The characterization of anther-specific promoter encoding chalcone synthase isolated from Lilium Oriental hybrid cv. Acapulco 东方百合花药特异性启动子编码查尔酮合成酶的特性研究。阿卡普尔科

Anthocyanins are the major pigments contributing to flower coloration. A 1584 bp 5' upstream sequence of ALCHS2 gene was isolated from Acapulco lily (Lilium Oriental hybrid cv. Acapulco). Computer-based analyses (GeneScan, AtPAN) predicted a CAATBOX1 and putative transcription factor-binding sites, including tissue-specific elements. When gALCHS7 promoter–gus fusion was introduced to petunia ('Dream Red'), all ten putative transgenic plants showed localized GUS activity in the anther, but five of them also showed weak GUS activity in the ovule. No distinctive signal in the leaf and petal was detected in the same stage. To clearly determine the operation of the promotor region, anther and ovule tissues of transgenic line 6 were fixed in paraffin for dark-field analysis. At 1 cm length of floral bud, a GUS signal was not observed in the anther, but weak expression was observed in the ovule. Before anthesis, GUS protein was highly expressed in the pollen, endothecium, and epidermis. Fluorometric GUS assays of individual organs taken from four transgenic plants demonstrated that all lines showed high GUS activity in the anther compared to 35S CaMV promoter (pBI1 121), except line 34. Using the truncated promoters by cis-acting elements, we found that minimal region (gALCHS7-7, 270 bp) displayed GUS expression only in the anther, though at weaker activity than in the original promoter.

A genetic linkage map of allo-octoploid strawberry (Fragaria × ananassa Duch.) using SNP markers 利用SNP标记建立异源八倍体草莓(Fragaria × ananassa Duch.)遗传连锁图谱

An allo-octoploid strawberry (Fragaria × ananassa Duch.) is one of the most important vegetable crops in Korea. However, there were few genomic researches of strawberry due to polyploidy and complexity of its genome. In this study, we aimed to construct a genetic linkage map of strawberry using single nucleotide polymorphism (SNP) markers that were developed through a next-generation sequencing (NGS) analysis. Two strawberry varieties, ‘Sulhyang’ and ‘Senga-sengana’, were used as a maternal and a paternal parent, respectively, and their F1 generation consisting of 94 individuals was used for construction of a genetic linkage map. A total of 19.0 Gbp (‘Sulhyang’) and 21.8 Gbp (‘Senga-sengana’) of genomic sequences were obtained through NGS analysis. Subsequently, approximately 87,000 SNPs were identified and 1,154 primer sets for high-resolution melting (HRM) analysis were designed through bioinformatic analysis. In result, a total of 224 polymorphic HRM markers were developed and 205 markers were mapped on the genetic linkage map of strawberry, which total length was 800.8 cM and the number of linkage groups were 30. This SNP-based genetic linkage map and the 224 SNP markers will be very helpful for the genomic and genetic researches of allo-octoploid strawberry.

‘Nokwoo’, a mid-late maturing, high dry matter yielding rice (Oryza sativa L.) cultivar with a good early growth and low-temperature germinability, was developed for whole crop silage (WCS) use. It was derived from a cross between a leafy tropical japonica ‘LK1A-2-12-1-1’ with high biomass and good germinability in low temperature and new plant type (NPT), ‘IR72225-29-1-1’ which had low tillering trait, large panicle, dark green leaf, thick and sturdy stem and vigorous root system. This cultivar had about 123 days growth period from seeding to heading, 122cm culm length, 29cm panicle length, 9 panicles per hill, 144 spikelets per panicle and 1,000-grain weight of 24.4 g as brown rice in central plain region, Suwon. This wide and long leafy WCS rice variety was weak to cold stresses similar to ‘Nokyang’ but was a little resistant to lodging in the field, strong to viviparous germination and good to low temperature germination. In addition, ‘Nokwoo’ was resistant to leaf and neck blast but susceptible to bacterial blight, rice stripe virus and brown planthopper. Its average dry matter yield for three years reached 16.5 MT/ha, 14% higher than that of ‘Nokyang’. This cultivar had 5.3% crude protein and 68.8% total digestible nutrients a little low compared to ‘Nokyang’. In Korea peninsular, ‘Nokwoo’ grows well in central and southern plain and is good to harvest between 15 to 30 days after heading to improve its feeding value and digestion rate of livestock (Grant No. 6072).

