Anti-cancer perspectives of resveratrol: a comprehensive review 白藜芦醇抗癌研究综述

Resveratrol (RVT) is well known for its chemo-preventive and therapeutic attributes against the various kinds of cancers such as breast, prostate, pancreatic, oral, brain, lung and liver. It has been found to inhibit glioblastoma cell growth, declining uPAR and its mediator ERK1/2, inhibiting the cell lines (EC-9706) growth and ADAM9 expression, up-regulating the lncRNAs and PPARγ, inhibition of MTA1 to modulate oncogenic miR-34a, miR-22 and miR-17, enhance the E-cadherin expression and decreased the expressions of Twist1 and vimentin, inhibiting ADAM9 expression, downregulation of ATP2A2 and ATP2A3 genes expressions in different breast cell lines, reduction in IL-6 and COX-2 expression, inhibition of Tax activity of LTR and HTLV-1 in SIRT1 deacetylase, suppression of epithelial-to-mesenchymal transition (EMT) and eEF1A expression and fracas interactions between TEAD and YAP and activation of Hippo/YAP signalling. Consequently, it can be suggested that RVT is a promising agent for treating aforementioned cancers due to its curing characteristics.

Development of multiplex immunochromatographic assay for the simultaneous detection of three polyether ionophore antibiotics in chicken muscle 多重免疫层析法同时检测鸡肌肉中三种聚醚离子载体抗生素的建立

A rapid, reliable, sensitive, and multiplex colloidal gold immunochromatographic assay (CGIA) was developed for the simultaneous detection of three polyether ionophore antibiotics (PEs) in chicken muscle. The IC50 values of the optimized CGIA were 1.63, 1.94, and 1.18 ng/mL with a linear range of 0.87–3.04 ng/mL, 0.85–4.41 ng/mL, and 0.57–2.44 ng/mL for maduramicin (MAD), monensin (MON), and salinomycin (SAL), respectively. The cut-off values were 10, 5, and 5 ng/mL, respectively. The whole testing process was completed within 15 min. A parallel analysis in blind chicken muscle samples was conducted by liquid chromatography–tandem mass spectrometry (LC-MS/MS), the developed CGIA did not appear false-positive and false-negative. The results show that the established method could provide a rapid and effective approach for on-site screening and determination of PEs residues in chicken muscle samples.

Sensitivity, specificity and positive predictive value of ALEX2 multiplex examination in patients suffering from atopic dermatitis and reaction to tomatoes ALEX2多重检查在特应性皮炎及番茄反应患者中的敏感性、特异性及阳性预测值

The aim of the study is to evaluate the clinical reactions to tomatoes in atopic dermatitis (AD) patients, and to analyse the results of specific IgE to allergens of tomatoes and to molecular components of pollen, seeds, fruits, vegetables and latex with the use of ALEX2 Allergy Xplorer test. According to our results, 15% of AD patients suffer from clinical reactions to tomatoes; we confirmed the low sensitivity and low positive predictive value of specific IgE to molecular component Sola l 6 and allergen extract Sola l, the specificity of this test is higher. The significantly higher sensitisation to other nonspecific lipid transfer proteins is observed in patients sensitised to tomato allergens. An elimination diet and an exposure test are recommended to detect a reaction to tomatoes. ALEX2 Allergy Xplorer test gives us a comprehensive picture of sensitisation and the possibility of evaluation of cross-reacting allergens.

Plant chemodiversity is a helpful tool for disease prevention and a basis for adjuvant treatments to conventional therapies. In this regard, the extract of Polypodium leucotomos rhizomes, PLE, has shown benefits fighting inflammation and recurrent infections but its molecular mechanism is poorly undersood. This work shows that Plesinox 3A, containing PLE, Vitamins A, C, and selenium, helps modulate the initial inflammatory response triggered by bacterial LPS through the upregulation of IL8 and IL10, together with downregulation of COX2, IL1B and TNF, in a more efficient manner than PLE alone. Additionally, this formulation enhances the antiviral response through the upregulation of MX1, IFNA1 and IFNG in different cell types. Finally, the addition of vitamins and selenium to PLE in Plesinox 3A greatly boosts the anti-bacterial properties of PLE alone. Overall, these findings support the combined use of PLE, vitamins, and selenium, in the form of Plesinox 3A as an immune booster to prevent recurrent infections, highlighting a gene set potentially involved in its beneficial effect, as well as showing its direct anti-bacterial properties, which are greater than those of PLE alone.

