A practical exercise is presented for determination of very low amounts (ng/l) of As(III) by flow injection (FI) and hydride generation and atomic absorption spectrometry detection. The exercise is directed towards students at the advanced undergraduate level, and it is intended to be an introduction to procedures of FI, which by relatively uncomplicated equipment provide analysis with excellent performance. With the method presented, it can be shown that assays of As(III) in natural waters can be performed only if an on-line preconcentration step is incorporated into the FI manifold. The method offers a unique illustration of the versatility of the FI approach in sample manipulation to reach low detection limits as well as a demonstration of cleverly exploiting kinetic discrimination schemes for promoting the main reaction and eliminating potential side reactions and interferences.

The analytical performance of an on-line focused microwave digestion method for the determination of selenium in enriched yeast by hydride generation–atomic absorption spectrometry (HG–AAS) is evaluated. The method is based on the direct injection of slurried samples into a carrier stream of water. Microwave-assisted digestion in the presence of 1% K2S2O8 permits the quantitative extraction of selenium species from the solid samples and their transformation with HCl acid to selenite. Based on a sample injection volume of 100 μl, limits of detection (L.D.) of about 1 ng Se and relative standard deviation (R.S.D.) of 4.5% were obtained. The results of comparison with traditional digestion procedures are reported.

Permanganate Time (PT) is a standard quality control test for oxidisable impurities in products such as ethanol, acetic acid, acetic anhydride, acetone, methanol, pyridine, propionic acid and tricresyl phosphate. The test relies on visual assessment of the change in pink colouration over a period of time of a sample of product to which has been added a standard solution of potassium permanganate. Typically, this time period is between 30 min and 2 h. The standard test, therefore, is very time-consuming as well as being highly susceptible to operator error and bias. An automated measurement system has been developed that employs a photodiode array spectrophotometer to monitor the colour of the mixture. Pre-processing of the spectrum eliminates any interference and provides accurate estimates of the PT, even in the presence of a heavy precipitate. The method by which the software determines the test result can be tailored by the user to suit particular products. Any number of such user configurations can be saved for future use.
