Preliminary DNA Barcode Report on the Marine Red Algae (Rhodophyta) from the British Overseas Territory of Tristan da Cunha 英属特里斯坦达库尼亚海域红藻DNA条形码初步报告

ABSTRACT The current manuscript is the first in a series intended to publish accumulating DNA barcode data to make them accessible to the scientific community. Focused on 135 specimens of red algae from the remote islands of Tristan da Cunha, part of the British Overseas Territory of Saint Helena, Ascension and Tristan da Cunha, the 47 (possibly 48; see notes with Lophurella sp. 1Tris) genetic groups uncovered during this project are compared to the only detailed floristic work for this region completed by Baardseth in 1941. A number of taxonomic anomalies are reported with indications for eventual solutions that await study of the type material of the associated morphospecies. Species previously assigned by Baardseth to the genus Epymenia Kützing are formally transferred to Rhodymenia Greville as R. elongata (Baardseth), comb. nov., (including E. marginifera Baardseth) and R. flabellata (Baardseth), comb. nov. A number of range extensions are reported including species such as Ceramium secundatum (Lyngbye) C.Agardh, Colaconema caespitosum (J.Agardh) Jackelman, Stegenga & J.J.Bolton, Helminthocladia calvadosii (J.V.Lamour. ex Duby) Setch. and Porphyra mumfordii S.C.Lindstrom & K.M.Cole, which have likely been distributed by human activities. We also note that the sporophyte of the supposedly narrowly distributed Schimmelmannia elegans has been collected from both British Columbia, Canada, and Queensland, Australia, consistent with other observations that sporophytes of red algal species with alternations of heteromorphic generations are commonly more broadly distributed than the gametophytic stage. This species, although originally described by Baardseth from these mid Atlantic islands, may also be introduced.

ABSTRACT This paper provides information on the morphology, reproduction and distribution of a freshwater brown alga, Porterinema fluviatile (Porter) Waern (Phaeophyceae). The alga was recorded in oligotrophic water from Mlava Spring, a karst limnocrene in eastern Serbia (44°11′298″N, 21°47′021″E). This is the first record of freshwater brown algae in general in Serbia, and the first record of P. fluviatile in southeastern Europe. The thallus of P. fluviatile formed small, dark-brown tufts, with multicellular, uniseriate filaments that were irregularly branched. An anastomosis was noticeable between some filaments. The vegetative cells of the young filaments were cylindrical (17-38 µm long × 2.5-4.0 µm wide). The cells had 1 or 2, rarely 3, plate-like parietal plastids. The reproductive organs of P. fluviatile (asexual reproduction) originate from the transformation of vegetative cells. Elongated vegetative cells develop inflated vegetative cells (5-10 µm in diameter) with lipid droplets. Further transformation of these cells leads to the formation of intercalary plurilocular sporangia with 4 to 8 (rarely 16) cells. Nonmotile, round aplanospores, which develop by protoplast division in elongated vegetative cells, are described for the first time in this species.

ABSTRACT The dinoflagellate Symbiodinium sp. establishes symbiotic relationships with the anemone Exaiptasia diaphana Rapp. The process that leads to the reciprocal recognition of the two symbiotic partners is still not very well understood. We hypothesize that chemical signals are exchanged between the Exaiptasia diaphana-Symbiodinium holosymbiont and aposymbiotic anemones or between free living Symbiodinium and holo- and aposymbiotic Exaiptasia, leading to changes in organic and elemental compositions in the aposymbiotic anemones. In order to test these hypotheses, bleached anemones were exposed to the presence of either free living Symbiodinium, previously extracted from the same Exaiptasia clone, or to holobionts. The ex-hospite algae and the holobionts were included in dialyses membranes with a cut-off of 14 000 Da. In the control treatments, the experimental samples were exposed to the presence of empty dialysis tubes. The organic composition and the elemental composition of the anemones were determined by Fourier Transform Infrared Spectroscopy and Total Reflection X-Ray Fluorescence Spectroscopy/Gas Chromatography, respectively. The fact that both the organic and elemental composition of the experimental aposymbiotic anemones differed significantly from the controls, in the absence of any obvious nutritional effect, is suggestive of an exchange of chemical signals between the aposymbiotic and holosymbiotic anemones.

ABSTRACT Pigment variation within and among algal species may have important ecological consequences because small changes in the concentration and composition of pigments can influence the photosynthetic efficiency and rate as well as the spectra of light utilized. Toward the goal of developing a rapid method for comparing pigment composition among algal thalli, we characterized the relationship between visual color information taken from photographs (e.g., red, green, and blue color values) and photopigment composition in the non-native red alga Agarophyton vermiculophyllum (Ohmi) Gurgel, J.N.Norris & Fredericq. We used a set of 19 thalli, collected from across the known native and non-native range in the Northern Hemisphere, which exhibited substantial color variation at the time of field collection, and sustained this variation after being maintained in a common garden. We identified a set of ecologically interesting pigment traits that are readily predicted by color information, including chlorophyll a and phycobilin concentration. Finally, we demonstrated the repeatability of estimating color phenotypes from photographs of thalli taken under a range of light conditions in order to evaluate the utility of this approach for field studies. We suggest this method could be useful for the rapid, high-throughput phenotyping of photopigments in other red algae as well.

