Growth and nutritional health of Pterophyllum scalare juveniles: Unleashing the benefits of feeding with Artemia sp. in aquariums 鳞叶蕨幼体的生长和营养健康:释放在水族馆中与蒿属一起喂养的好处

Pterophyllum scalare is a popular ornamental fish species, but current rearing methods result in high mortality, low fertility, disease incidence, and slow growth in aquarium conditions. Research on co-feeding for ornamental fish at the juvenile or adult stage needs to be completed. This study implemented a “snacking” feeding strategy using Artemia sp. to evaluate changes in fish survival rate, growth performance, and enzyme activity related to digestion, antioxidants, and immunity in a recirculating aquaculture system. Two feeding strategies were tested: one group was fed a commercial diet plus Artemia sp. (0.1% of diet) as “snacking,” and another group was fed only the commercial diet as normal. “Snacking” with Artemia sp. enhanced fish’s survival and growth performance, with higher relative weight rate, standard length, and total length than the control group. Specific growth rates for weight, standard length, and total length were also more significant in the supplemented group compared to the control group. The activity of amylase, superoxide dismutase, and glutathione peroxidase was substantially higher in the supplemented group. Adding Artemia sp. as a supplement under a “snacking” feeding strategy to the control diet was beneficial for P. scalare juvenile rearing and can guide managing recirculating farming activities.

Effects of combined rice flour and molasses use on the growth performance of Pacific white shrimp (Litopenaeus vannamei Boone, 1931) applied biofloc technology 米粉和糖蜜混合使用对应用生物絮团技术的凡纳滨对虾生长性能的影响

A 63-day completely random experiment with three replications was carried out to compare the effects of five different combination ratios of rice flour (R) and molasses (M) on the growth and survival rates of Pacific white shrimp (Litopenaeus vannamei Boone, 1931) postlarvae applied biofloc technology. Five biofloc (BF) treatments, including R90-M10, R70-M30, R50-M50, R30-M70, and R10-M90, formed with the addition of different combination ratios of rice flour and molasses, i.e., 90% R+10% M, 70% R+30% M, 50% R+50% M, 30% R+70% M, and 10% R+90% M, respectively, with C/N ratios of 15:1, and a control (neither rice flour nor molasses applied) was randomly arranged into the 18 plastic tanks of 1.0 m3 volume (with 0.5 m3 of water) each tank and salinity of 15‰. The postlarvae (0.095 g) were stocked into the tanks at a 150 ind. m−3 density and fed pelleted feed (40% protein). There was an improvement in growth (FMW, WG, DWG, and SGR) for all treatments. Besides, treatments with more than or equal to 30% molasses have improved SR, FCR, and FB. Especially the highest SR (94.2%) was obtained at the R70-M30, which perhaps created the highest FB (1.435 kg m−3) in this treatment. The lowest FCR (1.28) was also observed in the R70-M30 and significantly differed from the control and other treatments. Besides, water quality parameters were within the ranges recommended for Pacific white shrimp health during the experimental period. Our findings indicated the benefits of shrimp culture using the BF system when different combined ratios of rice flour and molasses were applied, of which a ratio of 70% rice flour and 30% molasses was considered as the best.

The drug resistance of Vibrio harveyi in aquaculture became more severe because the control of Vibriosis depends majorly on the current antibiotics. Transcriptomes of a wild-type strain (VS) and its quinolone-resistant mutants (VR) of V. harveyi were respectively sequenced by RNA-seq technology. A total of 2,082 unigenes were obtained after de novo splicing and assembly. 129 genes were identified with significant differential expression in strain VR compared to strain VS, among which 65 were up-regulated and 64 down-regulated. Then, functional annotation and enrichment analysis of these differentially expressed genes (DEGs) were performed. GO enrichment results showed that DEGs focused mainly on cell structure, substance metabolism, and transporter. COG classification of the DEGs mainly focused on amino acid transport and metabolism, cell wall/membrane biosynthesis, carbohydrate transport and metabolism, ribosomal structure, and biosynthesis. KEGG pathways related to a two-component system, ABC transport system and flagellar assembly (ko02040) were enriched significantly, and 9 genes associated with quinolone-resistance ability, including genes for resistance-related transport proteins, outer membrane proteins, and DNA repair-related proteins were discovered through analysis of the drug-resistance related genes. Ten DEGs (including the above part genes of 9 drug resistance-related genes) in the transcriptome data were taken to analyze their expression with real-time qPCR. The results were the same as the changes of the above transcriptome analysis, further confirming the reliability of the transcriptome sequencing and data analysis. In a word, genes from a two-component system, flagellar assembly, active efflux system and outer membrane proteins take great roles in the quinolone-resistance of V. harveyi. These results provide enough information for further study on the molecular mechanism of quinolone-resistance and give a helpful transcriptomic resource to unravel the contact between quinolone-resistance and metabolic pathways in Vibrios.

