Effects of RU 486 on electrical activity, on sexual steroid and prostaglandin F2α concentrations in the myometrium at mid-pregnancy in the rat ru486对妊娠中期大鼠肌层电活动、性类固醇和前列腺素F2α浓度的影响

Effects of RU 486 (10 mg·kg–1, per.os) were assessed at mid-pregnancy in the rat. One hour after RU 486 treatment, myometrial electrical activity stayed low. It increased from the 3rd hour after administration of RU 486 and a perfect synchronization of the bursts of the action potentials was observed from the 6th h to the 24th h. Tissular steroid hormones and PGF, evaluated at hour 6 after RU 486 administration, showed a decrease of progesterone concentrations in both myometrium and uterus. Estradiol levels decreased in uterus whereas, PGF levels increased in both myometrium and uterus. These results show for the first time that RU 486 strongly increases the myometrial electrical activity in the rat at mid-pregnancy. This action was closely related to E2, P4 and PGF concentrations.

The public decision-making methods in transport are based on cost–benefit analysis, by which the consequences of the decision (standards for vehicles, new infrastructures…) are converted in monetary amounts and compared to the cost of implementation of the decision. But some of these consequences, especially those related to environment, are not directly expressed in monetary terms. The article aims at offsetting this difficulty in the case of noise. The possible methods for getting money values of noise are presented; it is shown that the estimates to which they lead are coherent and consistent. Then a comparison is made between the present procedures and the procedures which could be implemented, and it is shown that large gains of efficiency could be obtained.

Hemochromatosis at the intersection of classical medicine and molecular biology 血色素沉着症在经典医学和分子生物学的交叉点

Hemochromatosis is a genetic disease of iron overload due to intestinal hyperabsorption of iron. It is one of the most prevalent autosomal recessive diseases in Caucasian populations. Hemochromatosis causes severe visceral and metabolic complications at adulthood, which include cirrhosis, diabetes, arthropathy and cardiac failure. A major breakthrough has been the discovery, in 1996, of the HFE gene which is strongly associated with the phenotypic expression of the disease. This discovery has, very quickly, provided a powerful genetic blood test which permits, in most cases, to establish the diagnosis in a non invasive way (i.e.without a liver biopsy). Hemochromatosis can be cured by repeated venesections provided the diagnosis has been detected sufficiently early. Moreover, an efficient preventive strategy can be applied to family members and should now be proposed to the general population. Finally, the identification of the HFE gene has paved the way for the identification of new iron overload entities.

Superficial architecture of the jaw-closing muscles of the cat (Felis catus): the temporo-masseteric complex 猫闭颚肌的表面结构:颞-咬肌复合体

The muscle fiber fascicles of the temporo-masseter complex of the cat were minutely dissected. Some heads were embedded in paraffin while others were put into methyl-methacrylate resin and sections were made. The results of this anatomical study demonstrate that this complex consists of the masseter muscle, the temporal muscle and two well individualized transitional fascicles: the maxillomandibularis and zygomatocomandibularis muscles. The masseter and temporal muscles are composed of individualized compartments in which the orientation and aponeuroses of the fibers of which they are composed differ with regard to the centric occlusion plane. The masseter muscle consists of a superficial fascicle made up of two layers, an intermediate fascicle, and a deep fascicle composed of two layers. The temporal muscle consists of one anterior orbital part and one posterior temporal part. This structure is in accordance with the mammalian archetype described by Gaspard and Saban. These findings should lead towards a homology-based nomenclature founded on comparative anatomy studies of mammalian species. Such a classification would permit the comparison of results obtained from physiological and histochemical studies of these complex muscle fibers when they are published by different researchers.

Is there chaos in the brain? I. Concepts of nonlinear dynamics and methods of investigation 大脑有混乱吗?非线性动力学的概念和研究方法

In the light of results obtained during the last two decades in a number of laboratories, it appears that some of the tools of nonlinear dynamics, first developed and improved for the physical sciences and engineering, are well-suited for studies of biological phenomena. In particular it has become clear that the different regimes of activities undergone by nerve cells, neural assemblies and behavioural patterns, the linkage between them, and their modifications over time, cannot be fully understood in the context of even integrative physiology, without using these new techniques. This report, which is the first of two related papers, is aimed at introducing the non expert to the fundamental aspects of nonlinear dynamics, the most spectacular aspect of which is chaos theory. After a general history and definition of chaos the principles of analysis of time series in phase space and the general properties of chaotic trajectories will be described as will be the classical measures which allow a process to be classified as chaotic in ideal systems and models. We will then proceed to show how these methods need to be adapted for handling experimental time series; the dangers and pitfalls faced when dealing with non stationary and often noisy data will be stressed, and specific criteria for suspecting determinism in neuronal cells and/or assemblies will be described. We will finally address two fundamental questions, namely i) whether and how can one distinguish, deterministic patterns from stochastic ones, and, ii) what is the advantage of chaos over randomness: we will explain why and how the former can be controlled whereas, notoriously, the latter cannot be tamed. In the second paper of the series, results obtained at the level of single cells and their membrane conductances in real neuronal networks and in the study of higher brain functions, will be critically reviewed. It will be shown that the tools of nonlinear dynamics can be irreplaceable for revealing hidden mechanisms subserving, for example, neuronal synchronization and periodic oscillations. The benefits for the brain of adopting chaotic regimes with their wide range of potential behaviours and their aptitude to quickly react to changing conditions will also be considered.

