Objective To investigate the clinical efficacy and safety of soluble hyaluronic acid microneedle eye patch on infraorbital wrinkles. Methods Seventy-five female subjects in Beijing and Shanghai were recruited and treated with soluble hyaluronic acid microneedle patch two days a week at the left lower eyelid (experimental side) and at least 30 minutes each time for 12 weeks. Investigator visual assessment, instrumentation measurement for skin moisture, elasticity, wrinkle area and depth were performed at week 0 (pre-treatment) and week 4 and week 12 and compared to the right (control side). The safety was assessed by documenting adverse reactions occurring during the study. Subject satisfaction was assessed at week 12. Results After 12 weeks of treatment, the visual assessment of skin wrinkles on experimental side (n=41, 54.67%) was significantly improved as comparied with that on control side (n=14, 18.67%). The skin hydration of treatment side skin increased with treatment time (t=-0.488 at week4, t=-3.054 at week 12, P<0.05). After 12 weeks of treatment, skin elasticity of the experimental side improved significantly. The improvement rate of the experimental side was more than that of the control side (t=2.724, P<0.05). The wrinkle area was significantly decreased in 4 and 12 weeks compared with pre-treatment (t=4.442 at week 4 and t=3.802 at week 12, P<0.001). The depth of wrinkles of the experimental side shallowed with the treatment (t=-2.435 at week 4 and t=-3.433 at week 12, P<0.05). Only 8% (n=6) of the subjects had temporary slight pruritus or acupuncture sensation on the treatment side, and no other adverse reactions occurred. The overall subject satisfaction rate was 88%(n=66). Conclusions Soluble hyaluronic acid microneedle eye patch is effective in improving wrinkles, skin elasticity and hydrating, and has no obvious adverse reaction. Objective To indentify the cognitive status of Chinese patients to acne and the influencing factors to theirs' cognitive status, so as to provide solid evidences for the prevention and treatment of acne. Methods A self-designed questionnaire was made to conduct this survey of 16, 156 acne patients, who seeked to the treatment in the dermatological departments from 112 hospitals in China. The survey consisted of several parts, including the general status of patients, the patients' cognition of occurrence, development and risk factors of acne, whether the first choice was seeking treatment at the hospital when the patients had acne and the condition of selection of skin care products. The factors were analyzed, which could impact the cognition of the patients' behavior of treatment, how did the patients' cognition to influence their medical behavior and skin care as well as the consistency of assessment of the severity of acne by doctors and patients themselves. Results The acne patients studied had the best knowledge of " acne is a skin disease" , " it not only occurs in the period of adolescence" and " the disease can be prevented and cured" , which accordingly accounted for 80.65%, 69.16% and 65.49% of the total patients respectively. However, the awareness of acne patients to heredity, high sugar and dairy products as risk factors for acne was insufficient, which accounted for 48.72%, 42.40% and 18.25% of the total patients, respectively. Gender, age, educational level, occupation and health knowledge were the main factors affecting the cognitive level of patients; the survey also found that men, patient with educational level of junior high or even lower educational condition, occupation of labor workers or farmers and patients were lack of health education with poor knowledge of the genetics and dietary were risk factors for acne; patients with age over 36 years or with mild illness had poor knowledge of dietary risk factors for acne; the difference was statistically significant (P<0.05). The analysis of the influence of cognitive status on medical treatment behavior and skin care showed that the better the cognition, the higher the probability of patients would choose medical treatment as the first choice as well as choosing functional skin care products; the difference was statistically significant (P<0.05). The consistency of assessment of the severity of acne by doctors and patients was poor (Kappa value <0.4), and the assessment of severity of acne by patients was more serious than doctors' assessment. Conclusions Patient's cognitive status will affect their medical behavior and skin care, and there is also a phenomenon that patients have a more serious assessment of their acne condition. It is suggested that health education for acne patients should be strengthened in clinical medicine so as to improve their knowledge of acne as well as preventing from acne effectively. Objective To observe the fat particles survival situations in autologous transplantation under different conditions of low temperature. Methods Three sample groups of adult Wistar rats were injected with fresh autologous fat particles, which were stored under 80 ℃ for two months and under 196 ℃ for two months respectively, and the volume, parallel histological detection, glucose transport measurement, MTT experiments and other data were compared and analyzed one month and two months after the transplantation. Results One month after transplantation, graft survival rate was higher in Group injected fresh autologous fat particles than that of Group injected autologous fat particles stored under 80 ℃ and Group injected autologous fat particles stored under 196 ℃ (P<0.05); Two months after transplantation, the differences among three groups had no statistical significance. Before the transplantation, the activity of autologous fat particles stored in 80 ℃ and 196 ℃ were 68.7% and 62.5% of fresh autologous fat particles, respectively, two months after cryopreservation. One month after the transplantation, we found that the activities of autologous fat particles were higher in fresh group than that of 196 ℃ and 80 ℃, There was no significant difference between the groups stored in 196 ℃ and 80 ℃. Two months after the transplantation, the activities of autologous fat particles were higher in fresh group than that of 196 ℃ and 80 ℃. Before transplantation, the particles of fat histology showed no significant difference among three groups. One month after transplantation, fat vacuoles and distinct inflammatory infiltration were observed in all three groups, with the group stored in 80 ℃ as the most serious affected. Two months after transplantation, fresh fat particles and Group stored in 196 ℃ showed large quantity of tightly packed intact fat cells, scattered around the central area in fat vacuoles, while Group stored in 80 ℃ still had a large quantity of fat vacuoles, but the fat vacuoles were obviously fused. Conclusions The fat stored at80 ℃ and196 ℃ could meet the needs of clinical use in the short time. The fat stored at 196 ℃ shows the best activity. The fat particles stored at196 ℃ could be used to establish better blood circulation. The heaviest inflammatory reaction occurs in the graft zone one month after fat transplantation and the decrease of transplanted particle activity is the most obvious. Objective To observe the effect of varying intensities of water-jet force on autologous fat graft viability. Methods Lipoaspirate was taken from 12 female patients undergoing water-jet assisted abdominal liposuction at our department. According to the intensity of water-jet force, the experimental group was divided into four subgroups: R1 (pressure, 30 bar), R2 (pressure, 50 bar), R3 (pressure, 70 bar) and R4 (pressure, 90 bar). Hand-held suction was taken as the control group C. Adipose tissue was filtered with cotton cushion and centrifuged at low speed, and the composition ratio of water and fat tissue from each group was observed. Calcein-AM/Hoechst 33342 staining was used to detect the viability of adipocytes. Results Fat aspirates was divided into four layers: oil layer, pure fat tissue, liquid and bottom sediment. Oil ratios of R1, R2, R3, R4 and C were (8.9±2.3)%, (9.6±2.1)%, (10.3±1.3)%, (14.2±1.6)% and (9.5±1.8)%, respectively. There was no statistically significant difference between R1, R2, R3 and C (P>0. 05). Statistically significant difference was found between R4 and other groups (P 0.05). Statistically significant difference was found between R3, R4 and C (P<0.05). Conclusions Viability of fat graft harvested under lower intensity of water-jet force (R1, R2) is higher than that harvested under higher intensity of water-jet force (R3, R4).
