Antioxidant and Anticancer Activities of Methanol Extract of Seeds of Datura stramonium l 曼陀罗种子甲醇提取物的抗氧化和抗癌活性

Background: Datura stramonium belongs to the family Solanaceae which is commonly dubbed as thorn apple or jimson weed, and is abundant throughout tropical and subtropical regions. Datura seeds and leaves are used as anti-asthmatic, antispasmodic, hypnotic and narcotic. Externally, the jimson seeds are used in treating fistulas and abscesses. It has been used by the British soldiers to treat respiratory problems. Datura has also finds its utility in Ayurveda for asthma symptoms where jimson weed’s leaves are smoked in cigarette or pipe. Objectives: Aim of the present study was to investigate the antioxidant and antiproliferative activities of methanol extract of seeds of D. stramonium. Materials and Methods: Antioxidant assays such as DPPH ̇ radical, superoxide radical, ABTS ̇+ radical cation, OH ̇ radical scavenging assays, Phoshomolybdenum reduction and Fe3+ reducing power assays were investigated. Thin layer chromatography was performed to find out compounds present in the methanol extract. in vitro cytotoxic activity for MCF7 (breast cancer) cell line was too studied by MTT reagent assay method. Results: IC50 values of DPPH ̇ radical, superoxide radical, ABTS ̇+ radical cation, OH ̇ radical scavenging assays were 35.26, 10.50, 49.36 μg/mL concentration respectively. TLC analysis showed the presence of compounds with Rf values of 0.76 and 0.58 in the methanol extract of D. stramonium. Cytotoxic activity for MCF7 cell line was 66.84% at 500 μg/mL concentration of methanol extract. Conclusion: Here Current study corroborates that D. stramonium is enriched with fine source of significant natural oxidants with anticancer agents and has therapeutic potential for treatment of diseases, induced by oxidative stress, Further research work is required to isolate active compounds for clinical

Phytochemical Evaluation and In vitro Antioxidant Activity of Various Solvent Extracts of Leucas aspera (Willd.) Link Leaves 不同溶剂提取物的植物化学评价及体外抗氧化活性研究链接的叶子

Background: Free radicals initiate the oxidative stress and damage the healthy cells. These damages contribute ageing, cancer, cardiovascular and inflammatory diseases. Phenolic compounds, flavonoids and tannins are directly contributed to antioxidant activity. Objective: The present study was attempted to evaluate the phytochemicals present in various solvent extracts obtained from Leucas aspera (Willd) Link leaves ( L. aspera ) and its antioxidant activity using different in-vitro models. Materials and Meth-ods: The free radical scavenging and antioxidant activity of petroleum ether, ethanol, isopropyl alcohol, ethyl acetate and chloroform extracts of L. aspera leaves were assessed by different in-vitro models include DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging capacity, total antioxidant activity, ABTS (2,2,-azinobis (3-ethylbenzoline-6-sulfonic acid) radical scavenging activity and nitric oxide radical inhibition assay, in order to ensure the pharmacological effects of the plant. Results: Petroleum ether extract (non-polar solvent) showed better antioxidant activity among the solvents with IC 50 values 18.96 μg/mL (DPPH assay), 17.22 μg/mL (total antioxidant assay), 16.00 μg/mL (ABTS assay) and 11.87 μg/mL (nitric oxide scavenging). Whereas, ethanol extract (polar solvent) exhibited better DPPH scavenging, ABTS assay and nitric oxide scavenging activity (IC 50 =19.90, 11.60 and 13.47 μg/mL respectively) compared to chloroform, ethyl-acetate and isopropyl alcohol extracts. Antioxidant Assay of Gold and Silver Nanoparticles from Edible Basidiomycetes Mushroom Fungi 食用担子菌、蘑菇真菌中金、银纳米颗粒的抗氧化测定