A New Single Cross Hybrid for Popcorn with Lodging Tolerance and High Yield 耐倒伏高产玉米单交杂交种新选育

: This study was carried out to create new popcorn variety. ‘G-Popcorn’ was made by single crossing with two inbred lines, the seed parent, GP3, and the pollen parent, GP4. The hybrid was made in 2009 and selected in 2014 after investigating characteristics for three years from 2012 to 2014. ‘G-Popcorn’ was evaluated on 2 or 3 places in Gangwon-do for three years. ‘G-Popcorn’ showed higher lodging tolerance and popping volume in contrast to the check variety ‘Oryunpopcorn’. The plant height is 213cm indicating 5cm lower than that of ‘Oryunpopcorn’. Also, ration ear height of plant height of ‘G-Popcorn’ is 59%. The weight of 100 seeds was 15.5g, similar to that of ‘Oryunpopcorn’. The popping volume of ‘G-Popcorn’ was about 28.3 ㎤ /g, higher than 25.0 ㎤ /g of ‘Oryunpopcorn’. The yield of ‘G-Popcorn’ was 467kg/10a in regional yield trials for three years, which was 7% higher than that of ‘Oryunpopcorn’.

Mungbean Cultivar, ‘Suhyeon’ with Short Stem Length, Disasters Resistance and High Yielding 短茎、抗灾、高产绿豆品种‘水玄’

Wilczek) was derived from the cross between ‘Jeonnam14’ and ‘IT208777’ at the Jeollanamdo Agricultural Research and Extension Services (JARES) in 2010. ‘Suhyeon’ has erect growth habit, heart leaflet, green hypocotyl, black and straight pod when matured, and dull green seed surface. The stem length of ‘Suhyeon’ was 66 ㎝ , which was 5 ㎝ less than check variety ‘Owool’. The pods number per plant of ‘Suhyeon’ was 24, which was 5 more than check variety. The sprout yield ratio of ‘Suhyeon’ was 8.67 times, which was 16% more than check variety. The contents of starch, protein and fatty acid of seed were 62.7, 27.3, 1.89%, respectively. The average yield of ‘Suhyeon’ was 1.74 ton/ ㏊ , which increased 8% more than that of check variety. (Variety registration number: 5107)

Effect of Gamma-ray on Survival and Mutation Rates of Rooted Cuttings and Unrooted Cuttings in Rose 射线对月季有根插枝和无根插枝成活率和突变率的影响

This study was carried out to compare the survival and mutation rates and mutation spectrum by gamma-irradiation on rooted and unrooted cuttings of three spray type (‘Lovelydia’, ‘Yellowbabe’, and ‘Haetsal’) and two standard type (‘Vital’ and ‘Aqua’) cultivars in roses. Two groups, rooted and unrooted cuttings were gamma-irradiated at 70Gy for 24 hours. The irradiated rooted and unrooted cuttings were planted in a greenhouse, and survival, mutation rates and mutation spectrum were investigated 30 weeks after planting, respectively. As a result, survival and mutation rates of gamma-ray irradiated plants were 16.4%~50.8% and 0~5.1% for unrooted cuttings, and 39.4%~55.1% and 0.7%~7.4% for rooted cuttings, respectively. In conclusion, both survival and mutation rates were a little higher on rooted cuttings than on unrooted cuttings. However, when only survived plants after gamma-ray irradiation were considered, mutation rates were 0~10% and 1.8%~14.1% for unrooted cuttings and rooted cuttings, respectively, showing no significant difference. In addition, diverse variations on color and number of petals or shape of flowers were detected both in plants from rooted and unrooted cuttings, which indicated that there was no significant difference in mutation spectrum between two groups.

Green Seed-Coat Colored Soybean Variety ‘Jungmo3005’ with Tolerance to Lodging and Shattering 耐倒伏、耐剥落的绿皮有色大豆品种‘中莫3005’