ABSTRACT The genus Meristotheca J. Agardh is the second largest in Solieriaceae with 13 species widely distributed whose thalli are mainly flattened. On the basis of molecular and morpho-anatomical evidence, M. spinella Núñez-Resendiz, Dreckmann & Sentíes, sp. nov. is the second species in the genus with cylindrical axes to be described from the southwestern Gulf of Mexico. rbcL sequences and genetic distance values indicate its separation from other species within Meristotheca and from other morphologically similar species occurring in its known distribution range as M. cylindrica Núñez-Resendiz, Dreckmann & Sentíes. The numerous short branchlets that cover the margins of main axes and branches, among other relevant anatomical and reproductive characters, is the main difference from M. cylindrica, its sister species.

Light and scanning electron microscope observations are provided on four new Hannaea Patrick species from Asia. The areas in Asia the new taxa are described from include Tibet, Mongolia and the Pamir mountainous region in western Asia. Both subgroups of the genus are represented, including H. tibetiana Q. Liu, Glushchenko, Kulikovskiy & Kociolek, sp. nov. and H. pamirensis Glushchenko, Kulikovskiy, Q. Liu & Kociolek, sp. nov., from the group that includes the generitype and has striae composed of single rows of areolae and one rimoportula per valve. Hannaea dorofeyukae Kulikovskiy, Glushchenko, Q. Liu & Kociolek, sp. nov. and H. mongolica Glushchenko, Kulikovskiy, Q. Liu & Kociolek, sp. nov. have striae composed of two rows of areolae and two rimoportulae; one at each end of the valve. The group with two rimoportulae per valve and biseriate striae is known only from Lakes Baikal, Khovsgol and the surrounding areas in Asia. New combinations are proposed for four other species. These taxa are also known from Asia. Two of them are known from Japan, one is from China and one is from Lake Baikal in Russia. It appears that Asia is an important center of diversity for Hannaea.

ABSTRACT We present the description of Mallomonas teres, sp. nov., from three localities, two peat-bogs in Northern Sweden and a peat-bog pool in the Polessian Nature Reserve, Northern Ukraine. The sites ranged from acidic to neutral pH, were low in conductivity, and contained other pH-indifferent or low-pH chrysophyte taxa. According to the scale structure M. teres, sp. nov., belongs to the section Heterospinae within the genus. The most distinctive scale character discerning M. teres, sp. nov., from the other species of the section Heterospinae is the distal part of the scale, which has a smooth dome and numerous parallel curved ribs. Mallomonas teres, sp. nov., is probably distributed at least in temperate, acidic European localities. Its rare occurrence could be a result of a poor dispersal capacity combined with a low growth rate.

ABSTRACT Macrocystis C. Agardh, Durvillaea Bory and Lessonia Bory are three brown seaweeds genera of commercial importance for Chilean fishermen. Macrocystis pyrifera (Linnaeus) C. Agardh (Laminariales, Laminariaceae) is one of the world's most ecologically and economically important seaweed collected in Chile for alginate extraction; and as food for abalone aquaculture, Lessonia spicata (Suhr) Santelices (Laminariales, Lessoniaceae) represents more than 70% of the total brown seaweeds annual landing in Chile; and Durvillaea antarctica (Chamisso) Hariot (Fucales, Durvillaeaceae) is consumed as food, being considered very healthy because of its iodine content. Despite the economic importance of these species their nuclear DNA content is unknown until this moment. The present research was initiated to determine the nuclear DNA content and the intraindividual ploidy level variation in these seaweeds. The DNA-localizing fluorochrome DAPI (4′,6-diamidino-2-phenylindole) and red blood cell (chicken erythrocyte) standard were used to estimate the nuclear DNA contents by image analysis. Durvillaea antarctica presented lower 2C DNA (1.2 pg) content and narrower range of DNA contents (1C-2C) than the Laminariales, which showed higher 2C DNA contents (1.4 -1.5 pg) and a wider range of ploidy level, achieving up to 4C in L. spicata and up to 8C in M. pyrifera. This higher ploidy levels observed would be related with the more complex cortical organization in the Laminariales than D. antarctica. 1C nuclei were only found in mature antheridia (D. antarctica) and sporangia (M. pyrifera and L. spicata). The 2C values observed for the taxa assessed were in the range of other previously published Fucales and Laminariales.

ABSTRACT Tunisia is located at a key position in the Mediterranean Sea where it constitutes a transitional area between the eastern and western basins benefitting from rich habitat diversity. The most recent inventory of marine macrophytes dated back to 1987 with an update in 1995, however these inventories were performed with identification based on morphological observations, which can be misleading for many seaweed species. To the best of our knowledge, this is the first study of the Tunisian macroalgal flora using DNA barcoding methods. Our target was to carry out a survey of macroalgae, to reveal cryptic species and allochthonous introductions, and to identify taxa with problematic taxonomy. COI-5P sequence analysis resolved 26 genetic species, including two new reports and three alien species. Of the remaining taxa, several displayed cryptic diversity relative to species reported from the Mediterranean versus from other geographical regions with taxonomic and nomenclature updates provided where possible. The aim of the present paper was to provide a DNA barcode-assisted floristic list of florideophycean algae collected along the Tunisian coast.
