Identification of polymorphisms in TYRP1, DCT and RAB38 genes and their association with coat color in alpacas 鉴定 TYRP1、DCT 和 RAB38 基因的多态性及其与羊驼毛色的关系

The goal of this study was to characterize and investigate the effect of single nucleotide polymorphisms (SNPs) or InDels in tyrosinase related protein 1 (TYRP1), dopachrome tautomerase (DCT), and Ras-related protein 38 (RAB38) genes on coat color in South American camelids. TYRP1 is among one of the major regulators of pigmentation in mammals. DCT is a pigment cell-specific gene that plays an important role in the melanin synthesis pathway and the expression of skin color. RAB38 is involved in the transport of tyrosinase (TYR) and TYRP1, which are key enzymes in melanin production.Five fleece color tones from 327 alpacas (white, black, brown, gray and light fawn) were investigated to characterize and identify polymorphisms that play a role in coat color. Among the SNPs and Indels identified in the coding region, statistical analysis was performed to determine the association between the polymorphisms and fleece color in camelids, as well as the determination of allelic frequencies.Twenty two polymorphisms in the coding region, seven of them were nonsynonymous, eleven were synonymous, three located in the 5’UTR region and one deletion in the 3’UTR region were identified in the TYRP1 gene; in the DCT gene were identified twenty two polymorphisms, where five of them were non-synonymous, twelve were synonymous, one polymorphism in the 5’UTR region and four in the 3’UTR region; in adition, in the RAB38 gene six polymorphisms were identified, one was nonsynonymous, two were synonymous, in addition to three polymorphisms in the 3’UTR region were found in the RAB38 gene. There were low to moderate polymorphisms in the populations. Significant differences (P<0.001) were found with color fiber in the alpaca populations studied for TYRP1 and DCT genes.

Alterations in energy balance and the antioxidant system due to heat stress: focus on animal production 热应激导致的能量平衡和抗氧化系统变化:关注动物生产

In areas where the ambient temperature is above the thermoneutral zone, the economic loss from animals for human consumption, such as pigs, is worrying. Heat Stress (HS) increases body temperature and causes a subsequent reduction in feed intake. This reduction results in the loss of essential nutrients, such as amino acids, which are crucial for growth rate and the activation of vital biochemical mechanisms. However, the effects of HS on reactive oxygen species (ROS) generation, and the activation of mechanisms such as the antioxidant system associated with nuclear factor erythroid-derived 2-like-2 (Nrf2), energy balance, protein synthesis and mitochondrial biogenesis have received limited attention. In this work, we provide a comprehensive review of the existing evidence regarding the significance of nutritional components and their association with HS. We particularly emphasize ROS generation, mitochondrial alterations, and the activation of Nrf2, AMP-activated protein kinase (AMPK), and mammalian target of rapamycin (mTOR) signaling pathways in response to the disruption of homeostasis.

The effect of yeast-derived β-glucans in reducing the adverse outcome of Lawsonia intracellularis in finishing pigs 酵母β-葡聚糖对减少育成猪细胞内劳森氏菌不良后果的影响

Chronic diarrhoea is one of the most recurrent health issues in pig herds. Among Q9 the causative agents of diarrhoea, Lawsonia intracellularis is quite frequent with worldwide distribution leading to detrimental economical losses, as a result of the poor growth rate of the infected pigs. A growing body of evidence has shown that branched yeast b-glucans are excellent inducers of innate immunity memory in humans and in pigs.In this study, yeast branched b-glucans were evaluated as a potential solution to control infection by L. intracellularis in pigs by reducing the shedding of L. intracellularis in the faeces of infected animals. To address this hypothesis, naturally infected grow finishing pigs were divided into two treatment groups (n=80) and fed either with a basal diet (Control) or the same basal diet supplemented with 250 g/ton b-glucans (BG-fed group).Our results have shown that BG-supplemented pigs during the growing and finishing period had significantly increased body weight (BW) by 4.22 kg on average on day 97 and ADG up to 5.6% over the whole experiment (p<5.0 x 10-2) with no significant increase in ADFI. Similarly, supplementation of pigs with BG during the grow-finish phase has significantly reduced (p<4.0x10-4) faecal shedding of the bacteria in infected pigs on day 97. Furthermore, our results indicate that microbial a-diversity was negatively correlated with the presence of L. intracellularis. Animals fed with yeast BG show a reduction of the two genera Sarcina and Catenibacterium and a deeper analysis indicated that the ratio of Fibrobacter to Lacnospiraceae_AC2044 group could be a potential marker for L. Intracellularis susceptibility.This study demonstrated the beneficial effect of adding branched yeast BG in the diet of grow-finish pigs in reducing the dissemination of L. intracellularis between pigs in a given herd.