A new colored soybean variety ‘Jungmo3005’ was developed as a breeding parent. ‘Cheongjakong’ and ‘Geomjeongkong3’ were crossed in 2000. F1 and F2 populations were grown for 2 years and selected by pedigree method from F3 to F5. The preliminary yield trial (PYT) and advanced yield trial (AYT) were conducted from 2006 to 2007, and regional yield trial (RYT) in 9 regions was conducted from 2008 to 2010. ‘Jungmo3005’ is determinate, white flower, green cotyledon, green spherical seed and yellow hilum. Flowering date and maturing date were July 30 and Oct. 7, respectively. Other quantitative characteristics of ‘Jungmo3005’ were similar to ‘Cheongdu1’, but it was more tolerant to lodging and shattering than ‘Cheongdu1’ at RYT field and indoor test. Although ‘Jungmo3005’ showed symptom of mosaic disease in inoculation test at greenhouse, it had high level of resistance to soybean mosaic virus and bacterial pustule diseases at field. The yield of tofu of ‘Jungmo3005’ was more than that of ‘Cheongdu1’. The mean yield of ‘Jungmo3005’ in RYT was 256kg/10a which was 97% of the yield of ‘Cheongdu1’. ‘Jungmo3005’ is expected to be widely used as a breeding parent to cross with other varieties and lines for creating colored soybean cultivars with tolerance to lodging, shattering and bacterial pustule.

Mungbean Cultivar, ‘Sanpo’ with Medium-Large Grain and High Yielding 中粒高产绿豆品种‘三坡’

A mungbean cultivar ‘Sanpo’ (Vigna radiata (L.) Wilczek) was derived from the cross between SM9710-3B-7 and Jangannogdu at the Jeollanamdo Agricultural Research and Extension Services (JARES) in 2012. ‘Sanpo’ has erect growth habit, green hypocotyls, heart leaflet, dull green seed surface, and black and straight pod when matured. The stem length of ‘Sanpo’ were 71cm, which was 5cm longer than check cultivar ‘Owool’. The pods number per plant, the seeds number per pod, and the seed weight of ‘Sanpo’ are similar to check cultivar. It has field resistance to cercospora leaf spot, mungbean mottle virus, and powdery mildew. In addition, ‘Sanpo’ has high lodging resistance. The hard seed rate of ‘Sanpo’ was 2.4%, which was 0.8%point lower than check cultivar. The sprout yield ratio of ‘Sanpo’ was 8.26 times, which was 10% (77%point) more than check cultivar. The average yield of ‘Sanpo’ was 1.62ton/ha, which was 14% more than the check cultivar ‘Owool’. (Cultivar registration number: 5250)

Soybean Cultivar, “Choseon” : Cultivar with Yellow Medium Seed Size and Lipoxygenase 2,3 Protein-free 大豆品种“Choseon”:黄色、中等大小、不含脂氧合酶2,3蛋白的品种

Soybean [Glycine max (L.) Merr.] is an important source for protein, oil, carbohydrates, isoflavones, and many other nutrients to humans and animals. Lipoxygenase protein is the main antinutritional factor in mature soybean seed. Soybean cultivar, "Choseon" with yellow medium seed size and free of lipoxygenase 2,3 protein was developed. "Choseon" was selected from the population derived from the cross between cultivar, "Jinpum 2" and germplasm, PI408155. "Choseon" has purple flowers, tawny pubescence, a determinate growth habit, and light yellow pods at maturity. The seed of "Choseon" has yellow hilum and yellow seed coat color. Seed protein and oil content on a dry weight basis were 34.2 and 16.9%, respectively. "Choseon" has shown a resistant reaction to soybean necrosis, soybean mosaic virus, Cercospora leaf spot and blight, black root rot, pod and stem blight, and bacterial pustule. "Choseon" matured in 5 October with plant height of 51 cm and a 100-seed weight of 15.1 g. Average yield of "Choseon" was 2.66 (Ton/ha) on the regional yield trials of 2013 at 4 locations. "Choseon" has been registered as a soybean cultivar (registration number: 5985, registration date: April 11, 2016) by Korea Seed & Variety Service, Republic of Korea.

Assessment of Genetic Diversity and Identification of Korean Wheat Varieties using RAPD Markers 韩国小麦品种遗传多样性评价及RAPD标记鉴定

Eleven RAPD primers were assessed to analyze genetic diversity of Korean wheat varieties and to develop DNA marker for cultivar identification. The average of the number of polymorphic bands was 5.2 and PIC values showed 0.48, respectively. Ten major clades were presented by phylogenetic analysis. Three cultivars containing Uri, Hanbeak and Jonong were distinct from the others in the phylogenetic dendrogram. Seven cultivar-specific fragments were detected from 11 RAPD fingerprinting among 35 wheat cultivars and they were sequenced. Four Korean wheat cultivars, Eunpa, Jopoom, Yeonbaek and Jeokjoong, were identified newly by four markers, 84, 173, 174 and KWSM011. We convince that these new DNA markers are useful for cultivar fingerprinting and are applied to marker-assisted selection in wheat breeding program.
