Isolierung von serinspezifischen transfer-ribonucleinsäure-fraktionen 有血清转移迹象的迹象

Soluble RNA was isolated from brewer's yeast; on the average 30–35 g were obtained from 50 kg. 350 mg of a serine transfer-RNA fraction were isolated from 32 g of soluble RNA by several subsequent countercurrent distributions in the tri-n-butylamin system (Fig. 1, Table I). The serine transfer-RNA fraction (31 mμmoles serine/mg RNA; less than 1 % of acceptor activity for other amino acids was detected) contained two serine transfer-RNA chains. The nucleotide composition of the serine transfer-RNA fraction was determined (Fig. 3, Table II). In a second series of countercurrent distributions the two serine transfer-RNA's were separated (Fig. 2) from each other and partially purified.

Studies on the “independence” of nucleolar ribonucleic acid synthesis 核仁核糖核酸合成的“独立性”研究

Using [14C]orotic acid as a precursor, the time course of labeling of nuclear and nucleolar RNA of rat liver has been studied in vivo. RNA from both nuclear and nucleolar preparations was fractionated into two classes with the aid of phenol. Each class of RNA was found to have a relatively specific kinetic profile after administration of the tracer. Labeling of nucleolar RNA was as rapid as nuclear RNA indicating that the extranucleolar RNA could not be the precursor of the nucleolar RNA. Actinomycin D inhibited nucleolar-RNA synthesis to a much greater degree than the synthesis of extranucleolar RNA. These data, together with previous observations on the base compositions of nucleolar-RNA fractions and other types of experiments provide evidence that nucleolar-RNA synthesis largely proceeds “independently” of extranucleolar-RNA synthesis.

On ribonuclease activity in reticulocyte ribosomes 网织细胞核糖体的核糖酶活性

RNAase (polyribonucleotide 2-oligonucleotidotransferase (cyclizing), EC 2.7.7.16) activity of reticulocyte ribosomes of rabbits was tested in comparison to the nucleolytic activity of liver ribosomes. Reticulocyte ribosomes showed practically no RNAase activity under conditions where such activity was demonstrated in liver ribosomes.