Effects of different chicken protein sources on palatability in dry adult dog food 不同鸡肉蛋白质来源对干成年狗粮适口性的影响

As the demand for dog food production continues to rise, along with the number of dogs per household in the US, sustainable ingredients must be considered for the pet food industry. Consumers have a negative perspective when it comes to animal by-products in their pet’s food. However, animal by-products contribute a quality source of dietary protein, vitamins, and minerals which contribute to nutritionally complete pet foods. Along with being nutritionally complete, foods must also appeal to the dog while eating. The objective of this study was to produce formulas utilizing four different chicken protein sources, and to evaluate the effects of those protein sources on palatability in dogs. Five treatments were produced, through extrusion, using fresh mechanically deboned chicken at two different inclusion levels: (FMDC14 and FMDC25), dried chicken (DC), chicken meal (CM), and chicken by-product meal (CBPM). Four trained panels of twenty-five adult dogs, made up of majority Beagles, randomly divided by sex, were used for palatability testing using the two-bowl test. Based on the palatability results from this study, the CBPM treatment was preferred (p<0.05) over the FMDC14, FMDC25, and DC treatments. The CBPM treatment was at parity to the CM treatment (p>0.05). It can be concluded that the CBPM treatment was considered palatable to the dogs.

Links between animal welfare and “One Health”: perception and implementation in Latin America 动物福利与 "同一健康 "之间的联系:拉丁美洲的认识与实施

The recent COVID-19 pandemic has highlighted the crucial role of the “One Health” (OH) concept in the prevention, early detection, and mitigation of health issues involving humans, animals, and the environment. Recognizing the intrinsic interdependence among human health, animal health, and environmental well-being is crucial, demanding heightened emphasis. Many health challenges stem from situations that compromise animal welfare (AW), human well-being, environmental sustainability, and vice versa. Recognizing the significance of AW across its five domains is essential for preventing future pandemics and advancing global objectives such as food security, reduction of human suffering, biodiversity conservation, and enhanced productivity in the agricultural-livestock sector.From March to May 2023, we conducted an online survey using QuestionPro® to explore perceptions, experiences, ongoing strategies, and activities within disciplines linked to AW and the OH approach in Latin America. Respondents included stakeholders from various disciplines associated with OH and AW.Our findings reveal that both concepts are currently undergoing an expansion phase in Latin America. However, the interdependencies between AW and OH are not widely recognized. While animal welfare frequently receives significant attention from diverse educational perspectives, the OH approach has gained considerable momentum over the past decade. Academic sectors and non-governmental organizations (NGOs) are driving initiatives bridging AW and OH in Latin America, underscoring the need for robust public policies to ensure sustainable strategies.Advocating for greater awareness of the interdependence between AW and OH, this study highlights knowledge gaps among the scientific community and policymakers. Actively engaging with these connections can foster comprehensive strategies to address global health challenges and enhance overall well-being. The “One Welfare” framework and other integrative frameworks hold promise in strengthening the linkages between AW and OH, facilitating theory translation into practical action. Establishing comprehensive, integrated policies that unite these domains is imperative for addressing complex health challenges and advancing the welfare of both animals and humans. Recognizing the intrinsic interdependence among human health, animal health, and environmental well-being is crucial, demanding heightened emphasis. Many health challenges stem from situations that compromise animal welfare (AW), human well-being, environmental sustainability, and vice versa. Recognizing the significance of AW across its five domains is essential for preventing future pandemics and advancing global objectives such as food security, reduction of human suffering, biodiversity conservation, and enhanced productivity in the agricultural-livestock sector.From March to May 2023, we conducted an online survey using QuestionPro® to explore perceptions, experiences, ongoing strategies, and activities within disciplines linked to AW and the OH approach in Latin America. Respondents included stakeholders from various disciplines associated with OH and AW.Our findings reveal that both concepts are currently undergoing an expansion phase in Latin America. However, the interdependencies between AW and OH are not widely recognized. While animal welfare frequently receives significant attention from diverse educational perspectives, the OH approach has gained considerable momentum over the past decade. Academic sectors and non-governmental organizations (NGOs) are driving initiatives bridging AW and OH in Latin America, underscoring the need for robust public policies to ensure sustainable strategies.Advocating for greater awareness of the interdependence between AW and OH, this study highlights knowledge gaps among the scientific community and policymakers. Actively engaging with these connections can foster comprehensive strategies to address global health challenges and enhance overall well-being. The “One Welfare” framework and other integrative frameworks hold promise in strengthening the linkages between AW and OH, facilitating theory translation into practical action. Establishing comprehensive, integrated policies that unite these domains is imperative for addressing complex health challenges and advancing the welfare of both animals and humans. Genetic parameters of subjective and image-based horn fly abundance phenotypes 基于主观和图像的角蝇丰度表型的遗传参数

