Quantification of Isoflavone Malonylglucosides in Soybean Seed during Germination 大豆种子萌发过程中异黄酮丙二醇苷含量的测定

Soybean produces three major types of isoflavones, daidzein, genistein, and glycitein aglycones and their glucosides and malonylglucosides. It has been known that malonylated glucosides are rapidly converted to their corresponding aglycones due to the unstable thermolabile glucoside malonates; therefore, the analytical study of malonylated glucosides has been insufficient. In this study, we analyzed the malonylglucoside content in soybean seeds. Isoflavone analysis of three soybean cultivars revealed that 81.5~90.0% of the total isoflavones were malonylglucosides, whereas aglycones were rarely detected. Moreover, the total isoflavone content increased during a 5-day germination period where growth regulators and coumaric acid treatments tended to yield higher isoflavone content than the normal germination treatment, however the differences were not significant; notably, the isoflavone accumulation trend continued with additional germination days. The content of malonylglucoside was higher than that of other isoflavones, which was 83.7~86.6% of the total isoflavone content in seeds with a 3-day germination period. Furthermore, isoflavones were significantly accumulated in the hypocotyl of seedlings with a 5-day germination period. The content of isoflavone in the hypocotyl of the Pungsannamul-kong was 10,240 ug/g when treated with coumaric acid, which was considerably higher than that of other cultivars and treatments. Additionally, soybean seeds heated at 60°C for 1 hour produced higher isoflavone content than non-heated soybean seeds. Our results show that it is possible to increase the isoflavone content in soybean seeds through various treatments.

Analysis of Genetic Variation in Pre-Harvest Sprouting at Different Cumulative Temperatures after Heading of Rice 水稻抽穗后不同积温收获前发芽的遗传变异分析

Pre-harvest sprouting (PHS) refers to seed germination during ripening, due to loss of dormancy before harvest. As PHS in rice causes decrease in grain yield and quality, tolerance to PHS is an important trait of Japonica cultivars in Korea. It is important to investigate the related genes and environmental factors, because PHS is a quantitative trait. In this study, we examined PHS rates at three different times according to the cumulative daily mean temperature after heading (CTAH) for 5 rice cultivars released in Korea for 5 years from 2013 to 2017 to determine the effect of environmental factors on PHS. ABA content in ripening spikelets was analyzed to understand how it was related to PHS tolerance. PHS rate increased as CTAH increased from 800°C to 1200°C. PHS rate was significantly different (p < 0.001) among the cultivars, showing Dasanbyeo, Jounbyeo, and Nampyeongbyeo to be PHS-tolerant, and Jopyeongbyeo and Gopumbyeo to be susceptible at all the CTAH of 800, 1000, and 1200°C. In 2015 and 2016, PHS rates were relatively higher, because of high temperature and frequent rainfall during the ripening period. In each cultivar, ABA content decreased as CTAH increased from 800°C to 1200°C. However, there was no significant correlation between ABA content and PHS tolerance among the cultivars.

Genetic Diversity and Population Structure of a Korean Rice Germplasm Based on DNA Profiles 基于DNA图谱的韩国水稻种质遗传多样性和群体结构研究

Information on the patterns of genetic diversity and population structure is essential for the rational use and efficient management of germplasms; accurate information aids in monitoring germplasms, and can also be used to predict potential genetic gains. In this study, we assessed genetic diversity, focusing on Korean rice accessions for theand their sustainable conserved diversity. Using DNA profiling with 12 simple sequence repeat (SSR) markers, we detected a total of 333 alleles among 2,016 accessions. The number of alleles ranged from 21 to 53, with an average of 27.8. Average polymorphism information content was 0.797, with the lowest being 0.667 and the highest 0.940. CA cluster analysis and the model-based population structure revealed two main groups that could be subdivided into five subgroups. Analysis of the molecular variance study based on the SSR profile data showed 5% variance among the profiles, whereas we recorded 93% variance among individuals and 2% variance within individuals. Specifically, the utilized diversity for of the breeding program is restricted in that cultivars were located in limited clades. These results revealed that preserving the diversity of Korean landraces could be useful sources for breeding new rice cultivars, and cwould be the basis for the sustainable conservation and utilization of a Korean rice germplasm.

Correlation of Quality Characteristics of Soybean Cultivars and Whole Soymilk Palatability 大豆品种品质特性与全脂豆浆适口性的相关性研究

The correlation between the nutritional composition of soybeans and whole soymilk palatability was investigated using nine soybean cultivars (Teagwangkong, Daewonkong, Saedanback, Jinpung, Daechan, Miso, Cheongmiin, Cheongja 3, and Socheongja). Physicochemical analysis of soybeans, showed that the protein and lipid contents were 37.7-46.0 and 15.2-20.9%, respectively. Unsaturated fatty acids were 81.1-84.8% of total fatty acids, of which linoleic acids was 49.7-56.8%. Total tocopherol was 243.5-361.3 μg/g of extract, of which γ-tocopherol was 67.14-86.49 μg/g. Total isoflavone contents varied within cultivars from 495.4-1,443.8 μg/g of extract. Daidzin and genistin were 252.1-556.0 and 241.8-730.7 μg/g, respectively, which were major isoflavones in soybeans. For the sensory evaluation, whole soymilk was made from nine soybean cultivars and 20 panels investigate its palatability. The Daechan cultivar had the highest (9.1), and Cheongmiin the lowest (5.6), overall palatability score. Interestingly, sensory results were strongly correlated with linoleic acid (0.746) and stearic acid (-0.716) content. In summary, the fatty acid composition of soybeans is the key factor in determining the palatability of whole soymilk. This study could be applied to determine the suitability of cultivars for soybean products, including whole soymilk.
