[Review] Extraction of Intact Heteroxylan by Ionic Liquid and Its Structural Characterization [综述]离子液体萃取完整杂氧蓝及其结构表征

植物細胞壁は,セルロース,ヘミセルロース,リグニン の主要成分から構成され,地球上で最大の炭素貯蓄源であ り,植物バイオマスと呼ばれている.人類は,植物バイオ マスを直接的な燃料や建築材料,紙パルプなどの資源とし て利用してきた.しかし,化石資源の枯渇が懸念される 中,これまで以上に,植物バイオマスをより多様な用途に 活用していくための技術革新が求められている.自然界の 循環系に目を向けてみると,光合成によって生産された植 物細胞壁の大部分が,担子菌をはじめとする森林の微生物 によって資化されており,この際に大きな役割を果たすの が,菌体外に分泌される酵素である.このことを考える と,我々が植物細胞壁をバイオマス資源として上手く利用 していくためには,酵素による細胞壁分解をより本質的に 理解することが重要だと言える.筆者らの研究室では,植 物細胞壁中の多糖類の利用を目的に,木材腐朽力が高い菌 として単離された白色腐朽菌ウスバタケ (Irpex lacteus) が Extraction of Intact Heteroxylan by Ionic Liquid and Its Structural Characterization イオン液体を用いた天然型キシランの抽出とその 構造的特徴
植物细胞壁由纤维素、半纤维素、木质素等主要成分构成,是地球上最大的碳储蓄源,被称为植物生物量。人类一直将植物生物量作为直接的燃料、建筑材料、纸浆等资源加以利用。但是,在化石资源枯竭的担忧下,植物生物量比以往更加多样化的用途为了灵活运用,需要技术革新。把目光转向自然界的循环系统,光合作用所生产的植物细胞壁的大部分,被担子菌等森林微生物所养分化,这时担子菌就发挥了很大的作用。但是,从菌体外分泌的酶。考虑到这一点,我们为了把植物细胞壁作为生物量资源好好利用,更本质地理解酶对细胞壁的分解是很重要的。在笔者的研究室里,为了利用植物细胞壁中的多糖类物质,分离出了一种具有很强的木材朽力的白色朽菌——薄糟朽菌(Irpex lacteus)。Heteroxylan by Ionic Liquid and Its Structural Characterization使用离子液体的天然型氧基烷的提取及其结构特征

[Review] Moving Mechanism of Chitinase A from Serratia marcescens [综述]粘质沙雷菌几丁质酶A的迁移机制

1.1 プロセッシブな糖質分解酵素 プロセッシビティとは,1回の基質酵素複合体形成あた りの触媒反応回数ということができる.糖質関連酵素の分 野では,セルラーゼ,キチナーゼなどの加水分解酵素につ いて解析がなされることが多いため分解反応回数と思われ がちである.しかし他分野では DNAポリメラーゼの合成 反応回数やモーター蛋白質の運動回数として使われる概念 であり,酵素の重要な性質の一つである. セルラーゼについては,X線結晶構造解析によりトンネ ル状の触媒ドメインの構造が明らかになりプロセッシブ反 応の可能性が示された.その後,分解反応後の結晶性セ ルロースが尖ることからプロセッシブ反応が検証された. またキチナーゼについても,分解後の結晶性キチンの形状 変化からプロセッシブ反応モデルが提案された.更にプ ロセッシビティ定量のために,生成物の奇数糖と偶数糖の 比から推定する方法,基質を改変するまたは可溶性と不 溶性の基質を混合して計測する方法などが提案されてき た.プロセッシビティの定義を考えると,生成物の量と Moving Mechanism of Chitinase A from Serratia marcescens セラチア菌由来キチン加水分解酵素の運動機構
1.1处理性糖质分解酶所谓处理性,可以说是指1次基质酶复合物形成值的催化反应次数。在糖类相关酶的分野中,由于多分析纤维素酶、激酶等水解酶,所以容易被认为是分解反应次数。但是在其他领域DNA聚合酶的合成是作为反应次数和马达蛋白的运动次数使用的概念,是酶的重要性质之一。关于塞尔酶,通过X射线结晶结构分析,发现了隧道状催化域的结构,处理反随后,由于分解反应后结晶性脊骨变尖,验证了处理反应。另外,激酶分解后结晶性甲壳素的形状。从变化中提出了处理反应模型。为了进一步定量焦糖,提出了根据生成物的奇数糖和偶数糖之比推算的方法,改变基质或混合可溶性和不可溶性基质测量的方法等。考虑处理器的定义,生成物的量和Moving Mechanism of Chitinase A from Serratia marcescens来自塞拉西菌的甲壳素水解酶的运动机制
