Anticonvulsant Luminal Affects the Arginine-Vasopressin Expression in Hypothalamus and the Locomotor Behaviour of Male Mice 抗惊厥药Luminal对雄性小鼠下丘脑精氨酸-抗利尿激素表达及运动行为的影响

Although it is reported that Luminal may affect the neuronal activity in the cerebral cortex and hippocampus, its impact on the hypothalamic nuclei including the paraventricular nucleus (PVN) and the supraoptic nucleus (SON) has not been elucidated. The PVN and SON are particularly important due to the release of arginine-vasopressin (AVP) which plays a crucial role in regulating cardiovascular functions, metabolism and locomotor behaviour, by their magnocellular neurons. In this study, we investigated the effect of chronic administration of Luminal (for 6 months) on the PVN and SON of male mice. We evaluated the expression of AVP by immunofluorescence and the changes in the cellular architecture by cresyl violet staining of PVN and SON. We also assessed the impact of Luminal administration on locomotor activity, which is largely influenced by AVP. Our findings indicated that chronic administration of Luminal decreased the expression of AVP in PVN and SON without significant changes in their neuronal architecture and influenced locomotor behaviour. Our findings provide novel insights into the central effect of anticonvulsant treatments on the AVP-producing neurons in the hypothalamus and could explain possible side effects on body physiology and behaviour. This may help optimize the therapeutic strategies used for seizure control.

The Effect of Ceftriaxon and Gentamicin on Antibiotic Resistance Pattern of Multiple Drug Resistance Klebsiella pneumoniae 头孢曲松与庆大霉素对多重耐药肺炎克雷伯菌耐药模式的影响

Klebsiella pneumoniae is one of the bacterial species with high antibiotic resistance. The present study aimed to investigate the effect of Minimal Inhibitory Concentration (MIC) of Ceftriaxon and Gentamicin on the antibiotic pattern of multi-drug resistant K. pneumoniae isolated from urinary tract infections. Sixty K. pneumoniae isolates have been isolated from 300 urine specimens collected from patients with urinary tract infections. Vitek 2 Compact System was used for identification and detection of antibiotic resistance patterns of bacterial isoltes. The highest percentage of antibiotic resistance of bacterial isolates to ampicillin (100%), followed by cefazolin (53.3%), while the lowest percentage of resistance was shown to Imipenem and tigecycline, which were 5% and 3.3%, respectively. The effect of MIC of gentamicin and ceftriaxon toward 10 selected isolates were used to select mutated isolates, then the effect of both antibiotic-on-antibiotic resistance of mutated isolates was detected using Vitek 2 Compact System. The results of the mutation experiment using ceftriaxone gave successful results to 7 isolates while 5 isolates gave positive results to gentamicin. The results of antibiotic resistance pattern of mutated isolates showed that all mutated isolates became resistance to most tested antibiotics in comparison with original isolates. We concluded that indiscriminate and wrong use of antibiotics would lead to emergence of MDR isolates not only for antibiotics belonging to the same class but also to other classes.

Emerging Role of Folate-Mediated One Carbon Metabolism in Leukemia: A Review 叶酸介导的一碳代谢在白血病中的新作用

Nearly 2.5% of all new cancer cases and 3.1% of cancer-related deaths were attributable to Leukemia. Leukemia can strike at any age geographical and ethnic differences in leukemia rates are also present. Folate's significance in cancer was exclusively tied to antifolate-based cancer treatment. After discovering Antifolate associated based chemotherapy, contrary to the inhibitory action of antifolate on tumors, epidemiologic, clinical, and experimental data show that folate deficiency in normal tissues may predispose them to neoplastic transformation and that folate supplementation may suppress the development of tumors in normal tissues. Epidemiologic research reveals a negative connection between folate levels and numerous cancers, including Leukemia. The exact association between folate status and certain cancers is unknown. Vitamin B control one-carbon metabolism and human health. Folate-mediated one carbon (1C) metabolism supports a of processes that are essential for the cell. Through a number of interlinked reactions happening in the cytosol and mitochondria of the cell, folate metabolism contributes to de novo purine and thymidylate synthesis, to the methionine cycle and redox defense. Targeting the folate metabolism gave rise to modern chemotherapy. The folate and methionine cycles complete the one-carbon route with vitamin B9 or folate. Vitamin B6 regulates carbon cycling. Vitamin B dysregulation alters biochemical signaling and causes several illnesses.

