Candida albicans and Candida tropicalis antigens studied by SDS polyacrylamide gel electrophoresis and Western blot. 采用SDS聚丙烯酰胺凝胶电泳和Western blot技术研究白色念珠菌和热带念珠菌抗原。
Pub Date : 1987-06-01 DOI: 10.1111/j.1439-0507.1987.tb03979.x
S M Bruneau, R M Guinet
Summary: The SDS extracts of blastospores from 14 Candida albicans strains and from 8 Candida tropicalis strains were studied by SDS polyacrylamide gel electrophoresis. The Coomassie blue staining revealed more than 40 polypeptides; 4 from Candida albicans and 5 from Candida tropicalis were major polypeptides with molecular weights (M.W.) of 34 to 54 kDa. The extracts obtained by the mechanical disruption of yeasts cells and those obtained by the SDS whole‐cell treatment showed several differences especially with polypeptides ranging from 67 to 94 kDa and from 20 to 30 kDa – the major polypeptides were extracted by the both procedures. All the gels run with the SDS extracts from the strains of same species strains gave identical patterns but several polypeptides including the major ones showed different M.W. when extracted from Candida albicans or Candida tropicalis. However the antigens of these two species previously studied by quantitative Immunoelectrophoresis techniques (QIE) had shown more than 85% of cross‐reactions. (Gabriel‐Bruneau and Guinet 1984 [5]).

Clinical and immunological follow-up of trichophytic kerion celsi. 毛癣性角质细胞的临床及免疫学随访。
Pub Date : 1987-06-01 DOI: 10.1111/j.1439-0507.1987.tb03974.x
M Papini, S Simonetti
Summary: Eighteen patients who had been affected by trichophytic Kerion Celsi within the past 10 years, and 18 control subjects, were investigated for immunofluorescent specific antibodies directed against the responsible mycetes and trichophytin skin reactivity. Significant titers of humoral antibodies and intensely positive skin reactions were observed in subjects whose infection pre‐dated the study by 1–5 years. The persistence of specific immunological memory could be a contributing factor to resistance to relapses in these patients.

Inhibitory effect of antifungal agents on germ tube formation in Candida albicans. 抗真菌药物对白色念珠菌芽管形成的抑制作用。

Summary: The allylamine derivatives naftifine and terbinafine were compared in vitro with 15 standard antifungals in terms of their inhibitory effects on the yeast‐mycelial (Y‐M) transformation in tests with 19 Candida albicans isolates. Y‐M transformation minimal inhibitory concentration (MICs) were determined in a microtiter‐broth‐dilution test using a chemically defined medium with N‐acetyglucosamine as inducer of hyphal growth. Clotrimazole was the most potent inhibitor with a geometric mean MIC (G‐MIC) value of 0.00038 μg ml‐1 followed by ketoconazole (G‐MIC,0.012 μg ml‐1) and itraconazole (G‐MIC,0.027 μg ml‐1) in that order. G‐MIC values ranging from 0.027–0.037 μg ml‐1 have been determined for itraconazole, econazole, amorolfine (RO 14–4767/002), miconazole and 5‐fluorocytosine. Among the orally active azole derivatives fluconazole was the least active (G‐MIC,0.14 μg ml‐1). Noteworthy activity included that of the allylamine terbinafine which was similar to that of bifonazole (G‐MIC 0.23–0.26 μg ml‐1). There was a statistically significant higher susceptibility of the test strains with terbinafine than with amphotericin B. Naftifine and amphotericin B were comparable active in terms of G‐MIC values (0.52–0.7 μg ml‐1). Significantly higher drug concentrations (> 1 μg ml‐1) were necessary to inhibit filamentation of C. albicans with ciclopiroxolamine, nystatin, griseofulvin and tolciclate. Tolnaftate was the least active drug tested with a G‐MIC of 74.7 μg ml‐1.
