Objective To explore the expressions of tripartite motif family protein 72 (TRIM72) in peripheral blood, pleural effusion and tumor tissue in patients with lung adenocarcinoma. Methods There were 80 cases diagnosed with lung adenocarcinoma collected in the affiliated hospital of Guangdong Medical University from August 2017 to February 2018 and 20 healthy subjects in the same period were taken as control group.The expression of TRIM72 was detected by immunohistochemistry, and ELISA kits were used to detect the TRIM72 in peripheral blood and pleural effusion.Also, clinical data were analyzed. Results TRIM72 was mainly expressed in normal airway epithelial cells but hardly expressed in lung adenocarcinoma tissues.TRIM72 level of peripheral blood was significantly lower in the patients with lung adenocarcinoma compared to control group (t=8.727, P<0.05), and negatively correlated with CEA (r=-0.556, P<0.001). TRIM72 level in malignant pleural effusion is significantly lower than that in inflammatory effusion (t=10.09, P<0.05). TRIM72, CEA, CA125 were used to make receiver operating characteristics curve for the diagnosis of lung adenocarcinoma.The areas under the curve were 0.927, 0.746, 0.851, with a cut-off 110.629 ng/L, 4.535 μg/L, 33.965 U/ml respectively.When TRIM72 combined with CEA and CA125, the area under the curve could reach 0.974. Conclusions Serum TRIM72 may be a potential biomarker for the diagnosis and the prognosis of lung adenocarcinoma. Currently, there are still many problems in screening and early diagnosis of lung cancer.Noninvasive and convenient methods with high specificity and sensitivity are urgently needed to improve the efficiency of screening and early diagnosis of lung cancer.A number of studies published in recent years have shown that test of a panel of tumor-associated autoantibodies in screening and early diagnosis of lung cancer exhibits perfect specificity and fine sensitivity.In addition, combined use of a panel of autoantibodies and low dose CT can reduce false positive rate, improve specificity and positive predictive value of CT, and could be helpful in screening and early diagnosis of lung cancer.This article reviews generation mechanism, characteristics, diagnostic performance and problems of tumor-associated autoantibodies in detection of lung cancer, especially in the diagnosis of early-stage lung cancer. Objective To investigate the role of Pokemon protein and P14ARF-MDM2-p53 pathway proteins in the pathogenesis of non-small cell lung cancer (NSCLC). Methods This study included 35 NSCLC patients who underwent surgical treatment from the First Hospital of Hebei Medical University from October 2015 to October 2016.At the same time, 29 cases of normal bronchus stump adjacent to the cancer and 25 cases of pneumonia were used as the bronchial control group and alveolar control group, respectively.Immunohistochemistry was used to detect the expressions of Pokemon, P14ARF, MDM2 and p53 proteins in NSCLC group, bronchial control group and alveolar control group, and to analyze their relationship with clinicopathological data and their correlations. Results The expressions of Pokemon, MDM2 and p53 proteins in NSCLC group were significantly higher than those in the bronchial control group and the alveolar control group (all P values <0.01). The expression of P14ARF protein in NSCLC group was significantly lower than that in the bronchial control group and alveolar control group (χ2=25.39, P<0.01). The expressions of p53 and MDM2 protein were correlated with the degree of differentiation of NSCLC (P=0.02, 0.03). In NSCLC group, Pokemon was negatively correlated with P14ARF protein expression (r=-0.58, P<0.05), and P14ARF was negatively correlated with MDM2 protein expression (r=-0.51, P<0.05), MDM2 was positively correlated with p53 protein expression (r=0.86, P<0.05). Conclusions Pokemon plays an oncogene role in the pathogenesis of NSCLC by regulating the P14ARF-MDM2-p53 pathway. Acute respiratory distress syndrome (ARDS) is a heterogeneous clinical syndrome with high morbidity and mortality.Diffuse alveolar damage (DAD) is the typical pathological feature of ARDS, but pulmonary autopsies and lung biopsies have found that only about half of patients meeting the Berlin