Analysis of Chosen Polymorphisms rs1982073 T/C, rs1800471 C/G TGFβ1 in Pathogenesis of Hashimoto's Diseases rs1982073 T/C、rs1800471 C/G tgf - β1在桥本病发病机制中的选择多态性分析

TGFβ-1 levels and its gene polymorphisms in Hashimoto's Thyroiditis (HT) patients and the immune system attack of the thyroid gland cells, by the presence of antibodies, including thyroglobulin antibody (TG-AB) and thyroid peroxidase antibody (TPO-AB), which serve as biochemical markers for Hashimoto’s Thyroiditis. A case-control study encompassed a cohort of 100 female subjects within the age range of 20 to 53 years, the study consisted of a group of diagnosed Hashimoto's thyroiditis patients and an equivalent number of healthy volunteers who were carefully selected to match the age and gender characteristics of the patient group. The study employed allele-specific arm-PCR technique to examine the genotype of TGF-β1 gene polymorphisms. Furthermore, levels of T3, T4, TSH, fT3, fT4, Anti TPO, and Anti-TG were quantified using the ELISA method, utilizing specific commercial kits designed for analysis within the respective study groups. Current work showed a highly significant variation in TGFβ1 levels in HT patients group when compared with healthy group, in addition to these findings suggest that the presence of the CG, GG genotype of rs1800471 and the CT, TT genotypes in rs1982073 are associated with an increased susceptibility to HT disease. The present study concluded that genotypes distribution and associated risks highlight the potential role of this genetic variation in the development and manifestation of HT disease.

The angiotensin-converting enzyme 2 (ACE2) is an enzyme of the renin-angiotensin-aldosterone system (RAAS), which catalyzes the conversion of some other angiotensin. ACE2 is widely expressed in different tissues. Many researchers reported many microRNAs involved in ACE2 expression, hsa-mir-181a one of these microRNAs which involved in ACE2 expression. In this study, we develop a comparative method (miR-Explore) to demonstrate that clustering of vaccinated miRNAs hsa-mir-181a of gene ACE2-alters the sensitivity and specificity of the prediction method even when a simple direct alignment of the positional secondary structure is used. The basic idea of the current approach was to generate a predictive visualization of the miRNAs hsa-mir-181a consensus structure for each vaccinated and unvaccinated miRNAs hsa-mir-181a class and use this consensus structure to perform alignment with the query sequence A set of data was taken from pre-miRNA by extracting total RNA and converting it to cDNA, then reading the DNA sequence and following up the in situ secondary structure of these sequences by means of dedicated software programs and websites and comparing them with each other and with sequences available in the microRNA database website

In this study, we develop a comparative method (miR-Explore) to demonstrate that clustering of vaccinated miRNAs hsa-mir-181a of gene ACE2-alters the sensitivity and specificity of the prediction method even when a simple direct alignment of the positional secondary structure is used. The Role of Serum Methylglyoxal and Glyoxalase 1 on Diabetic Peripheral Neuropathy 血清甲基乙二醛和乙草醛酶1在糖尿病周围神经病变中的作用

Diabetic neuropathy (DN) is a characteristic microvascular complication linked to diabetes. It affects about 50% of those who have this ailment. It has been revealed that Advanced Glycation End Products (AGEs) play a role in the development of diabetic neuropathy. Patients diagnosed with type 2 diabetes (T2DM) have higher quantities of methylglyoxal (MG), particularly harmful precursor of AGEs. One of the numerous defensive mechanisms responsible for MG metabolism and prevention of the generation of AGEs is the glyoxalase system. The cytotoxic byproduct methylglyoxal (MG) is removed by the detoxifying enzyme glyoxalase 1 (GLO1) via converting it to D-lactate, which is not harmful to tissues. This study intends to unveil the role of serum methylglyoxal and glyoxalase 1 on the onset of diabetic peripheral neuropathy (DPN). The present research was designed as a case-control study; including 160 individuals (males and females) aged (30-60) years. The subjects were divided into three groups: group one includes 40 type 2 diabetic patients with peripheral neuropathy, group two includes 40 type 2 diabetic patients without peripheral neuropathy and group three includes 80 apparently healthy as the control. Blood specimens were collected from all patients conducted at the National Diabetes Center, Mustansiriyah University in Baghdad, Iraq. Serum was used to measure glyoxalase 1 and Methylglyoxal levels using Enzyme-Linked Immunosorbent Assay (ELISA) technique. Whole blood with ethylene diamine tetraacetic acid (EDTA) was employed to measure glycated hemoglobin (HbA1c) levels with standard biochemical technique (the CLOVER system). Anthropometric measurements, such as height, weight, BMI and duration of diabetes were assessed. Individuals in control group showed higher serum level of glyoxalase 1 (26.4±4.06 ng/ml) than either patients with DPN (13.61±2.22 ng/ml) or patients with T2DM without peripheral neuropathy (15.25±1.68 ng/ml) with highly significant differences. In contrast, patients with DPN demonstrated higher level of methylglyoxal (29.45±6.06 ng/ ml) than either patients with T2DM without peripheral neuropathy (23.76±5.66 ng/ml) or controls (14.04±4.46 ng/
