Midgut health of Apis mellifera workers on oral administration of Lactobacillus rhamnosus and organic acids 口服鼠李糖乳杆菌和有机酸对蜜蜂工蜂中肠健康的影响

AbstractA study about the provision of Lactobacillus rhamnosus NR_113332) and organic acids was carried out. 108 colony-forming units of probiotics were provided to the caged workers with and without the addition of 2.99% lactic acid, 2.91% acetic acid, and 1.96% acetic acid. After two weeks of oral administration, on termination, five bees were collected and dissected from each cage, and their midguts were cut into 8 μm thick sections by microtome. Subsequently, these histologically processed preparations were observed under camera fitted microscope. It was observed that the experimental workers were provided with diets: (pollen+ 50% w/v sucrose in 1.96% acetic acid, pollen+ L. rhamnosus in 50% w/v sucrose in distilled water, pollen+ L. rhamnosus in 50% w/v sucrose in 2.99% lactic acid, pollen+ L. rhamnosus in 50% w/v sucrose in 2.91% acetic acid, pollen+ L. rhamnosus in 50% w/v sucrose in 1.96% acetic acid) had a considerable quantity of peritrophic membranes. The mean midgut lumen diameters (µm2) were 373.33 ± 98.38, 296.67 ± 23.33, 243.33 ± 71.72, 426.67 ± 40.55, 576.67 ± 93.87 as compared to control’s 240.00 ± 30.55. The provision of probiotic and organic acids resulted in a slightly disturbed midgut epithelial wall, the presence of more quantity of peritrophic membranes, saved regeneration crypts, more gastric cells discharging, and some cytoplasmic vacuolization of caged worker bees. Beekeeping under climate change 气候变化下的养蜂业

There is consensus that climate change is one of the grand challenges facing humanity in the twenty first century, inevitably the most confided and undeniably one of the most pressing. Profound effects are inevitable in global agriculture, and beekeeping is certainly no exemption. Indeed, extreme weather and natural disasters have already had an impact on honey bees. Thus, it appears evident that climate change will constitute a key stress factor for managed bees and beekeepers alike contributing to increased colony losses and reduced income. Here, we review the literature on the impact of climate change on honey bees and beekeeping. Based on the literature, it is evident that at present there is no inclusive strategy for beekeeping to adequately deal with the challenges global climate change will bring. Here, we call for such a strategy and briefly list the main challenges for future beekeeping due to a changing climate as well as suggest possible countermeasures. Ultimately, the impact of climate change and its mitigation are at present insufficiently understood in the beekeeping context. This calls for respective concerted efforts of scientists, beekeepers, and other stakeholders to find a sustainable future for beekeeping. Such efforts will inevitably require evidence-based mitigation measures to deal with the increasing global impact of climate change.

Increased levels of introgression evident in Irish honey bees 在爱尔兰蜜蜂中明显增加的基因渗入水平

Ireland is home to a significant population of the black bee, Apis mellifera mellifera, however, in recent years the numbers of imported queens have increased 10 fold and beekeepers are observing more bees that are not totally black. Here we confirm an increase in the proportion of introgressed honey bees in Ireland via multiple approaches. The use of colour and wing morphometrics is becoming less reliable in Ireland to identify native honey bees due to the increased presence of various hybrid phenotypes and genotypes.

AbstractGenetic analysis of the honey bee spermathecal content can be particularly useful to provide an estimate of the genetic diversity and purity of the surrounding populations. Here we compared the concentration and quality of DNA extracted from queen spermatheca using four commercial kits to determine the best method to obtain DNA suitable for single nucleotide polymorphism genotyping by next-generation sequencing. The four kits were tested with different adjustments in the lysis incubation time, use of RNA-carrier, elution conditions and number of re-elutions. Honey bees (Apis mellifera) have a peculiar and complex reproductive biology, with queens being polyandrous and mating with several drones during one or more mating (nuptial) flights in so-called drone congregation areas. Observing the virgin queens’ and drones’ flight behaviour provides data to understand and interpret a portion of the honey bees’ complex reproductive process. Observing the behaviour of the virgin queens on the hive entrance also serves to estimate the distance from the mating place or potential drone congregation areas (DCAs) as well as to detect the presence of airborne drones in the area.