Background: In recent years, nanotechnology research is emerging as cutting edge technology interdisciplinary with physics, chemistry, biology, material science and medicine. AuNPs and AgNPs are an important class of nanomaterial for a wide range of biomedical applications. Synthesis of AuNPs and AgNPs through biological route is preferred due to its environment-friendly and economic aspects. Mushrooms have been known for their nutritional values and used as medicines by humans for ages. In modern terms, they can be considered as functional foods which can provide health benefits beyond the traditional nutrients. The present study demonstrates an eco-friendly and low-cost method of biosynthesis of AuNPs and AgNPs using basidiomycetes mushroom fungal strains. Materials and Methods: The antioxidant potential of the basidiomycetes mushroom fungal strains was analysed by total flavonoid content, FRAP assay, ABTS assay, Metal chelating activity, Phosphomolybdenum assay, Assay of superoxide radical scavenging activity, Free radical scavenging activity on DPPH along with the determination of total phenolic and tannin contents in the mushroom mycelial extracts. Results: The synthesized gold nanoparticles (AuNPs) silver nanoparticles (AgNPs) were confirmed by the colour transformation and Ultraviolet-visible (UV-visible) spectroscopy. These biologically synthesized AuNPs and AgNPs were tested for antioxidant activities. The biosynthesized AuNPs and AgNPs showed significantly higher antioxidant activity. Conclusion: The present study explored that the mushrooms which are efficient producers of AuNPs and AgNPs, and could act as safe and cost-effective with potential antioxidant activities. These findings encourage studying these fungal strains further for their potential biological applications.

Effects of extraction conditions over the phlorotannin content and antioxidant activity of extract from brown algae Sargassum serratum (Nguyen Huu Dai 2004) 提取条件对褐藻褐藻提取物中褐皮素含量及抗氧化活性的影响(Nguyen Huu Dai 2004)

Introduction: This study focused on the discussion of effect of various extraction conditions for phlorotannin content and antioxidant activities extracted from brown algae Sargassum serratum. The algae was grown in the tropical coastal areas of Vietnam. Methods: The various extraction conditions include the following parameters: temperature (30-80 0 C), maceration time (1/2–2,5 hours; 8-48 hrs), the ratio of solvent to material (10:1–70:1 (v/w)), pH (2–8), various kind of solvents (ethanol, acetone, chloroform, ethyl acetate and n-hexan) and solvent concentrations. Phlorotannin content, antioxidant activities, and some phytochemical compositions of Sargassum serratum were evaluated. Results: The highest phlorotannin content and antioxidant activities was expressed when extracting in the following conditions: 100% ethanol solvent at 50 0 C in 32 hours with the ratio of solvent to material of 40/1 (v/w), pH 7 and one-step extraction. The extract contained flavonoid, terpenoid, alkaloid, fatty and oil. DPPH free radical scavenging of the extract was 87.53%. Conclusion: Phlorotannin content and antioxidant activities extracted from Sargassum serratum was depended on the extracting conditions. The condition of extraction for antioxidant phlorotannin and the chemical composition of extract was determined. Brown algae Sargassum serratum has high antioxidant phlorotannin content. A total of 6 compounds were identified from the extract of Sargassum serratum.

Objectives: The present study assessed the phytochemical components, In vitro antioxidant ability and cytotoxicity of leaf extract of Ficus racemosa . Methods: Preliminary phytochemical analysis in aqueous and ethanol was carried out for the presence of phytochemical components. Of the two extracts used ethanolic extract possessed the highest phytochemical constituents compared to aqueous extract. Hence the antioxidant activity of ethanolic extract of F. racemosa was performed by several antioxidant assays including 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging assay, nitric oxide (NO) scavenging assay, reducing power and superoxide radical (SO) scavenging assay. Results: From the results, F. racemosa has been found to have the significant antioxidant activity in a dose-dependent manner and IC 50 value was 150 μg/ml for DPPH and 100 μg/ml for both NO and SO scavenging assays. Further, the cytotoxicity analysis was determined against Dalton Lymphoma Ascites (DLA) cell line and the IC 50 value was found to be 175 μg/ml for ethanolic leaf extract of F. racemosa . Conclusion: Hence, the current study attests that F. racemosa is enriched in phytochemicals and a fine source of natural antioxidants with anticancer agents and can be used in pharmaceutical preparations for the treatment of diseases induced by oxidative stress.

