Today, the use of mesenchymal stem cells (MSCs) for treating human diseases has attracted wide attention. The aim of this study is the expression of dopaminergic genes such as Nestin, patched Tumor Suppressor (PTCH), Sonic Hedgehog (SHH), Tyrosine Hydroxylase (TH) and Nuclear receptor-related factor 1 (NURR1) in MSCs after induction with selegiline. Rat bone marrow mesenchymal stem cells (rBMSCs) were extracted from femur and tibia bones and incubated with alpha Minimum Essential Medium (α-MEM) and 10% Fetal bovine serum (FBS). The stemness of cells at passage 4 was determined by the positive response to CD71 and CD90 markers and their differentiation into adipocytes and osteoblasts. The expression of SHH, PTCH, TH, NURR1 and Nestin genes in the cells after induction by 10-8 M selegiline for 48 hours was investigated by Reverse transcription polymerase chain reaction (RT-PCR) and Real Time-PCR methods. Isolated rBMSCs expressed CD71 and CD90 markers in culture conditions and could differentiate into adipocytes and osteoblasts. Induced cells showed neuronal morphology, positive response to Nestin and TH immunostaining. There was a significant increase of dopaminergic genes TH and NURR1 compared to the untreated cells. The results showed that selegiline with a dose of 10-8 M for 48 hours can lead to dopaminergic differentiation in rBMSCs.

In the current study, we investigated the effect of creatine monohydrate (CrM) and newly synthesized creatine lysinate (CrLys) in tail suspension (TST) and rotarod tests and their influence on the histology of the skeletal muscles. In the TST, a slight decrease in the immobility time from the 1 st to the 3 rd week was observed in the group treated with CrM at a dose of 1.5 g/kg/day and CrLys at a dose of 6 g/kg/day. The rotarod test revealed that CrM (1.5 g/kg/day) and CrLys (3 g/kg/day) lead to a significant improvement in motor coordination in the 3 rd week. The results from histology showed an increase in the muscle fiber diameter of soleus muscle in animals treated with CrM (3 g/kg/day) and CrLys (6 g/kg/day). The results showed that supplementation with creatine derivatives appears to be a generally effective nutritional ergogenic aid for an improvement of physical performance.

Individual NAT2 genotype identity data should be enriched to prevent Isoniazid-induced liver injury (IDILI) and optimize the dose of Isoniazid (INH). Therefore, this study aims to present the level of IDILI risk for specific genotype alleles. The data collection involves literature indexed by Google Scholar, Scopus, and Pubmed databases. The search uses a combination of the following keyword variants “INH” OR “INH”, “liver injury” OR “hepatotoxicity”, “polymorphism” OR “pharmacogenomic”, and “N-acetyltransferase 2” OR “NAT2”. Furthermore, the screening results of library sources were narrowed to 11 original articles that met the inclusion criteria. The IDILI risk assessment analysis due to NAT2 enzyme polymorphism following the odds ratio has a 95% confidence interval. The results showed that the IDILI risk level of the slow acetylator group was 3.11 times higher than other populations. Meanwhile, the rapid and intermediate acetylator groups were not at risk. Three variants related to *6 allele were classified as high risk; *6A/*6A risk 5.76 times, *6A/*7B (5.54 times), and *6/*7 (4 times). The three allele configurations of the *5 and *7 were also classified as a risk; *5B/*7B (5 times), *7B/*7B (3.23 times), and *5/*7 (2,74 times).

This study aims to assess the hypoglycemic effects of Ngu-Vi-Tieu-Khat (NVTK) decoction, a traditional Vietnamese medicine, in a rat model of type-2 diabetes. The NVTK decoction was prepared using the maceration method and tested for its hypoglycemic effects by measuring blood glucose levels, insulin resistance indicators, and pancreatic mass. The results showed that NVTK decoction improved diabetes symptoms, increased insulin levels, reduced insulin resistance, restored pancreatic mass, and decreased total cholesterol (TC), triglycerides (TG) and low-density lipoprotein cholesterol (LDL-C) while increasing high-density lipoprotein cholesterol (HDL-C). The hypoglycemic effects of NVTK were comparable to those of gliclazide at a dose of 10 mg/kg. In conclusion, NVTK decoction possesses hypoglycemic properties and could be explored as a potential traditional medicine for treating type-2 diabetes in humans.

Headaches, especially migraines and tension-type headaches (TTH), are among the most prevalent problems encountered in neurology and medicine. Ongoing debate surrounds the pathophysiological mechanisms behind this condition. One of the potential ideas involves inflammation and diet. The purpose of the study was to compare the serum levels of C-reactive protein (CRP), D-dimer, vitamin D and magnesium in 23 migraineurs and 23 TTH-diagnosed sufferers. The CRP level (4.8±3.7 mg/L) was found to be significantly different between migraineurs and TTH sufferers, with a significant difference (p=0.03) between the two groups of headache sufferers. Vitamin D serum levels (19.5±7.9 ng/L) and TTH serum levels (26.7±8.4) were observed to differ significantly. Magnesium serum levels (p=0.692) did not differ significantly between the two headache patient groups.

The colorimetric paper-based device using lipase inhibition assay was developed for rapid and visual detection of orlistat in weight loss supplements. The paper device with five circular detection zones was simply fabricated from filter paper No 1 using a low-cost paper craft puncher with a design of a flower-like shape. The enzymatic reaction on the detection zone employed a small volume of a substrate, a-naphthyl acetate, a sample, and a lipase enzyme. After incubation, the Fast Blue B solution was used as a chromogenic reagent. The decrease in purple color can be observed by the naked eye, in the presence of orlistat. Under optimized conditions, the paper device showed satisfactory sensitivity and selectivity. The device was applied for rapid screening of orlistat in weight loss supplements and the results agreed with those obtained from TLC analysis.

