The behaviour of the in vitro-activities of an alkaline and an acid deoxyribonuclease (DNase I and II, resp.), and of an inhibitor of DNase I of the kidney of mice, as well as of the DNA- and protein-content of kidneys and thymus, was studied in about 500 mice 4 hours to 21 days after exposure to folic acid in doses of 60 — 180 mg/kg body-weight. The most important results can be summarized af follows: 1. Activity of DNase I decreased and activities of DNase II and of a DNase I-inhibitor increased under the influence of high doses of folic acid. Significant effects were observed 16 — 24 hours after folic acid-injections. Extreme values (80% decrease [DNase I], 180% increase [DNase II, DNase I-inhibitor]) were reached after 2 and 4 days and were dose-dependent. Control values reappeared within 1 — 3 weeks. 2. Protein- and DNA-content of the thymus behaved very similarly to DNase I-activity of the kidney. 3. The increase of the DNase II-activity of the kidney under the influence of folic acid resulted from enzyme induction. As to the behaviour of DNase I loss of enzyme out of damaged cells and the induction of a DNase I-inhibitor in the kidney must be taken into account. 4. In many systems DNase I may control DNA-synthesis. Preliminary studies on the behaviour of folic acid-induced reaction of the kidney, when inhibited by X-irradiation, Actinomycin D, Actidione, or poly (vinylsulfate), suggest that DNase I-inhibitor plays a certain role in combining protein- and DNA-synthesis by inhibiting DNase I.

Putrescine and spermidine were the dominant polyamines in Tetrahymena cells. Spermine was present only in very small amounts. Changes in the amount of putrescine during and after the synchronizing heat treatment followed a definite quantitative pattern. Changes in the amount of spermidine were similar, though not so pronounced. The changes may be due to a varied influx of polyamines into the cells. Moreover, the exchange of polyamines between the cells and the culture medium must be specifically regulated since there seems to be a relatively greater accumulation of spermidine than of putrescine in the cells, and since spermine was almost excluded from the cells.

A furocoumarin intercalated between two base pairs of native DNA can photoreact with two pyrimidine bases engaging both its 4',5'- and its 3,4-double bond. This fact was evidenced studying the behaviour of the fluorescence acquired by DNA during irradiation at 365 nm. When this double reaction occurs, a cross-linking is formed between the two strands of DNA, as denaturation and renaturation experiments have demonstrated. The various furocoumarin derivatives have a very diverse ability to form cross-linkages, which is independent of their photobinding capacity. This different ability is due mainly to the structure of furocoumarins and to the steric relationships that the intercalated furocoumarin molecules have with the stacked pyrimidine bases.

Zur Messung wurde das zentrifugierte Zellsediment in folgender Pufferlösung suspendiert: 30,0 MM Na 2 PH0 4 + 35,4 MM NaH 2 P0 4 , pH 6 , 7 6 , 8 , K®-Untergrund < 2 1 0 " 3 MM (flammenphotometrisch), KCl-Zugabe je nach Versuchsbedingung. Der Bakterienstamm ist auxotroph für His, Leu und Met; bezüglich Nährmedium und Züchtung siehe 12. Die Bakterien befanden sich bei Entnahme aus dem Nährmedium ca. 1 h in der stationären Wachstumsphase, pH 6,4, Extinktion (Amax = 4 1 8 n m ) der zehnfach verdünnten Suspension 0,30, Zellzahl ca. 2 -10 9 /ml (zur Messung beibehalten), Volumen der Proben 50 ml, Trockengewicht von 6 gleich vorbehandelten Proben 41,8 ± 1 , 2 mg, 12 h bei 80 °C im Vakuum getrocknet. Beim Versuch zu Abb. 1 verstrichen 6 h mehr zwischen Inkubation und Messung als bei der Versuchsserie zu Abb. 2. Temperatur der Versuche 25 — 26 °C.
