Metchnikovella dobrovolskiji sp. nov. (Microsporidia: Metchnikovellida), a parasite of archigregarines Selenidium pygospionis from the polychaete Pygospio elegans Metchnikovella dobrovolskiji sp.nov

Summary Spore sacs and free spores of a metchnikovellid were found in several specimens of the archigregarine Selenidium pygospionis isolated from polychaetes Pygospio elegans , collected from the littoral area of the Kandalaksha Gulf of the White Sea. The morphology of spore sacs suggests that this species belongs to the genus Metchnikovella ( sensu Caullery and Mesnil, 1914). The length of the spore sacs varied from 5.6 to 9.2 µm, the width of the spore sacs was 3.3–5 µm, each of them contained up to 12 oval spores. Both spore sacs and free spores were enclosed in vacuoles. The combination of morphological features and host range distinguishes the studied isolate from any known species. Results of the phylogenetic analysis based on the SSU rRNA gene showed that the sequence of this species had a significant difference from all available sequences of metchnikovellids and usually grouped close to the species Metchnikovella incurvata or formed a clade with it. Here we describe the studied isolate as a new species, Metchnikovella dobrovolskiji sp. nov. (Microsporidia: Metchnikovellida).

Diversity and abundance of naked lobose amoebae belonging to the classes Tubulinea and Discosea in pond sediments in Moscow urban parks 莫斯科城市公园池塘沉积物中管形纲和Discosea纲裸叶变形虫的多样性和丰度

Summary Naked lobose amoebae (also known as gymnamoebae) nowadays are split into the Tubulinea and Discosea lineages within the Amoebozoa supergroup; few genera belong to the Variosea lineage. Morphological identification of amoeba species is difficult because it requires study of clonal cultures. Most of species nowadays require molecular data for precise identification. The deficiency of these data results in the lack of reliable information on the local amoebae faunas, their abundance and distribution in natural and artificial habitats. In this paper we provide faunistic and quantitative data on naked lobose amoebae from bottom sediments of ponds located in urban parks of Moscow, Russia. We applied enrichment cultivation, followed by light-microscopic study, and in some cases – electron-microscopic and molecular studies. In total, 29 amoebae species were found. Most of recovered isolates might be new to science. The number of amoebae cells in the sediments, estimated using the most probable number method, varied from 75 to 288 cells ml -1 . Overall, the diversity and abundance of naked amoebae in urban park ponds is comparable with that in intact biotopes, and they are not affected by the urbanization-driven pollution. These results suggest that urban parks play an essential role in maintaining and preserving eukaryotic microbial diversity in the anthropogenic environment.

The morphology of two new species of centrohelid heliozoans, Choanocystis mylnikovi and C. punctata , from the brackish-water Tuzlukkol’ River, South-Eastern Russia, were studied using scanning electron microscopy. The new species are covered with two types of scales that are plate scales and spine scales with heart-shaped basal plates and proximally curved shafts. C. mylnikovi has spine scales that are highly variable in length, which differs up to six times (2.0–12.9 µm). Spine scales of C. mylnikovi are slightly tapering, with two small teeth of the same length on the apexes. This species also has oviform or oval plate scales (1.6–2.9 × 1.0–1.8 µm) with an axial ridge and thin marginal rim. C. punctata has both short (3.6–9.9 µm) and long (10.0–18.5 µm) spine scales with pointed apexes. Sometimes short spine scales of C. punctata end with a bent apex or a small bent hook. Plate scales (2.9–4.7 × 1.5–3.2 µm) and basal plates of spine scales are ornamented with numerous granules. C. punctata sporadically demonstrates the presence of spine scales with the shaft in a nearly central position and membranous extensions of the basal plate along the shaft, which are the characteristics for the other morphologically defined genera of centrohelids – Acanthocystis, Pterocystis or Raineriophrys . Similarities and differences between the new species and other members of the genus Choanocystis are discussed.

Changes of macronuclei structure during encystment in the ciliated protozoan Climacostomum virens 纤毛原生动物生囊过程中大核结构的变化

Summary Ciliate cysts are an important object for investigating changes in cell morphology during cryptobiosis. In this work, we studied the ultrastructure of Climacostomum virens resting cysts using electron and confocal microscopy. The main attention was paid to the morphological changes in the macronucleus, nucleoli and chromatin. Vegetative cells, cysts 1-2 h after cyst wall formation, 2-months old cysts, one-year old cysts, and cells after excystment were investigated. The process of encystment in C. virens , as in other ciliates, is accompanied by a decrease in cell volume and the formation of a cyst wall. The volume of the macronucleus reduces, too. This reduction is likely to be related to the compaction of the macronuclear chromatin since extrusion of chromatin and nucleolar material into the cytoplasm during C. virens encystment was not observed. The nucleoli in young and 2-months old cysts retained their interphase morphology with well-defined fibrillar and granular zones. Only at the age of one year, the nucleoli in the cyst reduced in size and consisted of a dense homogeneous material. The chromatin structure in the cyst micronuclei was morphologically identical to macronuclear chromatin at most other cell cycle stages. Macronuclear chromatin in vegetative C. virens was mainly represented by individual bodies, 100–200 nm in size. As the age of the cysts increased, the chromatin bodies formed extended fibrils. In one-year old cysts, the entire space of the macronucleus was filled with fibrils 100–200 nm thick. Such fibrils resemble chromonemes in the chromosomes of higher eukaryotes and probably are formed by similar cellular mechanisms

Macronuclear chromatin in vegetative C. virens was mainly represented by individual bodies, 100–200 nm in size. As the age of the cysts increased, the chromatin bodies formed extended fibrils. In one-year old cysts, the entire space of the macronucleus was filled with fibrils 100–200 nm thick.