Métabolisme lipidique du champignon endomycorhizien : Glomus intraradices

The use of monoxenic cultures of the obligately biotrophic vesicular arbuscular fungus Glomus intraradices now permits investigation of the lipid metabolism of this organism. In bicompartmental culture plates, sporulating extraradical hyphae can be obtained, totally free of roots, and then provided with 14C-acetate as lipid precursor. Three experimental stages were studied: i) stage A, symbiotic stage corresponding to the fungus still attached to the host plant roots, ii) stage B, consisting of the fungus detached from the host roots, iii) stage C, germinating spores. In each case, the fungus proved to be able to synthesise its own lipids: 1,2- and 1,3-diacylglycerols, triacylglycerols, phospholipids, sterols and free fatty acids, de novo. Lipid metabolism varied with the experimental conditions. Phospholipid synthesis was intensive in germinating spores. Thus the obligately biotrophic status of this fungus cannot be explained by a deficiency in synthesis of these various lipid classes.

Immunocytochemical characterization of viruses and antigenic macromolecules in viral vaccines 病毒疫苗中病毒和抗原大分子的免疫细胞化学特性

Gold immunolabeling combined with negative staining (GINS) provides a valuable immunocytochemical approach that allows a direct ultrastructural definition of all viral vaccine constituents that share common antigenic features with pathogenic viral particles. These results have implications for the development of viral vaccines since it has been demonstrated that incomplete viral particles such as natural empty capsides and Rotavirus-like particles lacking the infective genome are potential candidates for the production of neutralizing antibodies. Furthermore comparative results of the application of GINS to either inactivated vaccines or unfixed samples provide direct evidence that even after inactivation specific antigenic sites are still available for gold immunolabeling.

Phosphorylation by PKA of a site unique to B-Raf kinase PKA对B-Raf激酶特有位点的磷酸化作用

The Raf kinases serve as central intermediates to relay signals from Ras to ERK. Cell-specific effects of these signals on growth, differentiation and survival can be observed due to the recruitment of different isoenzymes of the Raf family. The in vitro phosphorylation of a site unique to B-Raf (Ser429) has been proposed to be responsible for the negative regulation of the isoenzyme by Akt. Using phosphopeptide mapping and site-directed mutagenesis we showed that Ser429 is phosphorylated upon cAMP elevation in PC12 cells and proposed that PKA is a major kinase phosphorylating the B-Raf-specific site in vivo.

Régime alimentaire d’un ver de terre anécique des savanes colombiennes : une remise en question des types écologiques 哥伦比亚大草原枯萎蚯蚓的饮食:生态类型的挑战

An ecological classification based on a set of morphological characters may be used to describe earthworm functions in soil. However these relationships have been seldom verified. Gizzards of the earthworm Martiodrilus carimaguensis (Glossoscolecidae) were studied to find out if its anecic morphology was in agreement with its diet (shallow litter remains and deep organo-mineral material). By direct observation of 13 adults’ gizzard contents with a dissecting and a phase contrast microscope, the volumetric percentage of plant fragments, roots, amorphous organic matter and mineral particles reached respectively 63, 5, 6.5 and 30 % v/v. Eighty percent of plant fragments were microscopic, and mostly incorporated into the organo-mineral earthworm casts well preserved in the gizzard. Coprophagy and rhizophagy were the dominant feature of M. carimaguensis diet. Yet, the casts produced revealed a high variability in its diet, what could be related to transitional adoption of a typical anecic diet during some periods.

Phylogenetic relationships between seven species of the catfish species flock from Lake Malawi (genus Bathyclarias) and other Clariid catfish have been investigated using cytochrome b partial sequences. Here we demonstrate that this species flock originated from a widespread, generalist species, Clarias gariepinus, still occurring in the lake. Bathyclarias species and their ancestor C. gariepinus form a simple model that can be used to understand the mechanisms of adaptation and rapid speciation in African Great lakes.
