In recent year a couple of interesting immunological functions of keratinocytes have been discovered. With dopa as substrat the enzyme were incubated 20 min in 0.1 M phosphate buffer, pH 6.8. After that the activity was detected at wavelengths of 313 and 492 nm using a layer thickness of 10 mm. Employing this conditions higher activities were estimated in the commercial preparates as indicated. The high sensitivity commends this standardized method to use in melanoma research.

In the last years a new therapy of psoriasis was developed, which consists in a treatment with salt solutions, resembling the water of the Dead Sea, and ultraviolet light (Tomesa-therapy). We studied the influence of the used salt on ATPase positive epidermal Langerhans cells in murine ear skin. An irreversible partial reduction of the Langerhans cell ATPase was found after salt treatment of separated epidermis or of full skin preparations. These results may have implications for the optimization and broader application of this therapy.

Using a multilayer membrane system and human horny layer the difference in the penetration of salicylic acid (SA) and its sodium (Na-S) and choline (Ch-S) salts from topical formulations was studied. It was found Na-S and Ch-S were markedly accumulated in the first membrane of the three layer membrane system used. In contrast, a rapid penetration into all three membranes was observed when SA was used. Similar penetration profiles were obtained in human horny layer. Hence, the use of the salts of SA appears to be more suitable for the application as keratolytic.

15 cases of human malignant melanoma were studied and classified into 5 superficial speading (SSM), 5 nodular melanomas (NM), and 5 melanoma metastasis (Met). The tissue was fixed with formaldehyde and cetylpyridium chloride (CPC). Glycosaminoglycans (GAG) or proteoglycans respectively were characterized by Alcian blue staining following the method of critical electrolyte concentration (CEC) (Scott, Dorling 1965) and by testes hyaluronidase. The staining intensities were quantified by a Leitz MPV photometer microscope in basement membranes (BM) and tumor septa. Tumor septa, which may be looked on as correlates of epithelial BM material, show increased straining intensities as compared to the normal BM (nBM) around the tumor. It is concluded from the sensitivity to testes hyaluronidase and the straining pattern that these are caused by increased straining of GAG of the type of chondroitin sulfates and possibly of dermatan sulfate while unsulfated GAG are rather decreased. The GAG pattern in BM in SSM shows characteristics of tumor septa and of nBM as well. The staining of the tumors shows higher intensities than that of all structures in the normal skin. It is concluded that increasing malignancy is accompanied by increasing changes in GAG which can be quantified by the method used topohistochemically discerning the healthy tissue from malignant structures.

Zinc is able to form complexes with 8-hydroxyquinoline (HC). Therefore the penetration of HC from topical formulations into the ear of guinea-pigs and into a multilayer membrane system was decreased by Zi. Using the AUC a suitable in vitro-in vivo correlation was obtained. An exception was observed when there were interactions between the ointment base and the in vitro model system. Furthermore, it was found that also zincoxinate is able to penetrate into the skin of guinea-pigs.

Microcirculation in the hands of patients with progressive systemic sclerosis shows some peculiarities. For diagnostic purposes functional thermography and the acrale rewarming test may be used. Typical features of progressive systemic sclerosis are a general hypothermia of the hands and localized hypothermic areas and refractory hyperthermic areas. In the acrale rewarming test reactive vasodilatation after cold stimuli is often not inducible and the significantly retarded rewarming shows a marked local restrict dependence. The significance of these facts for a more intensive physical therapy is discussed.

This open clinical study was designed to evaluate and compare the efficacy and side-effects of lindane (1% and 0.3%), benzyl benzoate (20% and 10%) and permethrin (5% and 2.5%) after twice-, three times- and once application at bedtime, respectively, in the treatment of scabies in 104 adults and 70 children aged between 0 and 5 years. Treatment failures were registered afer lindane in 3 adults and 2 children, while benzyl benzoate and permethrin cured all patients as assessed after 3 weeks follow-up. The number of irritations and postscabious eccematous reactions was increased after benzyl benzoate. Permethrin revealed to be very reliable and exhibited only a few side-effects when applied once at bedtime. Because of the risk of the percutaneous absorption and neurotoxicity of lindane, the application of permethrin can be recommended as an useful alternative in premature infants and small children, patients with seizures and neurological complications, in cases of treatment failures of lindane with the need to repeat the therapy, in scabies crustosa as well as in children, pregnant woman and nursing mothers.