The genetic basis of horn fly abundance remains largely unknown and only few heritability estimates are available based on small scale studies. In this study, one subjective and two image-based fly abundance phenotypes were analyzed. Each animal was assessed subjectively for fly abundance by at least one trained agent (SUB). Additionally, several digital images were taken and the best image for each animal was used to assess fly abundance. A box that runs roughly from the withers to the hook and from the chest floor to the fore of the udder was established and all horn flies (PA) and sampling-based estimated number of flies (PR) within the box were calculated. Horn fly counts (SUB, PA, and PR) were discretized into 4 classes based on the quartiles of their respective distributions. Heritability estimates ranged between 0.10 and 0.15 and between 0.14 and 0.16 for the continuous and discrete cases, respectively. The genetic correlation was 0.66 between PA and PR and decreased significantly between the image-based (PA and PR) and the subjective (SUB) assessments for the continuous case. For the discrete case, the genetic correlations between the three traits were very similar, indicating a high concordance between traits.

Mycotoxins are fungal secondary metabolites that can adversely affect animals consuming contaminated feeds. This 71-day feeding trial was conducted to assess the effects of dietary deoxynivalenol plus zearalenone (DON+ZEN = 1.6 + 0.3 ppm), and fumonisins (FUM = 15 ppm), and three adsorbent additives on the production performance, hematological parameters, and liver histology of juvenile Nile tilapia. A mycotoxin-free diet (Control) formulated to contain 35% protein and 8% lipid was spiked with either DON+ZEN or FUM using contaminated corn meals replacing portions of non-spiked corn. Subsequently, three out of four DON+ZEN- and FUM-spiked diets were supplemented (0.5%) with an adsorbent. The research was carried out in a recirculating water system (2,500 L) with a controlled temperature of (25.9 ± 1.1°C), feeding was carried out twice a day at rates ranging from 5 to 8%. The experiment included nine treatments with five replications, each experimental unit consisting of an aquarium with a useful volume of 75-L and fifteen juvenile Nile tilapia (average initial weight of 4.0 ± 0.1 g). Mycotoxin-spiked diets without added adsorbent supported lower final biomass and survival of Nile tilapia relative to control and adsorbent containing diets (P ≤ 0.05). Histological examinations revealed liver inflammation evidenced by lymphocytic infiltration adjacent to pancreatic tissue in fish fed mycotoxin-spiked diets without added adsorbent. Mycotoxin contamination significantly increased HSI (hepato somatic index), which was reverted to the Control value or reduced further by adsorbent addition. Lowest and intermediate hematocrit values were observed in groups fed mycotoxin-spiked diets without and with added adsorbents, respectively. Our results reinforce the importance of using adsorbents and the need to investigate the effect of sub-lethal concentrations of mycotoxins in aquaculture feeds.
