Zusammenfassung Hier wird die spanische Ubersetzung des Artikels prasentiert, den Alexander von Humboldt im Jahr 1825 in der deutschen Zeitschrift Hertha veroffentlichte und in welchem er seine Reise durch Spanien sowie seine geographischen Untersuchungen der Hochebene kommentiert. Aufgrund seiner genauen wissenschaftlichen Argumentation, der Darstelllung der barometrischen Nivellierung sowie des topographischen Profils der Halbinsel, wurde die Notwendigkeit einer spanischen Ubersetzung dieser humboldtschen Arbeit mit dem Titel Uber die Gestalt und das Klima des Hochlandes in der iberischen Halbinsel erkannt. Die Art in der Humboldt seine auf der spanischen Halbinsel durchgefuhrten Forschungen prasentiert ist bemerkenswert, da er trotz des streng wissenschaftlichen Tones dieser Schrift, den Leser durch einen an seinen Herausgeber Professor Berghaus gerichteten Brief fuhrt, unter Hinzufugung von Anmerkungen aus seinen Tagebuchern sowie Daten, die er von seinen spanischen Briefpartnern erhalten hatte. Abstract We introduce the article originally published in the German periodical Hertha in 1825, in which Alexander von Humboldt made comments on his trip to Spain and on the geographic study of the plateau he had undertaken, with an introductory essay that presents the Spanish travel in its context. Due to the precise scientific argument used and the given barometric levels and topographic profile of the Iberian penninsula, we have considered the translation into Spanish of Uber die Gestalt und das Klima des Hochlandes in der iberischen Halbinsel ( Sobre la configuracion y el clima de la meseta de la Peninsula Iberica ) as necessary. The way in which Alexander von Humboldt presented his research upon the penninsula is a rather peculiar one since, in spite of the strictly scientific style used in his writings, he leads the reader by using a letter to his editor, Professor Berghaus, and inserts excerpts from his diary as well as data the he had obtained from his Spanish correspondents. Resumen Presentamos el articulo publicado en la revista alemana Hertha en 1825, donde Alexander von Humboldt comento su viaje por Espana y el estudio geografico de la meseta, con una introduccion a modo de ensayo, que situa el viaje espanol de Humboldt en su contexto. La argumentacion cientifica es precisa, se dan las nivelaciones barometricas y se ofrece el perfil topografico peninsular, lo que nos ha hecho considerar la necesidad de la traduccion al espanol de este trabajo de Alejandro de Humboldt titulado Sobre la configuracion y el clima de la meseta de la Peninsula Iberica ( Uber die Gestalt und das Klima des Hochlandes in der iberischen Halbinsel ). Se rescata la funcion critica y anticipativa del gran arte y se pone en cuestion el caracter presumiblemente progresista y democratico del arte contemporaneo. The slow dissociation reaction of the tubulin-colchicine complex has been characterized in purified calf brain tubulin and microtubule protein preparations, using [3H]colchicine and fluorometric measurements. It fits to a single exponential phase, within the accuracy of these measurements. The dissociation is a kinetically unfavorable reaction, with activation energy values of 114 +/- 10 and 94 +/- 10 kJ mol-1 (purified tubulin and microtubule protein, respectively). The kinetic scheme previously proposed for the tubulin-colchicine association (Lambeir, A., and Engelborghs, Y. (1981) J. Biol. Chem. 256, 3279-3282) is: T + C K1 in equilibrium TC k2 in equilibrium k-2 (TC)' where step 1 is a fast reversible binding and step 2 is a slow conformational change, whose backward rate constant (k-2) was neglected for the association study. This kinetic scheme has now been completed to include the measurements of the rate-limiting dissociation step (k-2), and of the purified calf brain tubulin preparation. The overall binding standard free energy change, calculated from the kinetic measurements, is -42.0 +/- 0.1 kJ mol-1 (fast phase of binding in 10 mM sodium phosphate buffer, 0.1 mM GTP, pH 7.0, at 37 degrees C). The binding is exothermic and the calculated enthalpy change is -26 +/- 13 kJ mol-1, which coincides with the recently determined calorimetric enthalpy value, -21 +/- 2 kJ mol-1 (Menendez, M., Laynez, J., Medrano, F. J., and Andreu, J. M. (1989) J. Biol. Chem. 264, 16367-16371), suggesting that the kinetic scheme and measurements are essentially correct.