Haemolytic Bacillus subtilis, a potential pathogen of hemorrhage disease in farmed yellow catfish Pelteobagrus fulvidraco 养殖黄颡鱼出血病的潜在病原体溶血性枯草芽孢杆菌

The hemorrhagic disease has become a major bottleneck in yellow catfish Pelteobagrus fulvidraco aquaculture. Yet scarce information is available on Bacillus subtilis as a pathogen of hemorrhagic disease in cultured yellow catfish. In this study, a virulent isolate, temporarily named HS5, was isolated from hemorrhagic disease-infected cultured yellow catfish, and was identified phenotypically and molecularly as B. subtilis. In addition, isolate HS5 showed an LD50 value of 5.20×106 CFU/ml in yellow catfish, exhibited beta-haemolytic activity, harbored the virulent entFM gene, and displayed high susceptibility to enrofloxacin and florfenicol in aquaculture use. To the best of our knowledge, this is the first report of B. subtilis as a pathogen of hemorrhagic disease in farmed yellow catfish.

Gut health is essential for animal growth because it can absorb nutrients more efficiently, promote growth, and protect the body from invasion by pathogenic microorganisms. Temperature, dissolved oxygen, water quality, nutrition level, pathogen, net fishing, and so on will affect the gut health of aquatic animals. Intestinal diseases in aquatic animals were often not easily detected, making them difficult to prevent and control. Therefore, establishing a stable and scientific evaluation standard of gut health was of great significance. This study evaluated the effects of gross pathological and histological parameters of largemouth bass and channel catfish infected by Aeromonas hydrophila. A total of 150 largemouth bass and 150 channel catfish were randomly divided into 5 groups: 1 control group (NC) and 4 challenge groups (CH, fish were infected with A. hydrophila by intraperitoneal injection with a concentration of 200 µL 1x108 CFU/mL, 1x107 CFU/mL, 1x106 CFU/mL, and 1x105 CFU/mL, respectively). After the challenge, largemouth bass’ gross pathological and histological changes were observed on the 2nd, 6th, 9th, 12th, and 14th days. In contrast, channel catfish’s gross pathological and histological changes were observed on the 3rd, 6th, 9th, 12th, and 15th day. In this study, regarding the “I See Inside” (ISI) methodology of poultry, a pathological evaluation method of fish intestinal health was established to comprehensively evaluate the intestinal health status of fish by gross pathological and histopathological score. In gross pathology, it was scored by the body surface and intestinal lesions. In histology, it was a metric evaluation of histological alterations in the intestine, converting macroscopic and microscopic alterations into numbers and then scoring. Each evaluation parameter sets up an “influence factor” (IF) according to the degree of influence of its pathological changes on intestinal function. The final score was the sum of the IF product and this parameter’s score. The results of two kinds of fish showed that the gross pathological and histopathological scores correlated with the infection dose and time. And the significant difference in the score between the enteritis and control groups was observed. The results showed that the higher score, the more serious the pathological changes of fish infected by A. hydrophlia. From this, it can be concluded that the pathological evaluation method established can be used to evaluate the gut health of fish.

In this study, regarding the “I See Inside” (ISI) methodology of poultry, a pathological evaluation method of fish intestinal health was established to comprehensively evaluate the intestinal health status of fish by gross pathological and histopathological score. In gross pathology, it was scored by the body surface and intestinal lesions. In histology, it was a metric evaluation of histological alterations in the intestine, converting macroscopic and microscopic alterations into numbers and then scoring. Each evaluation parameter sets up an “influence factor” (IF) according to the degree of influence of its pathological changes on intestinal function. The final score was the sum of the IF product and this parameter’s score. The results of two kinds of fish showed that the gross pathological and histopathological scores correlated with the infection dose and time. And the significant difference in the score between the enteritis and control groups was observed. The results showed that the higher score, the more serious the pathological changes of fish infected by A. hydrophlia. Spotted seabass (Lateolabrax maculatus) is a popular food fish in Asian countries. Not only does the fish tasted well, but it is also widely utilized as nutraceuticals. However, the knowledge about the composition of the fish is lack. In this work, the proximate composition of spotted seabass flesh was determined. The content of proximate composition (moisture, proteins, fat, and ash) of spotted seabass flesh was fell within the range of other reported fish species, while it was a high-quality food fish with low oil-very high protein. The ratio of essential amino acids (EAAs) to nonessential amino acids (NEAAs) was 0.74, and the amount of EAAs accounted for approximately 42.34% of the total amino acids. Unsaturated fatty acids composed the majority of the fatty acid components, with linoleic acid was the predominate one. The content of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) were abundant in spotted seabass flesh. The proximate composition of the dorsal, ventral, lateral, and tail flesh was similar, except for the ventral flesh exhibited a higher content of fatty acids and lower content of moisture. Our research will be helpful for the general population to know the nutritional traits of spotted seabass.

