Effects of different extraction methods on the physicochemical properties and biological activities of polysaccharides from maca roots 不同提取方法对玛咖根多糖理化性质及生物活性的影响

Maca is rich in bioactive compounds and has considerable potential for commercial application as a functional food material. In this study, maca root polysaccharides (MRPs) were prepared via hot water extraction, enzyme-assisted extraction, ultrasound-assisted extraction (UAE), and deep eutectic solvent (DES)-based UAE, and their physicochemical properties and biological activities were compared. The MRP obtained using DES-based UAE (MRP-DU) exhibited the highest yield (30.85%), oil-holding capacity (5.21 g/g), emulsifying capacity (50.58%), and emulsion stability (48.14%). Additionally, MRP-DU exhibited the highest antidiabetic and absorption-retarding effects on glucose and bile acid. These results suggest that MRP obtained using DES-based UAE have high application potential as functional food materials and could also serve as antidiabetic and cholesterol-lowering agents.

Effects of different treatment subjects of Lactiplantibacillus plantarum SCS5 on oxidative stress in type 2 diabetic mice 不同处理对象植物乳杆菌SCS5对2型糖尿病小鼠氧化应激的影响

The purpose of this study was to determine the effect of different subjects from Lactiplantibacillus plantarum SCS5 (L. plantarum SCS5) on type 2 diabetes by modulating oxidative stress levels.The normal group (NG) and the model group (MG) of mice were fed normally, while the three test groups (LP1,LP2,LP3) received 10 ml/kg (body weight) of the L. plantarum SCS5 suspension, intracellular material, or heat-killed intracellular material daily for 10 weeks. The weight loss and the increase in blood glucose were alleviated in the LP2 and LP3 groups. Insulin, catalase, superoxide dismutase, and glutathione peroxidase significantly increased in the LP3 group of mice compared with the MG group. Additionally, the pathological sections showed that the lesions in the LP3 group were less severe than those in the MG group. These findings indicate that the heat-killed intracellular L. plantarum SCS5 material was most effective in alleviating oxidative stress in type 2 diabetic mice.

Development of two biosensors to detect biogenic amines was carried out using immobilized diamine oxidase from Pisum sativum on 3-APTMS-functionalized SiO2 substrates with either glutaraldehyde or terephthalaldehyde as crosslinkers. The assembly process was validated by FT-IR, SEM-EDX, and AFM. Biosensor responses to biogenic amines were c.a. 3-fold lower compared to the free enzyme kinetics as assessed by UV/VIS spectrophotometry. The calculated initial velocities (ΔAbs·min-1) for glutaraldehyde and terephthalaldehyde biosensors in the given order were (3.2 ± 0.68) × 10–2, (0.48 ± 0.09) × 10–2 for putrescine; (4.6 ± 1.2) × 10–2, (0.35 ± 0.47) × 10–2 for cadaverine; (5.8 ± 2.4) × 10–3, (19.0 ± 155) × 10–3 for spermidine, and (3.0 ± 2.0) × 10–4, (0.0 ± 7) × 10–4for histamine. The 2-way t-tests indicated that the crosslinker type affected the catalytic activity of immobilized diamine oxidase as the responses of glutaraldehyde biosensors to putrescine and cadaverine were significantly higher than those of the terephthalaldehyde. Molecular Docking simulations showed that glutaraldehyde has a stronger affinity for DAO’s lysine and arginine residues while terephthalaldehyde to phenylalanine.

Health compromising components in French fries and fried chicken available in the markets of Dhaka city, Bangladesh 孟加拉国达卡市市场上出售的炸薯条和炸鸡中有害健康的成分

French fries (FF) and fried chicken (FC) are known contributors to overweight and obesity among adolescents worldwide. Top selling five brands for both FF and FC were analyzed to determine their health compromising contents, which were sampled from the markets of Dhaka, Bangladesh. Sodium (Na) was determined by flame photometry, trans fatty acids (TFAs) by gas chromatography and heavy metals like arsenic (As), lead (Pb), cadmium and chromium by graphite furnace atomic absorption spectrophotometry. The median concentration of Na, TFA, As and Pb in FF was 0.45 gm/100 gm, 0.11 gm/100 gm, 0.93 mg/kg and 0.03 mg/kg, respectively. Similarly, in FC the median concentration of Na, TFA, As and Pb was 0.46 gm/100 gm, 0.13 gm/100 gm, 0.053 mg/kg and 0.06 mg/kg, respectively. The study findings suggests that consuming FF and FC impose a health risk among the exposed population.