A simple and rapid DNA extraction method from meat by microneedle patch for detection of adulterated components 采用微针贴片法提取肉类中掺假成分的DNA

ABSTRACT The simple and rapid identification of meat via DNA-based methods is still subject to the conditions of complex and time-consuming preprocessing. It usually takes 3–4 h to obtain DNA sample using conventionally available DNA extraction kits. The existing approaches are cumbersome, require large equipment, and are not suitable for field operation. This study developed a swelling microneedle patch, constituted by micron-sized copolymers of methyl vinyl ether and maleic acid (PMVE/MA). The microneedle patch was applied to the meat sample for 1 min before being pulled out and rinsed with Tris-EDTA (TE) buffer to obtain DNA, which could be used for polymerase chain reaction (PCR) without purification. The DNA extracted by the microneedle patch could meet the needs for amplification and identification of meat samples. The developed approach which combines the patch-based extraction method and conventional PCR amplification is quick, simple, and reliable for DNA extraction and identification of meat samples.

Influence of Porphyra-derived polysaccharides and oligosaccharides on attenuating food allergy and modulating enteric microflora in mice 卟啉多糖和寡糖对减轻小鼠食物过敏和调节肠道菌群的影响

ABSTRACT Recently, there have been reports on the immunomodulatory properties of polysaccharides (PP), oligosaccharides (PO), and mixtures of PO/PP derived from Porphyra. To explore their potential in alleviating allergic responses and enteric dysbiosis, this study delved deeper into their effects using a murine model of food allergy. Daily administration of Porphyra samples mitigated allergic diarrhea, enteritis, and allergen-specific IgE production. Moreover, Porphyra samples exerted a significant reduction in the secretion of IL-2, IL-4, IFN-γ, and IL-17A, along with an elevation in the IL-10/IL-4 ratio in splenocytes upon allergen stimulation. Similarly, expression of IL-4 and IFN-γ in the duodenum was suppressed. Notably, allergic mice displayed a distinct fecal microflora profile compared to that of normal mice. Interestingly, PP and PO treatments exhibited different effects on improving enteric dysbiosis. These findings collectively suggest the potential of PP and PO as functional food candidates for the management of allergic enteritis and dysbiosis.

ABSTRACT This study was carried out to investigate the effect of different iron sources on haemoglobin and myoglobin (Mb) synthesis, and the mRNA expression of the key genes related to iron metabolism in the skeletal muscle of piglets. Forty-eight piglets were assigned into four treatments including control group, ferrous sulphate (FeSO4) group, ferrous glycinate (Fe-Gly) group and amino acid-Fe(II)-chelator complexes group. The results showed that iron supplementation increased Mb concentration in muscles (P < 0.05). In addition, iron supplementation increased hephaestin (HEPH) expression in the longissimus dorsi muscle (P < 0.1), decreased transferrin receptor 1 (TFR1) expression in the longissimus dorsi muscle (P < 0.05) and increased lipocalin 2 (Lcn2) expression in gastrocnemius muscle, respectively (P < 0.05). In summary, compared with high-dose inorganic iron, low-dose organic iron supplements have the same effects in haemoglobin and myoglobin biosynthesis, and mobilize iron in muscles into circulation.

