Optimization of the ultrasonic-assisted enzymatic extraction of polysaccharides from Dendrobium offcinale Kimura et Migo and bioactivity study 超声辅助酶法提取铁皮石斛多糖的工艺优化及生物活性研究

Ultrasonic-assisted enzymatic extraction (UAEE) technology was employed to extract the polysaccharides from Dendrobium offcinale Kimura et Migo (D. offcinale). The extraction conditions were optimized by central composite design (CCD) found in the response surface methodology (RSM). The polysaccharide yield (34.05 ± 1.03%) was well matched with the predicted value, as obtained under the optimum parameters: enzyme activity of 2040 U/g, a liquid-solid ratio of 71 mL/g, and a time of 84 min. Compared to other extraction methods, UAEE was able to yield D. offcinale polysaccharide (DOP) highly efficiently in a shorter extraction time period and at a lower extraction temperature. The extracted polysaccharide (DOPUAEE) showcased a better nitrite scavenging ability, a high antioxidant activity, and more potency in inhibiting α-glucosidase, expanding its versatility in food applications.

Protective effect of bamboo root dietary Fibre on hyperlipidaemia mice induced by high-fat diet 竹根膳食纤维对高脂日粮致高脂血症小鼠的保护作用

Bamboo root as by-product of bamboo shoot processing is rich in dietary fibre. The protective effects of bamboo root dietary fibre (BRDF) with different intake dose on the hyperlipidaemia mice were investigated. The mice weight in the high dose (HD) group was 31.18 g, lower than that of high fat diet (HFD) group (33.71 g, p < .05), after consuming BRDF. The contents of TC, TG and LDL-C in HD group mice were significantly reduced (p < .05). The mice liver index significantly decreased in the BRDF group compared to the HFD group. TC and TG contents in the liver of HD mice were 0.85 mmol·g−1 and 0.83 mmol·g−1, respectively, significantly lower than 2.25 mmol·g−1 and 1.46 mmol·g−1 in the normal control group (p < .05). H&E staining showed a reduction in inflammatory infiltration. These results suggested that BRDF in the daily diet could reduce blood lipids in vivo and prevent hyperlipidaemia.

Targeting disorders of glucose metabolism and the occurrence of hyperglycemia provides a potential therapeutic strategy for balancing energy to reduce insulin resistance such as type 2 diabetes. The present study investigated whether theasaponin E1 regulate energy metabolism in skeletal muscle cells. C2C12 mouse myoblasts were differentiated into myotubes . An MTT colorimetric assay verified cell viability. Theasaponin E1 (30, 45 or 60 mg/ ml), or metformin (2.5 mM), insulin (150 nM) for reference were used to treat the C2C12 myotubes. The expressions of energy metabolism were measured by western blotting in C2C12 myotubes. 2-NBDG kit to detect the glucose uptake capacity. Theasaponin E1 Treatment increases glucose uptake,GSK 3 β, as well as increases to the glycogen synthesis through the PI3K/AKT/mTOR signaling pathway, while reducing AMPK and ACC activities, and reducing the adipose synthesis pathway. These results suggest that theasaponin E1 may be increases glucose uptake by improving muscle energy metabolism.

The effects of propolis on doxorubicin-induced hepatorenal damage: a comparison of ethanolic and oily extracts of propolis 蜂胶对阿霉素诱导的肝肾损害的影响:蜂胶乙醇提取物和油提取物的比较

The protective effect of ethanolic propolis extract (PE) in the toxic effects of doxorubicin (DOX), a chemotherapy drug, has been reported previously. Since ethanol usage has some restrictions, in the present study, olive oil as an alternative to ethanol has been tested in DOX-induced hepatorenal injury and the effectiveness of the two extracts were compared. Male rats were orally given PE or oily propolis extract (PO) for 15 days (50 mg/kg/day) and DOX was injected (15 mg/kg, intraperitoneally) on the 13th day. The animals were sacrificed and then hepatorenal oxidative stress and functional parameters were evaluated. DOX-induced reduction in hepatorenal functions and oxidative stress were significantly reduced in both propolis treatment groups. Treatment with PO was more effective in reducing DOX-induced oxidative stress, yet both extracts showed similar efficacy in improving hepatorenal functions. We suggest that the PO could be preferred to avoid the side effects of ethanol.

Development and optimization of methylcellulose-based edible coating using response surface methodology for improved quality management of ready-to-eat pomegranate arils 基于响应面法的甲基纤维素可食性涂层的开发与优化,以改善即食石榴皮的质量管理

In this study, the methylcellulose edible coating additives, namely, salicylic acid (SA) (1–2.5 mM), canola oil (0.05–0.1%) and tween-80 (0.05–0.1%) were optimized using RSM to improve quality management of pomegranate arils. The central composite design was used, and the response variables included mass loss (ML), total soluble solids (TSS), total phenolic content (TPC) and 2,2-Diphenyl-1-picrylhydrazyl (DPPH scavenging activity). The results showed that ML was affected by the quadratic terms of canola oil, tween-80 and SA, whereas TSS was affected by the quadratic terms of canola oil and tween-80. The interaction of SA and canola oil showed a significant effect on TSS and TPC, whereas DPPH scavenging activity was affected by linear terms of SA and canola oil. The optimized conditions for the independent variables were 1.55 mM, 0.079% and 0.079% for SA, canola oil and tween-80, respectively. The predicted responses were consistent with experimental values, indicating the model accuracy and suitability.

