Activity in serotonergic axons in visuomotor areas of cortex is modulated by the recent history of visuomotor coupling. 皮层视觉运动区血清素能轴突的活动受视觉运动偶联的近期历史调节。

Visuomotor experience is necessary for the development of normal function of visual cortex (Attinger et al., 2017) and likely establishes a balance between movement-related predictions and sensory signals (Jordan and Keller, 2020). This process depends at least in part on plasticity in visual cortex (Widmer et al., 2022). Key signals involved in driving this plasticity are visuomotor prediction errors (Keller et al., 2012; Keller and Mrsic-Flogel, 2018). Ideally however, the amount of plasticity induced by an error signal should be a function of several variables - including the total prediction error across all of cortex at that moment, the animal's experience in the current environment or task, stability of the current environment, and task engagement - for optimal computational performance. Candidates for regulators of visuomotor prediction error driven plasticity are the three major neuromodulatory systems that innervate visual cortex in the mouse: acetylcholine, noradrenaline, and serotonin. While visuomotor mismatch acutely triggers activity in noradrenaline (Jordan and Keller, 2023) but not acetylcholine (Yogesh and Keller, 2023) axons in visual cortex, how serotonergic axons in cortex respond to visuomotor mismatch is unknown. Here, we characterized the activity of serotonergic axons in visual cortex (V1) and in area A24b, a motor cortical area in anterior cingulate cortex (ACC), of awake head-fixed mice using two-photon calcium imaging. Our results reveal cortical region-specific responses to visuomotor stimuli in serotonergic axons, but no evidence of a response to visuomotor mismatch. However, average activity in serotonergic axons was modulated by the recent history of visuomotor coupling. We speculate that serotonin could function to regulate visuomotor plasticity as a function of the predictability of the environment with a slow integration time constant.

Factors shaping vaginal microbiota long-term community dynamics in young adult women. 影响年轻成年女性阴道微生物群长期群落动态的因素。

The vaginal microbiota is known to affect women's health. Yet, there is a notable paucity of high-resolution follow-up studies lasting several months, which would be required to interrogate the long-term dynamics and associations with demographic and behavioural covariates. Here, we present a high-resolution longitudinal cohort study of 125 women, followed for a median duration of 8.6 months, with a median of 11 samples collected per woman. Using a hierarchical Bayesian Markov model, we characterised the patterns of vaginal microbiota community persistence and transition, simultaneously estimated the impact of 16 covariates and quantified individual variability among women. We showed that "optimal" (Community State Type (CST) I, II, and V) and "sub-optimal" (CST III) communities are more stable over time than "non-optimal" (CST IV) ones. Furthermore, we found that some covariates - most notably alcohol consumption - impacted the probability of shifting from one CST to another. We performed counterfactual simulations to confirm that alterations of key covariates, such as alcohol consumption, could shape the prevalence of different microbiota communities in the population. Finally, our analyses indicated that there is a relatively canalised pathway leading to the deterioration of vaginal microbiota communities, whereas the paths to recovery can be highly individualised among women. In addition to providing one of the first insights into vaginal microbiota dynamics over a year, our study showcases a novel application of a hierarchical Bayesian Markov model to clinical cohort data with many covariates. Our findings pave the way for an improved mechanistic understanding of microbial dynamics in the vaginal environment and the development of novel preventative and therapeutic strategies to improve vaginal health.

Domestication is a process marked by complex interactions between demographic changes and selective pressures, which together shape genetic diversity. While the phenotypic outcomes of domestication are well documented, its genetic basis-particularly the dynamics of selection-remain less well understood. To investigate these dynamics, we performed simulations designed to approximate the demographic history of large domestic mammals. These simulations used selection coefficients as a modeling tool to represent changes in selection pressures, recognizing that such coefficients are abstractions rather than direct representations of biological reality. Specifically, we analyzed site frequency spectra (SFS) under varying distributions of fitness effects (DFE) and proportions of mutations with divergent selective pressures. Our results show that the discretized deleterious DFE can be reliably inferred from the SFS of a single population, but reconstructing the beneficial DFE and demographic history remains challenging, even when using the joint SFS of both populations. We further developed a novel joint DFE inference model to estimate the proportion of mutations with divergent selection coefficients (p c), although we found that signals of classic hard sweeps can mimic increases in p c, complicating interpretation. These findings underscore both the utility and limitations of DFE inference and highlight the need for caution when interpreting demographic histories in domesticated populations based on such modeling assumptions.