Dihydromyricetin promotes LDL metabolism in HepG2 cells through the PCSK9/LDLR pathway 二氢杨梅素通过PCSK9/LDLR途径促进HepG2细胞的LDL代谢

Low-density lipoprotein receptor (LDLR) and proprotein convertase subtilisin/kexin type 9 (PCSK9) play a pivotal role by regulating plasma low-density lipoprotein cholesterol (LDL-c) levels. Dihydromyricetin (DMY), the most abundant natural flavonoid in rattan tea, has proven anti-atherogenic effects, but the underlying molecular mechanisms remain poorly understood. Therefore, we studied the effects of DMY on LDLR and PCSK9. The results showed DMY promoted LDLR protein and mRNA expression and increased LDL uptake in HepG2 cells. DMY inhibited intracellular PCSK9 protein and mRNA expression. And it also significantly reduced PCSK9 levels in the cell culture medium. Furthermore, DMY inhibited the expression of PCSK9 through the liver nuclear transcription factor 1 A (HNF1A) and increased the protein expression of LDLR. Taken together, our results support the idea that DMY regulates LDL-c metabolism through PCSK9/LDLR signaling. This study reveals the potential mechanism of DMY’s anti-atherogenic effect and provides a theoretical basis for dietary DMY supplementation.

Simvastatin retards cartilage degradation and subchondral bone deterioration induced by a high-fat diet in mice 辛伐他汀延缓小鼠高脂肪饮食诱导的软骨退化和软骨下骨退化

To evaluate the effects of simvastatin on the degeneration of cartilage and subchondral bone in a mice model of osteoarthritis (OA) induced by obesity. Thirty male 3-month-old C57BL/6J mice were randomized into three groups: mice with normal diet + vehicle (distilled water) (Control), mice with High-fat diet + vehicle (HFD), mice with HFD + Simvastatin (HFD+S). HFD+S group were treated with simvastatin (10 mg/kg/day) for 12 weeks. The pathology of OA was assessed by histomorphology analyses, immunohistochemistry, micro-computed tomography, and enzyme-linked immunosorbent assay. Histomorphological analysis revealed that OA was significantly exacerbated by the HFD-induced obesity and markedly alleviated by the simvastatin intervention. In details, simvastatin ameliorated the abnormal metabolic status and cartilage lesions, significantly increased aggrecan and collagen-II expression and decreased the expression of MMP-13 and leptin in cartilage. Furthermore, the results of micro-CT analysis revealed that the HFD+S group exhibited higher BMD, BV/TV, and Tb.N values but a lower Tb.Sp value than that of the HFD group. Serum glucose, leptin, and IL-1β concentrations were significantly correlated with the OARSI score. Histomorphological analysis revealed that OA was significantly exacerbated by the HFD-induced obesity and markedly alleviated by the simvastatin intervention. In details, simvastatin ameliorated the abnormal metabolic status and cartilage lesions, significantly increased aggrecan and collagen-II expression and decreased the expression of MMP-13 and leptin in cartilage. Furthermore, the results of micro-CT analysis revealed that the HFD+S group exhibited higher BMD, BV/TV, and Tb.N values but a lower Tb.Sp value than that of the HFD group. Serum glucose, leptin, and IL-1β concentrations were significantly correlated with the OARSI score. The HFD-induced obesity aggravates articular degeneration and deterioration in subchondral bone, which could be improved by the intervention of simvastatin, suggesting that simvastatin may be a potential candidate for amelioration of the progression of obesity induced-OA.

This research studies the impacts of microcrystalline cellulose (MCC) concentrations on the physicochemical properties of spray-dried water-in-oil-in-water (W/O/W) vitamin C double emulsion powder Four MCC concentration (1%, 1.5%, 2%, and 2.5%) were used to produce the vitamin C double emulsion powder. The results of the emulsion’s creaming index, flow behaviour index, zeta value, and particle size indicated that these parameters decreased when the MCC concentration increased. Scanning electronic microscopy (SEM) analysis showed that the higher MCC concentration resulted in a smoother surface In general, the MCC concentration increased, the particle size, hygroscopicity, solubility, water content, and water activity of the double emulsion powder decreased. Double emulsion powder containing 2% MCC presented the highest level of antioxidant activity and was mostly stable after storage for 56 days. Before and after drying, transmission electron microscope (TEM) analysis of the morphological characteristics of the liquid emulsion showed that the concentration of 2% MCC in the emulsion could form a W/O/W double emulsion structure. In conclusion, the addition of MCC significantly affect the properties of vitamin C double emulsion.