definition of ARDS have DAD.Compared with non-DAD-ARDS patients, the mortality rate of DAD-ARDS patients is significantly higher, there may be different cellular and molecular mechanisms.This article aims to explore the relationship between ARDS and DAD and its clinical significance, in order to provide new ideas for the better diagnosis and treatment of ARDS. 2019 is the fourth year for the global to enter the era of sustainable development goals in 2030 with end tuberculosis strategy.It is also the end year whether the first milestone goal set by 2020 can be achieved.Based on the full recognition of the progress made by countries in tuberculosis control, the global tuberculosis report emphasized further analyzing the problems and challenges existing in national tuberculosis programme carefully and objectively, clarifying the action requirements.Learning good practices and experience shown in the committed action towards end tuberculosis strategy from global will facilitate us to make precise actions for ending tuberculosis in China. Objective To study the role of hypoxia-inducible factor-1α (HIF-1α) in regulating the acute inflammation of lung. Methods Hif-1α knockout (Hif-1α+ /-) mice and their wild-type (Hif-1α+ /+ ) were stimulated with lipopolysaccharide to establish acute lung injury (ALI) model, respectively.The bronohoalveolar lavage fluid (BALF), blood and lung tissue were collected at 6th, 12th, 24th and 48th hour after LPS treatment.The number of total leukocytes and neutrophils in BALF of ALI model group and control group were counted.The levels of inflammatory factors such as tumor necrosis factor-α and KC in the serum of each group were detected by enzyme linked immunosorbent assay.The neutrophil infiltration in airway and lung tissue of all groups was assessed by hematoxylin-eosin staining.And the neutrophil apoptosis in lung tissue of all groups was detected by TUNEL. Results The number of leukocytes and neutrophils in BALF of Hif-1α+ /- and Hif-1α+ /+ ALI mice was significantly higher than that of control mice.And there was no significant difference in the number of leukocytes in BALF between Hif-1α+ /- and Hif-1α+ /+ ALI mice.But compared to Hif-1α+ /+ ALI mice, the number of neutrophils in BALF of Hif-1α+ /- ALI mice was significantly lower (P<0.05). The levels of TNF-α and KC in serum of both ALI model mice were significantly higher than those in control group.But the levels of TNF-α and KC in the Hif-1α+ /- ALI mice were significantly lower than those in the Hif-1α+ /+ ALI mice.Hematoxylin-eosin staining indicated that there were a large amount of neutrophils infiltrating into the lung tissue and airway of Hif-1α+ /- ALI mice, which were much more than those of Hif-1α+ /+ ALI mice.TUNEL staining further revealed that there were a large number of apoptotic neutrophils in the lung tissue of Hif-1α+ /- ALI mice, which were significantly more than those of Hif-1α+ /+ ALI mice. During the long-term evolution of the host environment, Mycobacterium tuberculosis can completely or partially adapt to survive in the host cells by avoiding or modifying the host response to infection.Many factors contribute to the successful invasion of this pathogen into host cells.Over the past few decades, a great deal of researches have led to deeper understanding of the complex pathogenesis of Mycobacterium tuberculosis, especially its unique Ⅶ secretion system and the cell membrane with a variety of complex lipid have become the hot topic of the aspects.This review summarizes the recent research results of Mycobacterium tuberculosis associated virulence factors, hoping to find new drug targets from these virulence factors. Objective To summarize the clinical features of interstitial pneumonia with autoimmune features (IPAF) and to explore the correlation between autoantibodies and imaging findings. Methods IPAF patients hospitalized in the Department of Respiratory and Critical Care Medicine, Zhongnan Hospital of Wuhan University from January 2015 to September 2018 were enrolled in this study, who met with the serology and imaging features.The clinical data were retrospectively analyzed. Results 517 patients with interstitial lung disease were collected in this study, including 78 cases of IPAF (15.1%). Most of the autoantibodies were antinuclear antibody (ANA)(48.7%) and anti-Ro-52 (47.4%). Non-specific interstitial pneumonia (NSIP) was the most common type of high-resolution computed tomography (HRCT) in the chest, accounting for 62.8%.Usual interstitial pneumonia (UIP) was mainly seen in male (P=0.019) smokers (P=0.004). Patients with organized pneumonia (OP) were significantly younger than Patients with UIP (P=0.040) or NSIP (P=0.002). When the autoantibodies were positive for ANA (P<0.001) and anti-Ro-52 (P=0.011), the most common type of HRCT was NSIP.The incidence of mixed ventilation dysfunction in NSIP patients with OP was significantly higher than that in NSIP patients (P=0.020), and the incidence of diffusion dysfunction in NSIP patients was significantly higher than that in UIP patients (P=0.029). Conclusions When the autoantibodies are positive for ANA and anti-Ro-52, the HRCT type is mainly NSIP. Objective To investigate the possibility of cecal ligation puncture (CLP) combined with lipopolysaccharide (LPS) to establish acute respiratory distress syndrome (ARDS) model in rats, and to explore its significance and theoretical basis. Methods 176 SD rats were randomly divided into four groups, 44 rats in each group: (1)CLP group; (2)injection of LPS (2 mg/kg) through tail vein (LPS group); (3)CLP combined with injection of LPS (2 mg/kg) through tail vein (CLP+ LPS group); (4)injection of physiological saline (2 ml/kg) through tail vein after sham operation (control group). Each group was divided into four subgroups.The respiratory rate of rats, total number of white blood cells, percentage of polymorphonuclear leukocytes, total protein content, tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in bronchoalveolar lavage fluid, and ratio of lung wet to dry weight were measured at 4th, 12th, 24th, and 48th hour.Observation of histopathological and transmission electron microscope on lung tissue was performed. Results Compared with the control group, the respiratory rate, total protein content, TNF-α, white blood cells, percentage of polymorphonuclear leukocytes, and IL-6 level increased at 4th hour in LPS group and CLP+ LPS group (all P<0.05), and increased at 12th hour in CLP group (all P<0.05). The above parameters in CLP group and CLP+ LPS group increased significantly at 24th hour (all P<0.05), while those tended to recover in LPS group.Respiratory frequency and IL-6 level were increased in CLP+ LPS group at all time points (all P<0.05). The results of pathological morphology and transmission electron microscopy showed that the inflammatory response and alveolar cell damage were the most severe in CLP+ LPS group at 24th hour. Conclusions Compared with the CLP or LPS models, lung injury is earlier and duration is longer in the rat models established by CLP combined with LPS.It is closer to the diagnostic criteria of ARDS on clinical manifestations and lung injury, and it is an ideal ARDS model. Objective To explore the prognostic value of neutrophil-to-lymphocyte ratio (NLR) in acute respiratory distress syndrome (ARDS). Methods ARDS patients visiting the Second People′s Hospital of Suzhou Xiangcheng and Eastern District of Suzhou Municipal Hospital from January 2016 to October 2018 were enrolled and clinical information were recorded.Correlation analysis, multivariate regression analysis, and survival analysis were used to explore the prognostic value of NLR. Results A total of 80 ARDS patients were included and the median value of NLR was 14.The age, proportion of invasive mechanical ventilation, NLR, and creatinine in non-surviving patients were significantly higher than those in surviving patients, while oxygenation index and plasma albumin in non-surviving patients were significantly lower than those in surviving patients.NLR was significantly associated with overall survival (HR=1.011, 95%CI: 1.004-1.017, P=0.001). In the multivariate Cox regression model, it was further identified that NLR>14 as an independent prognostic factor for overall survival (HR=1.532, 95%CI: 1.095-2.143, P=0.001 3). Hospitalized mortality, 28-day mortality, 90-day mortality, and proportion of invasive mechanical ventilation in ARDS patients with NLR>14 were significantly higher than those with NLR≤14.