Introduction: A number of plants of Kashmir valley are unexplored and Eremurus himalaicus is one amongst them. Eremurus himalaicus is an edible herb of North Western Himalayas which is yet to be evaluated for possession of antioxidant and antimicrobial activities. Method: The antioxidant potential of the plant extracts (Ethyl acetate, EHE; Methanol, EHM and Aqueous, EHA) was assessed by determining the total phenolic content, total flavonoid content, DPPH (1, 1-diphenyl-2-picrylhydrazyl) radical scavenging activity, ferrous metal ion chelating activity, hydrogen peroxide scavenging assay and total reduction capability assay. The plant was further investigated for antimicrobial activity using agar well diffusion method. Results: EHM showed highest total phenolic content followed by EHA and EHE with 270 mg GAE/g, 240 mg GAE/g and 110 mg GAE/g respectively. The flavonoid content was found to be highest in EHA with 85 µg QE/g followed by EHM with 65 µg QE/g and least in ethyl acetate extract with only 20 µg QE/g. Ferrous metal ion chelating potential was also highest for EHA with IC 50 value of 200.336. For DPPH assay and H2O2 scavenging assay the activity was highest for EHM with IC50 value of 148.1788 and 182.3371 respectively. In total reduction capability assay again EHM showed highest reducing power. Besides, antibacterial activity was also screened on some human pathogenic bacterial strains where again the highest activity was shown by EHM. Conclusion: In conclusion, EHM and EHA showed highest antioxidant potential and EHE showed the least. Metabolite Fingerprinting and Antioxidant Potential of Tartary Buckwheat- an Underutilized Pseudocereal crop from Kashmir Region 克什米尔地区未充分利用的假谷类作物苦荞代谢物指纹图谱及其抗氧化潜力

Objective: Buckwheat is an underutilized pseudocereal crop used as a staple food especially in Himalayan regions. The aim of the present study was to evaluate the phytochemical screening, antioxidant potential and metabolite profiling of tartary buckwheat extract. Methods: Tartary buckwheat leaf samples were tested for total phenols, flavonoids and in vitro antioxidant potential in terms of total antioxidant activity, free radical scavenging (superoxide, hydrogen peroxide) and DPPH assay. GC-MS profiling was done to identify and quantify the various metabolites from the methanolic leaf and groat extract. Results: Preliminary phytochemical screening of methanolic extract revealed the presence of alkaloids, flavonoids, phenols, tannins, saponins, phlobatannins, coumarins, glycosides and anthoquinones. Methanolic extract exhibits higher TPC (28.32 ± 5.31 mg gallic acid equiva- lent g -1 DW) and TFC (25.18 ± 3.5 mg rutin equivalent g -1 DW). DPPH radical (EC50=1.8 µg ml -1 ) and H2O2 scavenging (EC50=0.103 µg ml -1 ) potential of tartary buckwheat leaf extract shows promising results. From GC-MS metabolite fingerprinting, over 111 and 24 metabolites were identified among leaf and groat extract respectively. The major compounds present in the extracts were fatty acids, hydrocarbons, steroids, terpenoids, esters, organic acids and aldehydes with excellent pharmaceutical properties. Conclusion: The tartary buckwheat extract were found to contain numerous metabolites with potent antioxidant and other pharmacological actions. Thus, tartary buckwheat could be a promising alternative in functional food sector to improve social well-being and neutraceutical to diminish malnutrition especially for the impoverished community.

Antioxidant, Antibacterial Activities and Identification of Bioactive Compounds from Terminalia chebula Bark Extracts chebula树皮提取物的抗氧化、抗菌活性及活性物质的鉴定

