The effect of mitochondrial ATP-dependent K(+)-channel (K+(ATP)-channel) opener diazoxide (DZ) on the oxygen consumption, functional state and the opening of cyclosporine-sensitive pore in the rat liver mitochondria has been studied. It has been established that K+(ATP)-channel activation results in the increase of the oxygen consumption rate (V4(s)) and the uncoupling due to the acceleration of K(+)-cycling, the decrease in state 3 respiration rate (V3) and the respiratory control ratio (RCR). Under K+(ATP)-channel activation an inhibition of oxidative phosphorylation takes place which reduces the rate of ATP synthesis and hydrolysis as well as ATP production and consequently results in the seeming increase of P/O ratio. It has been shown that the increase in ATP-dependent K(+)-uptake accompanied by the opening of mitochondrial permeability transition pore (MPTP) leads to dramatic uncoupling of the respiratory chain due to simultaneous activation of K(+)- and Ca(2+)-cycling supported by MPTP and Ca(2+)-uniporter as well as K(+)-channels and K+/H(+)-exchange. K+(ATP)-channel activation leads to the partial inhibition of MPTP, but insufficient for the restoration of mitochondrial functions. Elimination of Ca(2+)-cycling after MPTP opening is necessary to return mitochondrial functions back to the control level which shows that MPTP could serve as the mechanism of reversible modulation of bioenergetic effects of K+(ATP)-channel activation.

The effect of metal-nanocomposites (Me-NC) of cobalt and zinc (Co- and Zn-NC, correspondingly) synthecized on the basis of vinylpyrrolidone (PS) on the metal-accumulative proteins with antioxidant potential metallothioneins (MT) in crucian carp (Carassius auratus gibelio) was studied. Fish was subjected to the effect of Co-NC, Zn-NC, Co2+, Zn2+ or polymer carrier (PC) in the concentrations correspondent to 50 microg x Co/l or 100 microg x Zn/l during 14 days. It was shown that the MTs response is highly specific for the nature of metal, both in ion and Me-NC form: the effect of Co and Co-NC provoked the elevation of total MT concentration (MT-SH) and activation of antioxidant defence, whereas Zn and Zn-NC induced the decrease of the concentration of MT-SH and the inhibition of antioxidant defense. All the exposures provoked the decrease of the concentration of immunoreactive chelating MT form (MTi) and reduced glutathione, activation of anaerobiosys and Mn-superoxide dismutase, and also decrease of the concentration of proteins and lipids oxidative injury products. It was accompanied by the increase of the content of erythrocytes with nuclear abnormalities but did not cause the decrease of choline esterase activity. According to the rate of MT-SH and MTi concentrations, antioxidant potential of MTs is determined by its apoform. Our data indicate that partial biodegradation of Me-NC occurs in the organism of crucian carp.

Creation of new biocompatible nanomaterials, which can exhibit the specific biological effects, is an important complex problem that requires the use of last accomplishments of biotechnology. The effect of pristine water-soluble fullerene C60 on ATPase activity and superprecipitation reaction of rabbit skeletal muscle natural actomyosin has been revealed, namely an increase of actomyosin superprecipitation and Mg2+, Ca(2+)- and K(+)-ATPase activity by fullerene was investigated. We conclude that this finding offers a real possibility for the regulation of contraction-relaxation of skeletal muscle with fullerene C60.

It is known that extracts of cryopreserved organ fragments of pigs and piglets stimulate the processes of reparative regeneration. Therefore, the study of the extracts is essential for understanding the mechanism of their biological effects. In this paper was studied the molecular mass distribution of peptides in the extracts of cryopreserved heart fragments of pigs' and piglets', i.e., the chromatograms of pigs' heart extracts show 3 picks, whilst piglets' heart extract show 6 picks. The peculiarity of pigs' heart extracts is the absence of peptides with molecular mass of 10,000 and more. The differences in the intensity of extracts fluorescence prove that the peptides in pigs' heart extracts contain greater amount of tryptophan residues accessible for solvent. The synchronous fluorescence spectra of extracts were obtained which allows the identification of extract without assessment of their components. Results shown in this research could be used for control and standardization of heart extracts peptide composition under investigation of their biological quality.

The intensity of free-radical processes upon long-term omeprazole-induced hypoacidity in the rat pancreas was investigated. Significant violation of oxidative-antioxidative balance in pancreatic tissue upon gastric hypochlorhydria was established: overproduction of superoxide anion, quantitative changes of lipid functional groups, increased level of lipid peroxidation products, augmentation of xanthine oxidase, superoxide dismutase and glutathione transferase activity, as well as depletion of catalase, glutathione peroxidase activity and reduced glutathione content. The inflected expression of Cckbr gene in the rat pancreas upon these conditions was also observed, thus suggesting an increased risk of pathological changes development in the gland. Abovementioned parameters were only partially restored to control values in the case of simultaneous administration of multiprobiotic "Symbiter" with omeprazole, thus indicating the ability of this preparation to efficiently counteract the development of oxidative damages in pancreatic tissues upon long-term hypoacidic conditions.

The effects of urocanic acid (UA) on thermodynamic parameters of model dipalmitoylphosphatidylcholine (DPPC) lipid membrane have been studied by means of differential scanning calorimetry (DSC). The observed ordering effect of UA on the lipid bilayer is reflected in the increase in both the main phase transition temperature and cooperative unit size of the lipid membrane. Analysis of FTIR spectra suggests localization of UA molecules in the vicinity of the polar heads and carbonyl groups of DPPC due to electrostatic interactions and H-bonds. On the basis of experimental data obtained and geometry parameters of UA and DPPC molecules, some variants of the UA localization in DPPC bilayer were discussed.

The purpose of this study was to examine activity of adenylate cyclase (AC) and its cAMP-dependent phosphorylation by cAMP-dependent protein kinase (PKA) in the membrane of rabbit myometrium. Isoproterenol (IP) significantly increased AC activity of nonpregnant rabbits myometrium plasma membranes. However, during pregnancy AC of myometrium plasma membranes did not respond to IP. Phosphorylation of the myometrium membrane by PKA was followed by significantly decreased response of AC to IP.

A single injection of hydrocortisone to adrenalectomized animals increases the content of amino acids in muscles and lowers their total content in the liver; a single injection of ACTH lowers the total content of amino acids in the liver and blood plasma "Substitutive therapy" by hydrocortisone essentially increases the content of amino acids in the liver, muscles and blood plasma. Under conditions of the experiment the least changes in the content of amino acids were found in the brain tissue.

Isoenzymic spectra of lactate dehydrogenase and malate dehydrogenase were detected in unfertilized eggs of Misgurnus fossilis. They do not change significantly in the embryos during blastula, gastrula and early organogenesis stages of development. The intraspecific polymorphism of one lactate dehydrogenase subunit was found in the unfertilized eggs and embryos. The polymorphism is pronounced in the existence of three types of isoenzymic spectra of different frequencies in the given population of the loach.
