenic,and it adjusts the inflammation of asthma in pregnancy. The immune imbalance of CD4+T subsets affects the immune tolerance of pregnant body and the development of asthma during pregnancy.

Aim To explore the reversal of MDR(multidrug resistance) by nemadectin and milbemycin β1 from the milbemycin family to adriamycin-resistant human breast carcinoma cells(MCF-7 / adr). Methods MTT assay was used to evaluate the cell suppression rates and reversal effects. Intracellular accumulation of adriamycin was determined by HPLC. The accumulation of rhodamine 123(Rh123) was measured with spectrofluorometer. The expressions of P-gp and the MDR1 gene encoding P-gp were tested by flow cytometry and reverse-transcriptase PCR. Results Nemadectin and milbemycin β1at the high dose of 5 μmol·L- 1significantly increased the sensitivity of MCF-7 / adr cells to adriamycin. They also increased the intracellular accumulation of adriamycin and Rh123 in MCF-7 /adr cells in a dose-dependent manner. Expression of both P-gp and MDR1 was down-regulated to different level. Conclusion Nemadectin and milbemycin β1can partly reverse multidrug resistance of MCF-7 / adr,and the effect of nemadectin is better than that of milbemycin β1.

Aim: To explore the underlying mechanism of hyperthyroidism induced by subcutaneous injection of L-levothyroxine sodium and and intervention effects of Liuwei Dihuang pills on sprague dawley rats via serum metabolomics. Methods: After rats were injected with L-levothyroxine sodium at a dosage of 35 μg·(100 g)-1 every day for 1 week,and then Liuwei dihuang pills(LDPs) were treated at a dose of 4.7 g·kg-1 for 3 consecutive weeks.The serum samples for rats were obtained after the treatment of LDPS and the metabolic profiles of serum were obtained with ultra performance liquid chromatography coupled with time-of-flight mass spectrometry(UPLC-QTOF-MS). Results: The levels of oxaloacetic acid,Dihydroxyacetone phosphate(DHAP),ethyl glucuronide,triglyceride,lysophospholipids and cholesterol were significantly increased in the model group as compared to the normal rats.Moreover,these levels could be regulated to be close to normal after the treatment of LDPS. Conclusions: There is severe disturbance in the tricarboxylic acid cycle as well as sugar and lipid metabolism for hyperthyroidism rats.LDPS can exhibite obvious effects on the treatment of hyperthyroidism.Serum metabonomics analysis reveals the changes of metabolites traits during the treatment of LDPS,thus providing a supplementary evaluation method of LDPS in the treatment of hyperthyroidism.