This research aimed to determine the minimum amounts of chicken blood plasma (X1) and sorbitol (X2) that affect the protein solubility (Y1), cohesiveness (Y2), and whiteness (Y3) of surimi. Response surface methodology (RSM) generated 13 experimental designs ranging from 0.5% to 2.5% for chicken plasma protein (CPP) and 2% to 6% for sorbitol. The results show that optimal conditions for protein solubility, cohesiveness, and a whiteness of surimi were achieved with a CPP concentration of 0.79% and a sorbitol concentration of 4.68%. This resulted in a predicted protein solubility of approximately 49.09 mg/ml, cohesiveness of 0.654 and whiteness value of 75.55. Meanwhile, the optimal experimental values for protein solubility, cohesiveness, and whiteness were found to be 46.56 mg/ml, 0.65, and 75.55, respectively. The measured values and predicted values have no statistically significant difference (p > .05), indicating the validity of the RSM model. These findings are valuable for improving surimi production.

Untargeted metabolomics and quantitative label-free proteomics analysis of whole milk protein from Chinese native buffaloes 中国本土水牛全脂牛奶蛋白的非靶向代谢组学和定量无标记蛋白质组学分析

This study aimed to analyze metabolite abundances and proteome differences between buffalo milk from Binglangjiang (BBM) and Dehong (DBM). Untargeted UPLC-MS/MS, label-free proteomics approach, and bioinformatics analyses including GO, KEGG, and PPI were performed. The results revealed that catechol, L-proline, γ-aminobutyric acid, hippuric acid, creatinine, and betaine were detected in BBM and DBM as the major metabolites. One hundred thirteen proteins, mainly within the molecular weight range of 20–80 kDa, were identified, and the differential protein numbers of BBM were higher than those of DBM. These proteins were primarily involved in cholesterol metabolism, Staphylococcus aureus and Salmonella infections, the peroxisome proliferator-activated receptor signaling pathway, and coagulation cascades. Relative abundances of α-s1-casein, α-s2-casein, and β-casein were plentiful in BBM, while κ-casein, albumin, and α-lactalbumin were in DBM. PPI network analysis indicated that the identified proteins were highly interconnected. These findings promote a better understanding of quality biomarkers in dairy product improvements.

Changes in physicochemical and conformational properties of myofibrillar proteins isolated from mandarin fish (Siniperca chuatsi) treated by atmospheric pressure plasma jet 常压等离子体射流处理鳜鱼肌纤维蛋白理化和构象特性的变化

The effects of atmospheric pressure plasma jet (APPJ) treatment on the physicochemical and conformational properties of myofibrillar protein (MP) isolated from mandarin fish. The findings revealed that after exposing MP to APPJ for 0–12 s, there was a significant (P < .05) decline in the solubility and total sulfhydryl concentration. Turbidity and surface hydrophobicity increased significantly, while pH value decreased significantly at first but then stabilized. Gel electrophoresis confirmed that the extracted protein was unaffected. FTIR spectroscopy indicated that the APPJ treatment caused slight changes in secondary structure. However, the UV absorption spectral and intrinsic tryptophan fluorescence demonstrated that the tertiary structure of MP was altered following APPJ treatment. According to the rheological characteristics, APPJ treatment could improve protein cross-linking, raise storage modulus, and promote the development of protein gel network structure. These findings demonstrated that APPJ treatment might improve the functional characteristics of MP isolated from mandarin fish.

Physicochemical properties and process contaminants analysis of flaky egg-free rolls incorporated with buckwheat ( Fagopyrum esculentum Moench) 荞麦(Fagopyrum esculentum Moench)无蛋卷的理化性质及工艺污染物分析

The fortification of buckwheat (Fagopyrum esculentum Moench) with rheological characteristics, nutritional properties, and formation of contaminants in wheat flour-based flaky rolls was studied. Higher content of buckwheat flour increased the antioxidant capacity, and phenolic, protein, fiber, rutin, and quercetin contents, leading to the development of a nutritionally improved product. The contents of the process contaminants, for example, the acrylamide, hydroxymethylfurfural (HMF), and furfural ranged from 147.25–262.19 μg/kg, 19.17–73.56 mg/kg, and 1.34–4.55 mg/kg, respectively, when buckwheat was added in the range of 0–30%. Parallelly, the production levels of methylglyoxal (MGO) and glyoxal (GO) were 4.09–23.55 and 0.69–2.05 mg/kg in the buckwheat substitution range of 0–30%. Shortly, even though incorporating 10% buckwheat flour had the best sensory scores, 20% was the most suitable substitution rate in terms of controlling the contaminants produced during processing while enhancing the nutritional properties of flaky rolls.

Streamlined approaches for image classification using principal component analysis and hierarchical clustering of extrudates from coffee and sorghum blends 利用主成分分析和从咖啡和高粱混合物中挤出物的分层聚类简化图像分类方法

This article describes simple methods to group images including principal component analysis (PCA) and hierarchical clustering of principal components (HCPC). Images of expanded and low expanded extrudates were processed using two optimization alternatives: a) image size reduction (from 2126 to 25 pixels); and b) grayscale conversion before size reduction. After applying PCA and HCPC, all tests yielded consistently similar results with the same PCA distribution and identical HCPC groups. Furthermore, expanded and low expanded extrudates formed groups with their respective peers. The RAM allocated to images and the time required to process them was reduced from 1727 Mb to less than 5 Mb and from ~ 2000s to just 0.1s, respectively. These results demonstrate the feasibility of using these two simple multivariate statistical techniques for image classification.
