Tetrandrine: a potent abrogator of G2 checkpoint function in tumor cells and its mechanism. 粉防己碱:肿瘤细胞中G2检查点功能的有效抑制剂及其机制。

Objective: To assess the ability of tetrandrine (Tet) to enhance the sensitivity to irradiation and its mechanism in cell lines of human breast cancer p53-mutant MCF-7/ADR, p53-wild-type MCF-7 and human colon carcinoma p53-mutant HT-29 as well as in C26 colorectal carcinoma-bearing BALB/c mice.

Methods: MCF-7/ADR, HT-29 and MCF-7 cells were exposed to irradiation in the absence or presence of tetrandrine. The effect of Tet on the cytotoxicity of X-irradiation in these three cells was determined and the effect of tetrandrine on cell cycle arrest induced by irradiation in its absence or presence was studied by flow cytometry. Moreover, mitotic index measurement determined mitosis of cells to enter mitosis. Western blotting was employed to detect cyclin B 1 and Cdc2 proteins in extracts from irradiated or non-irradiated cells of MCF-7/ADR, HT-29 and MCF-7 treated with tetrandrine at various concentrations. Tumor growth delay assay was conducted to determine the radio-sensitization of tetrandrine in vivo.

Results: Clonogenic assay showed that tetrandrine markedly enhanced the lethal effect of X-rays on p53-mutant MCF-7/ADR and HT-29 cells and the sensitization enhancement ratio (SER) of tetrandrine was 1.51 and 1.63, but its SER was only 1.1 in p53-wt MCF-7 cells. Irradiated p53-mutant MCF-7/ADR and HT-29 cells were only arrested in G2/M phase while MCF-7 cells were arrested in G1 and G2/M phases. Radiation-induced G2 phase arrests were abrogated by tetrandrine in a concentration-dependent manner in MCF-7/ADR and HT-29 cells, whereas redistribution within MCF-7 cell cycle changed slightly. The proportion of cells in M phase increased from 1.3% to 14.7% in MCF-7/ADR cells, and from 1.5% to 13.2% in HT-29 cells, but 2.4% to 7.1% in MCF-7 cells. Furthermore, the levels of cyclin B 1 and Cdc2 expression decreased after X-irradiation in MCF-7/ADR and HT-29 cells, and the mitotic index was also lower. Tet could reverse the decrease and induce the irradiated cells to enter mitosis (M phase). Endosomatic experiment showed that tetrandrine caused tumor growth delay in irradiated mice.

Conclusion: Tetrandrine boosts the cell killing activity of irradiation both in vitro and in vivo. Tetrandrine is a potent abrogator for G2 checkpoint control and can sensitize the cells to radiation.

Contamination of aflatoxins in different kinds of foods in China. 中国不同食品中黄曲霉毒素的污染。

Objective: To study the contamination of total aflatoxins (AFs) in different kinds of foods including corn, peanut, rice, walnut, and pine nut in six provinces and two municipalities in China.

Methods: A total of 283 samples of corn, peanut, rice, walnut and pine nut were randomly collected from local markets in Fujian, Guangdong, Guangxi, Hubei, Jiangsu, and Zhejiang provinces, as well as in Shanghai and Chongqing municipalities. The samples were ground to which acetonitrile/water solution was added. After filtering, the extract was transferred into a MycoSep purifying column and was pressed slowly. Then the purified liquid was derivatized with trifluoroacetic acid (TFA) and assayed using high performance liquid chromatography (HPLC).

Results: AFs were detected in 70.27% of corn samples, with a mean level of 27.44 microg/kg and the highest level of 1098.36 microg/kg. In peanut, the AFs detection rate was 23.08%, with a mean level of 0.82 microg/kg and the highest level of 28.39 microg/kg. Very few rice samples with AFs were detected. The AFs levels were very low in walnut and pine nut.

Conclusion: Corn is the food most seriously contaminated with AFs in China. AFB1 is the main aflatoxin which is found as a contaminant in foods.

Characteristics of Zn2+ biosorption by Saccharomyces cerevisiae. 酿酒酵母对Zn2+的生物吸附特性

Objective: To investigate the characteristics of Zn2+ biosorption and the release of cations during the process of Zn2+ biosorption by intact cells of Saccharomyces cerevisiae.

Methods: The batch adsorption test was used to study the biosorption equilibrium and isotherm. Zn2+ concentration was measured with atomic adsorption spectrophotometer (AAS) AAS 6 Vario.

Results: When the initial concentration of Zn2+ ranged between 0.08 and 0.8 mmol/L, the initial pH was natural (about 5.65), the sorbent concentration was about 1 g/L and the capacity of Zn2+ biosorption was from 74.8 to 654.8 micromol/g. The pH value increased by 0.55-1.28 and the intracellular cations (K+, Mg2+, Na+, Ca2+) of the cells were released during the process of Zn2+ biosorption.

Conclusion: Ion exchange was one of the mechanisms for Zn2+ biosorption. The biomass of Saccharomyces cerevisiae is a potential biosorbent for the removal of Zn2+ from aqueous solution. More work needs to be done before putting it into practical application.