Bacteriophage applications in aquaculture 噬菌体在水产养殖中的应用

Aquaculture has grown tremendously due to the big demand for its products. However, diseases affecting aquaculture and economic losses are worldwide problems and it needs low cost, sustainable, highly efficient, specific and eco-friendly therapeutants. Due to the rising up antibiotic resistant-microorganism, bacteriophage therapy has reinvigorated to replace antibiotics in agriculture, medicine, food safety and the environment. Likewise, it also holds great promise to avoid, control and treat bacteria in aquaculture to decrease the mortality level of different aquatic animal diseases. The isolation and characterization of new phages and phage application therapy to eliminate bacterial fish and shellfish pathogens such as Vibrio, Aeromonas, Pseudomonas, Lactococcus, Yersinia, Flavobacterium, and Streptococcus was gradually reported in aquaculture literature. The present review summarizes large-scale reports in vitro or in vivo use of aquaphage studies and applications in fish diseases from the 1980s to 2022 and future directions.

Bogue (Boops boops, Linnaeus, 1758) that was gathered in Antalya Bay had their ages, growth rates, and mortality examined. Using the von Bertalanffy equation, Lt = 37.50 (1 - e-0.15(t + 1.32)) (R2 = 0.988), growth in length was expressed for the entire sample without revealing any disparities between the sexes. The values of the allometric coefficient (b) of the length-weight relationships specify negative allometric growth (b < 3) in females, males, and overall sexes. Total and natural mortality were found to be Z= 1.12 y-1, and M= 0.36 y-1. The exploitation ratio E = 0.68 indicated that the population was overexploited.

Effect of aqueous ozone treatment on some quality changes of vacuum-packed meagre (Argyrosomus regius) fillets 水臭氧处理对真空包装松茸鱼片品质变化的影响

In this study, microbiological, sensory and color changes in vacuum-packed meagre fillets treated with aqueous ozone for different periods of time were investigated. Meagre fillets were treated with ozone for 15 (O-15) and 30 (O-30) minutes and vacuum-packed and stored at +2 °C for 12 days. Control group (C) was not treated with ozone and stored after vacuum packaging. For microbiological changes, Total Mesophilic Aerobic Bacteria (TMAB), Pseudomonas sp., Lactic Acid Bacteria (LAB) and Total Psychrophilic Aerobic Bacteria (TPAB) counts were analyzed. At the end of storage, the highest bacterial counts were found in group C (>7.00 log cfu/g). In ozone-treated samples, Pseudomonas sp. counts did not exceed 7.00 log cfu/g only in the O-30 group. In the sensory evaluation of the samples, it was determined that the total demerit points were 18 points for group C, 16.25 points for O-15 and 14.75 points for O-30 at the end of storage over 30. No significant change was found according to the color results of the samples, but at the end of storage, it was observed that the L value of the ozone-treated samples was higher and the samples had a lighter color. In the conclusion of the results, it was observed that ozone treatment prolonged the shelf life of vacuum-packed meagre fillets for 4 days for the O-30 group compared to control and O-15 group without causing the sensory and color loss.

A heat shock protein 70 (Hsp70) was identified from Procambarus clarkii (named as PcHsp70). The open reading frame of the PcHsp70 cDNA was 1917 bp, encoding a 638 aa protein with signature sequences of the Hsp70 family. Tissue distribution results showed that PcHsp70 has relatively high expression in immune-related tissues such as hepatopancreas, gill, and hemocytes. The expressions of PcHsp70 in hepatopancreas and gill were significantly induced by Gram-positive bacterium, Gram-negative bacterium, and white spot syndrome virus stimulation, indicating that PcHsp70 may be involved in the immune response to pathogen invasion. The research will provide a foundation for understanding the resistance and stress mechanism of Hsp70 in P. clarkii.