Background: Free radical induces numerous diseases by damaging biomolecules such as lipids, proteins, RNA and DNA. Several scientists reported that numerous plants and plant extracts have potent antioxidant activities to scavenge free radicals. The present study was aimed to screen phytoconstituents, in vitro antioxidant and antibacterial potential in various solvent extracts of Terminalia chebula bark. Methods: Phytochemical analysis, estimation of various metabolites, in vitro antioxidant and antibacterial activity were done by adopting standard protocols. Selected bioactive (acetone) extract of T. chebula was analyzed for their phytochemical profile by GC-MS analysis. Results: The results of preliminary phytochemical screening analysis revealed that presence of various phytochemicals like, alkaloids, flavonoids, glycosides, terpenoids, phenolics, saponins and carbohydrates in most of the tested extracts. Acetone extract possess significant high quantity of both primary and secondary metabolites when compared with other extracts. Remarkable free radicals scavenging potential was observed in acetone extract with lowest IC50 values on all tested radicals namely, DPPH. (IC50=144.77 μg/ml), NO . (IC50=149.46 μg/ml), .OH (IC50=121.18 μg/ml), O2 .(IC50=159.41 μg/ml), Reducing power (IC50=35.85 μg/ml), Fe 2+ ion chelating (IC50=137.56 μg/ml) and TBARS (IC50=201.96 μg/ml). Acetone extract expressed significant high antibacterial activity against S. typhi (15 mm). The result of GC-MS analysis of acetone extract shows the presence of 32 major bioactive compounds, including various phenolic, sesquiterpene, flavonoid, triazine and gibberellin compounds. Conclusion: The present study suggested that T. chebula bark extract serves as a good source of phytochemicals, natural antioxidant and antibacterial agent.

Background: Plant’s are ability to manage with abiotic stress factors are mainly altered the metabolic pathways and produces a varied range of secondary metabolites. Secondary metabolite manufacture is a serious part of the defense response in stress circumstances. Objectives: The study was aim to find out the impact of drought stress factor on in vitro antioxidant potential of chloroform insoluble fraction (phenolics) of leaves and flowers extract of Tridax procumbens. Materials and Methods: The matured leaves and flowers of Tridax were collected widely from the four different locations;- Hilly dry-terrain; Hilly wet-terrain; Low dry–land; Low wet-land. The collected leaves and flowers of Tridax were washed with distilled water, shade dried, coarsely powdered and extracted using 70% ethanol and fractionated by chloroform to obtained chloroform insoluble fractions. The chloroform insoluble fraction then subjected into various in vitro free radical scavenging activities. Results: The drought stress significantly increased the sucrose content in the leaves, chloroform insoluble fractions (water soluble), total phenolics and decreased relative leaf water contents and chloroform soluble fractions when compared with well water state of Tridax. The in vitro free radical scavenging activities results revealed that drought had constructive effects on radical scavenging and antioxidant capacity. Conclusion: In dry land of the Tridax procumbens (both hilly dry-terrain and low dry-land) induced drought stress-related metabolic alterations and free radical production. In this condition Tridax need oversupply of reduced phenolics for tolerance and prevent the oxidative damage of the cells. As a result the metabolic processes are shifted towards biosynthesis of highly reduced phenolics in the Tridax for better adaptation.

Chemical- and Cell-based Antioxidant Capacity of Methanolic Extracts of Three Commonly Edible Plants from Zingiberaceae Family 姜科三种常见食用植物甲醇提取物的化学和细胞抗氧化能力

Background: Edible plants belonging to the Zingiberaceae family display various antioxidative properties and widely used as folklore medicines. Three edible plants in Zingiberaceae family; namely Boesenbergia rotunda, Phaeomeria imperialis and Zingiber officinale were selected for secondary metabolites extraction using methanol. All crude extracts were investigated to evaluate their phenolics and flavonoids contents and further compare their antioxidant properties. Methods: Total phenolic content and total flavonoid content were evaluated using Folin-Ciocalteu method and Aluminium Complexation Reaction respectively. Conventional DPPH (2,2-diphenyl-1-picryl-hydrazyl), revolutionary CUPRAC (cupric ion reducing antioxidant capacity) and cellular antioxidant activity (CAA) in vitro assays were employed to evaluate the antioxidant activities of methanolic plant extracts for the first time. Results: DPPH and CUPRAC antioxidant assays resulted in similar trend to total flavonoid content in the order Z. officinale >P. imperialis>B. rotunda where as P. imperialis revealed highest phenols content and displayed highest CAA. Conclusion: Methanolic extract of Z. officinale showed highest free radical scavenging ability and hence flavonoids present may potentially act as natural source of antioxidant.