Background : Burnout is described as an emotional, mental and physical exhaustion observed in healthy people and directly related to their professional context. The objective is to study the level of burnout among community pharmacists and to identify the workplace factors affecting it. Materials and methods : Using a questionnaire, specifically designed by the authors of the study, an anonymous survey was conducted in the region of Varna, as the largest region of Northeastern Bulgaria. The questionnaire contains questions about the demographic characteristics; a standardised burnout measure tool (MBI-HSS-MP), and questions regarding the specifics of the work environment. According to MBI-HSS-MP, burnout is associated with high levels of emotional exhaustion (EE) and depersonalisation (DP) and low level of personal accomplishment (PA). Results : The participants in the pilot study were 127 masters of pharmacy. Regarding individual factors, a statistically significant relationship was found between age and the scales emotional exhaustion EE (p = 0.012), depersonalisation DP (0.028) and personal accomplishment PA (p = 0.028). Most susceptible to ЕЕ (p = 0.014) and DP (p = 0.008) were the age group 25–45 years while most satisfied with their PA were in the age group 46–55 years (p = 0.016). From the organisational factors, the large number of patients had the greatest impact on burnout (DP р = 0.000 /ЕЕ р = 0.006). The other key factors for burnout were an insufficient number of pharmacy staff, a heavy administrative burden and a wide range of medicinal products. Conclusion : It is necessary to develop training courses for young professionals to introduce them gradually into the work process and build up skills for effectively coping with stress. Proposals need to be made for reducing the administrative burden of pharmacists and maintaining staffing levels that are adequate to the patient flow and expanding the capabilities of The Specialised Electronic System for Tracing and Analysis System of Medicinal Products (SETASMP).

Background : Atrial fibrillation (AF) is the most common heart rhythm disorder. AF ablation based on endovascular or surgical techniques is a standard of care in everyday practice. Early recurrence of atrial arrhythmias is a significant problem after ablative procedures. Prophylactic use of antiarrhythmic drugs has become a possible solution. Materials and methods : We performed a retrospective observational cohort study of 59 patients with heart disease requiring surgery under extracorporeal circulation (ECC) and have received radiofrequency ablation (RFA) for chronic or persistent AF with a successful conversion to sinus rhythm. Patients were divided into two groups: 36 were treated with amiodarone for three months postoperatively (treatment group), and 23 had no concomitant amiodarone therapy (control group). We compared the groups regarding freedom from recurrent atrial arrhythmia and the frequency of conduction disorders requiring permanent pacemaker implantation. Results : No differences between the groups were found regarding demographic characteristics, risk profile, and heart diseases indicating cardiac surgery. During follow-up, a significantly higher proportion of patients with sinus rhythm in the group receiving amiodarone was found (80.56% vs. 52.17%; p < 0.041). No cases of premature discontinuation of amiodarone for side effects or noncompliance were registered in the treatment group. Conclusion : Three months of amiodarone therapy post-surgical RFA of AF is an effective and safe strategy for relapse prevention.

AF ablation based on endovascular or surgical techniques is a standard of care in everyday practice. Early recurrence of atrial arrhythmias is a significant problem after ablative procedures. Prophylactic use of antiarrhythmic drugs has become a possible solution. Materials and methods : We performed a retrospective observational cohort study of 59 patients with heart disease requiring surgery under extracorporeal circulation (ECC) and have received radiofrequency ablation (RFA) for chronic or persistent AF with a successful conversion to sinus rhythm. Patients were divided into two groups: 36 were treated with amiodarone for three months postoperatively (treatment group), and 23 had no concomitant amiodarone therapy (control group). We compared the groups regarding freedom from recurrent atrial arrhythmia and the frequency of conduction disorders requiring permanent pacemaker implantation. Results : No differences between the groups were found regarding demographic characteristics, risk profile, and heart diseases indicating cardiac surgery. During follow-up, a significantly higher proportion of patients with sinus rhythm in the group receiving amiodarone was found (80.56% vs. 52.17%; p < 0.041). No cases of premature discontinuation of amiodarone for side effects or noncompliance were registered in the treatment group. Conclusion : Three months of amiodarone therapy post-surgical RFA of AF is an effective and safe strategy for relapse prevention.","PeriodicalId":20086,"journal":{"name":"Pharmacia","volume":"29 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-09-28","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135425154","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
This scoping review aimed to examine the existing evidence on complementary and alternative medicine interventions for preeclampsia, emphasizing empowering midwives in postpartum treatment. Three databases (PubMed, Web of Science, Scopus) were used following the PRISMA-2020 guidelines to find articles from January 2012 to December 2022 and synthesized using Rayyan AI software. A total of 707 articles were initially identified, 7 were included in the review. Traditional Chinese Medicine combined with Western medicine showed promise in treating severe preeclampsia. Massage therapy reduced anxiety, aromatherapy positively affected postpartum well-being, and warm foot therapy reduced swelling associated with postpartum preeclampsia. Using herbal ingredients, lavender aromatherapy, magnesium sulfate, massage, and physical therapy contributed to successful postpartum preeclampsia management. Complementary and alternative medicine, collaboration with community health workers, clear guidelines, targeted training, and future research are crucial for effective postpartum preeclampsia management.
