Bioconversion of waste paper materials to sugars: an application illustrating the environmental benefit of enzymes 废纸原料生物转化为糖:说明酶的环境效益的应用

Recycling of waste materials into useful products is very topical not only to protect natural resources, but also to limit environmental pollution. The enzymatically catalyzed bioconversion of organic based waste such as used paper materials to soluble sugars like glucose can be performed in an environmental-friendly way. A basic experiment is proposed to show how different paper materials exhibit varying susceptibilities towards cellulase catalyzed saccharification, and that this approach can be applied by biochemistry or biotechnology students in the bioconversion of different cellulose related waste materials.

Sex determination using the polymerase chain reaction 用聚合酶链反应测定性别

A practical class experiment on the PCR is described which has been used over several years as part of an undergraduate biochemistry and molecular biology course for science students. A major aim is to provide experience in the use of the polymerase chain reaction (PCR) and its interpretation. Students are given small coded DNA samples and use the PCR reaction to determine whether the sample is from a male or a female.

The electrophoretic mobility shift assay is a simple and rapid method for visualizing the existence of specific DNA-protein interactions and is a useful teaching experiment. In the experiment described here the students prepare nuclear extracts, a DNA probe, and can successfully produce specific DNA-protein complexes and estimate physicochemical parameters characterizing the interaction. The system described is easy to perform. The synthesis of the oligonucleotides required and the labeled deoxyribonucleotide used for labeling the probe are the main costs of the experiment, but since the synthesized oligonucleotides can be used in several courses and the amount of labeled deoxyribonucleotide required for labeling the probe is small the experiment is relatively inexpensive to put on with a class.

This article describes a laboratory class experiment for an undergraduate Biochemistry course for Chemistry students. The experimental work involves the extraction of lovastatin from a medicament utilized in the treatment of hypercholesterolemic patients, and its determination by spectroscopy and by reversed-phase high-performance liquid chromatography. The entire experiment can be performed in one laboratory period, e.g. less than one day.