Ecotoxicity of lanthanides to Daphnia magna: insights from elemental behavior and speciation in a standardized test medium 镧系元素对大型蚤的生态毒性:从标准化测试介质中的元素行为和种类中获得的启示

Lanthanides (LNs) are a group of 15 elements with steadily increasing economical importance due to their multiple uses in technologies essential for sustainable ecological, digital and energetic transitions. Although knowledge on LN ecotoxicology has greatly improved over the last decade, uncertainty persists with regard to their actual hazard and risk in freshwater environments. In particular, only limited information is available on i) the actual relationships between LN speciation vs . ecotoxicological responses in standardized laboratory tests and ii) the existence of regular and predictable patterns in LN ecotoxicity (expressed as e.g. , EC 50 ) along the LN series. The present paper provides the first report on the ecotoxicity of all lanthanides (except Pm) for the freshwater crustacean Daphnia magna along with an unprecedented level of detail on LN speciation in the exposure medium. Experimental data show that exposure concentrations can decrease by up to 95 % over the test duration, with the percentage decrease being inversely related with LN atomic mass. Thermodynamic speciation calculations confirm the possible formation of insoluble species, mainly LN carbonates. However, the corresponding theoretical solubility limits do not fully agree with measured concentrations at the end of the tests. Experimental verification of exposure concentrations (as a minimum at the beginning and end of laboratory tests) remains therefore mandatory to reach proper conclusions as to the ecotoxicity of each LN. A decreasing trend in ecotoxicity can actually be observed along the LN series when temporal changes in the exposure concentrations are properly accounted for. However, this trend remains dependent on exposure time and selected exposure metrics. This and other caveats must be considered in future research to reach a community-based consensus for the proper hazard and risk assessment of LN towards daphnids and other aquatic organisms.

Multiproxy analysis exploring patterns of diet and disease in dental calculus and skeletal remains from a 19th century Dutch population 从 19 世纪荷兰人的牙结石和骨骼遗骸中探索饮食和疾病模式的多代理分析

Dental calculus is an excellent source of information on the dietary patterns of past populations, including consumption of plant-based items. The detection of plant-derived residues such as alkaloids and their metabolites in dental calculus provides direct evidence of consumption by individuals within a population. We conducted a study on 41 individuals from Middenbeemster, a 19th century rural Dutch archaeological site. Skeletal and dental analysis was performed to explore potential relationships between pathological lesions and presence of alkaloids. Dental calculus was analysed using ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-ESI-MS/MS). We were able to detect nicotine, cotinine, caffeine, theophylline, and salicylic acid, suggesting the consumption of tea and coffee and smoking of tobacco on an individual scale, which is also confirmed by historic documentation and identification of pipe notches in the dentition. Nicotine and/or cotinine was present in 56% of individuals with at least one visible pipe notch. There is some influence of skeletal preservation on the detection of alkaloids, with higher quantities of compounds extracted from well-preserved individuals, and we observe a positive relationship between weight of the calculus sample and quantity of detected compounds, as well as between chronic maxillary sinusitis and the presence of multiple alkaloids. There are many limitations that will need to be addressed going forward with this type of analysis; we stress the need for more systematic research on the consumption of alkaloid-containing items and their subsequent concentration and preservation in dental calculus, in addition to how mode of consumption may affect concentrations in the dentition. Despite the limitations, this preliminary study illustrates many benefits of using calculus to target a variety of compounds that could have been consumed as medicine or diet. This method allows us to directly address specific individuals, which can be especially useful in individuals that are not always well-documented in historic documentation, such as rural populations, and especially children and women.

Social Network Analysis of Ancient Japanese Obsidian Artifacts Reflecting Sampling Bias Reduction 反映取样偏差减少情况的日本古代黑曜石文物社交网络分析

This study aims to investigate the dynamics of obsidian trade networks during the Jomon period (15,000–2,400 years cal BP), the hunting and gathering era in Japan. To improve regional representation and reduce the distortions caused by small sample sizes, we performed clustering based on a large-scale dataset and conducted social network analysis. The research results revealed that the trade networks during the Jomon period were not constant; they expanded throughout the southern Kanto region during the Middle Jomon period (5,500–4,500 years cal BP) and ceased to function during the Late Jomon period (4,500–3,200 years cal BP). Furthermore, to enhance the readability and inter-pretability of the dataset, we implemented clustering using the density-based spatial clustering of applications with noise (DBSCAN) method. The results showed that in every time division of the Jomon period, the mean intra-cluster cosine similarity of each cluster was higher than the similarity between sites outside the clusters
