Protein patterns of Streptomyces griseus strains during development. 灰色链霉菌菌株在发育过程中的蛋白质模式。

Protein patterns from mycelial extracts of Streptomyces griseus strains No. 45H and No. 52-1 were studied by one dimensional gradient PAGE with 20 cm run. The results are reproducible, the protein patterns are the same for the same developmental stage in a given strain. There are well-defined characteristic changes of the protein patterns during the life cycle. The proteins of spores show conspicuous differences compared to protein patterns of the old (72 h) mycelia in the same culture. There is no difference between protein patterns of spores from submerged and from solid media. The protein patterns of two closely related Streptomyces griseus strains significantly differ from each other. After pulse labelling with (14C)-labelled protein hydrolyzate for 40 min, the specific activities of the proteins show considerable differences. There are characteristic bands with low specific activity and others with high incorporation. The fluorograms of the 16, 40 and 64 h cultures show that different proteins are being synthesized intensively at different ages of the culture.

Nuclear inheritance of the biosynthesis of cyclopenin and cyclopenol in Penicillium cyclopium. 环青霉环青霉素和环酚生物合成的核遗传。

Balanced heterokarions were grown from Penicillium cyclopium aux-glu 1, a glutamic acid auxotroph producing benzodiazepine alkaloids of the cyclopenin-cyclopenol group, and P. viridicatum aux-met 1, a methionine auxotroph forming these alkaloids in traces only. In contrast to the hyphae of the parent strains, the hyphae of the heterokarions were dark orange-brown and grew well on media without the auxotrophic factors. In surface cultures they synthesized the benzodiazepine alkaloids cyclopenin and cyclopenol in amounts similar to those formed by the hyphae of P. cyclopium aux-glu 1. From the monokariotic conidiospores of the heterokarions homokariotic daughter strains were obtained which were similar to the parent strains in every respect. Hence no exchange of features of cyclopenin-cyclopenol biosynthesis took place between the parent strains at the stage of the heterokarion. This result indicates that the formation of cyclopenin and cyclopenol in P. cyclopium aux-glu 1 and the nearly complete lack of biosynthesis of these compounds in P. viridicatum aux-met 1 is encoded within the nucleus and is not influenced by plasmic genetic material.

Occurrence of gentamicin-producing micromonosporae in Lake Balaton (W. Hungary). 匈牙利西部巴拉顿湖产庆大霉素小单孢子菌的发生。

The total number of platable bacteria in the wet mud material characterized by the predominant occurrence of high-magnesian calcite and protodolomite of the Lake Balaton fluctuates between 3.5 X 10(4) and 2.3 X 10(5) g-1. Actinomycetes constitute about 25-30 per cent of this mud-community and not more than half of them belong to the genus Micromonospora. The presence of 10 species of this genus was detected among the isolated and studied strains identified by means of conventional methods and computer aided numerical analysis in which 20 authentic, mostly type strains of Micromonospora spp. were also involved. All of the three species of gentamicin-producing Micromonospora (M. echinospora, M. purpurea and M. sagamiensis) known so far were found in mud samples. These three species form stable but less numerous populations (less than 0.5 per cent of the total platable microflora) in the Balaton's mud, the members of which are capable to produce gentamicins at about 40-50 micrograms ml-1 level at most.