ABSTRACT Cordyceps cicadae is a rare edible and medicinal entomogenous fungus. Here, the structural characteristics and immunomodulatory activity of C. cicadae polysaccharide CP80-1 were preliminarily elucidated. It showed that CP80-1 of 250.431 kDa was mainly composed of glucose, xylose and rhamnose. Physiologically, CP80-1 can regulate the immune of RAW264.7 cells by promoting the proliferation, phagocytosis, and secretion of nitric oxide (NO), tumor necrosis factor (TNF)-α, interleukin (IL)−1β, and interleukin (IL)−6. However, these effects were significantly attenuated when Dectin-1 receptor expression was inhibited by laminarin. Moreover, laminarin down-regulated the mRNA levels of relevant genes, inhibited CP80-1-driven expression of proteins related to spleen tyrosine kinase (Syk) and nuclear factor kappa-B (NF-κB) signaling pathway. Additionally, CP80-1 induced the nuclear translocation of NF-κB subunit p65 in RAW264.7 cells. These results suggest that CP80-1 may activate RAW264.7 cells by regulating the Dectin-1/Syk/NF-κB signaling pathway, which provides theoretical basis for the development of C. cicadae in immunomodulation.

Molecular mechanisms and biomarkers underlying the protective roles of the nutraceutical laminarin against ischaemic strokes 营养药物层压蛋白对缺血性中风的保护作用的分子机制和生物标志物

ABSTRACT Stroke is the second leading cause of death and a major cause of disability worldwide. Ischemic stroke accounts for approximately >60% of all stroke, and >80% of strokes can be prevented. Middle cerebral artery occlusion (MCAO) is a common cause of stroke in humans. MCAO is associated with blood vessel-related problems, such as reduced vascular plasticity and hypertension. These problems can be attenuated by the use of nutraceuticals such as laminarin. Laminarin is a storage glucan commonly found in brown algae. Accumulating evidence and studies have revealed that laminarin is a promising candidate drug for treating ischaemic stroke. However, details on pharmaceutical targets and molecular mechanisms underlying laminarin’s beneficial effects for treating ischaemic stroke remains largely unknown. Herein, we applied network pharmacology, bioinformatic analysis, and middle cerebral artery occlusion model to delineate the protective role of laminarin. By comparing the laminarin- and MCAO-associated genes, we identified 23 potential targets including tumour necrosis factor (TNF), vascular endothelial growth factor A, selectin P, presenilin 1, fibroblast growth factor 2, microtubule-associated protein tau, caspase 3, matrix metallopeptidase 1, 5-hydroxytryptamine receptor 2A, telomerase reverse transcriptase, interleukin 2, signal transducer and activator of transcription 3 (STAT3), ATP binding cassette subfamily B member 1, catalase, superoxide dismutase 2, adenosine A1 receptor, adenosine A2a receptor, 5-hydroxytryptamine receptor 1B, heat shock protein 90 alpha family class A member 1, matrix metallopeptidase 8, BCL2-like 1, galectin 3, and epoxide hydrolase 2 of laminarin against ischaemic stroke. The gene ontology enrichment and Kyoto Encyclopedia of Genes and Genomes enrichment analyses highlighted the importance of this gene cluster in the development of blood vessels, neuronal cell death, brain functions, and neuroinflammation. Furthermore, molecular docking analysis suggested a direct binding of laminarin to its target proteins STAT3 and TNF. Our results provide the pharmaceutical targets and delineate the details regarding the molecular mechanisms underlying the beneficial effects of laminarin against ischaemic stroke. Moreover, our findings support those of previous studies suggesting laminarin as a promising drug for treating ischaemic stroke.

ABSTRACT The microbe Paecilomyces cicadae (Miquel) Samson (P. cicadae) is an asexual strain of Cordyceps cicadae (C. cicadae) used in traditional Chinese medicine. Due to the abundance of secondary metabolites found in cultured metabolites, including polysaccharides, hyaluronic acid, ergosterol peroxide, and active nucleosides, P. cicadae and/or C. cicadae exhibit several therapeutic activities, including immunoregulatory, antioxidant, and anti-diabetic effects. Given the great attention for health care and the desirable medical value of C. cicadae and/or P. cicadae, they are becoming potential alternatives of Cordyceps sinensis (C. sinensis). Although there are many studies on P. cicadae/C. cicadae, most of them are experimental research articles. There are limited reviews to summarize the latest researches on P. cicadae/C. cicadae. This review mainly focuses on the metabolites, physiological activities and mechanisms of action of P. cicadae and/or C. cicadae. This review can serve as a theoretical basis for the more efficient use of this medicinal fungus.
