ABSTRACT The aim of this study was evaluated the effect of selected bioactive substances and nanoparticles on the immunoreactivity of edible packages based on chitosan using the ELISA. The analysed protein was the tropomyosin. The results confirmed the presence of the tropomyosin (3.77 ± 0.79–5.75 ± 0.01 µg/g) in control samples. This study demonstrated that the bioactive substances in the form of grape (0.61 ± 0.34–0.43 ± 0.16 µg/g), blueberry (0.58 ± 0.32–0.39 ± 0.27 µg/g), and parsley extracts (2.09 ± 1.28–0.79 ± 0.40 µg/g) reduces immunoreactivity (p < 0.05) of the tropomyosin. The elder pollen had no significant effect (p > 0.05) on immunoreactivity. ZnO and TiO2 nanoparticles also demonstrated immunoreactivity reduction (p < 0.05). The exception was silver nanoparticles, where the immunoreactivity increased with increasing concentration of grape extract (1.29 ± 0.01–5.47 ± 0.25 µg/g). The results confirmed the inhibitory effect of bioactive substances on the immunoreactivity of the used ELISA and also showed the need to consider immunoreactive substances when interpreting ELISA results.

ABSTRACT The aim of this study was to reveal the mechanism of its wound healing function at the molecular level using transcriptome sequencing technology. The results showed that EAE mainly upregulated Cyclin A gene through AMPK signalling pathway, activated cell cycle pathway, promoted DNA synthesis, accelerated mitosis, and thus promoted the proliferation of Human immortalized keratinocytes (HaCaT) cells. In addition, analysis of downregulated gene enrichment revealed that these genes were significantly or very significantly enriched not only in the cytokine-cytokine receptor interaction pathway but also in the TNF and the NF-κB signalling inflammatory classical pathway. It was mainly through downregulation of genes such as Fn14, CCL2, CCL5, CARD14 and LTB that prevented inflammatory responses while delaying apoptosis. The results of this study provided important information for the molecular mechanism of wound healing of EAE and provided a basis for further development of functional products of raspberry.

ABSTRACT Schizophyllum commune is a fungus with applications both in medicine and foods, which produces IPS and EPS. However, there is no comparative study on the activity comparison of IPS and EPS and its mechanism of antioxidant effect. In this study, the antioxidant activities of the two polysaccharides were measured in biochemical, cellular and molecular levels, and H2O2 induced oxidative damage model of HSFs was established to explore the protective effects of EPS and IPS. We found IPS and EPS both achieved a good antioxidant effect. They can also improve the cellular levels of SOD and reduce MDA. Insights regarding the molecular mechanism were studied, we found that both EPS and IPS could increase the expression of Nrf2 in Keap1-Nrf2/ARE signaling pathway. At the same time, they could inhibit the expression of the negative feedback gene Keap1 and promote the expression of downstream gene GCLC, Gstt-1, Nqo1 and Ho-1.

Litopenaeus vannamei fermentation using selected Lactobacillus spp. to reduce its allergenicity 用选定的乳杆菌发酵凡纳滨对虾以降低其致敏性

ABSTRACT Shrimp is a common cause of food allergies in allergic individuals. The management of patients suffering from shrimp allergy relies on avoidance of the specific food, which compromises nutritional requirements. In this study, fermentation was performed using Lactobacillus to reduce the allergenicity of Litopenaeus vannamei. The optimal fermentation conditions using Lactobacillus helveticus TS6024 were 1% glucose, 30% minced shrimp, 2.5% inoculum level, 3% NaCl, and 60 h of incubation at 37°C, whereas those using Lactobacillus acidophilus 6005 were 2% glucose, 20% minced shrimp, 15% inoculum level, 2% NaCl, and 48 h of incubation at 42°C. Under optimal conditions, Lactobacillus helveticus TS6024 and Lactobacillus acidophilus 6005 fermentation reduced the allergenicity of shrimp by 78.97% and 70.09%, respectively. SDS-PAGE analysis showed that the band intensity of tropomyosin (36KDa) reduced evidently after the selected Lactobacillus fermentation. The hypoallergenic shrimp/shrimp products will provide optimal nutrition with reduced risk of adverse reactions in hypersensitive individuals.

ABSTRACT Our previous studies reported that viridicatol isolated from the deep-sea-derived fungus Penicillium griseofulvum could regulate the stabilisation of mast cells to relieve food allergy. To understand the molecular role of viridicatol in stabilising mast cells, transcriptomes of viridicatol-treated RBL-2H3 cells were analysed by RNA-sequencing. There were 128 differentially expressed genes in activated RBL-2H3 cells with or without viridicatol treatment. The mast cell activation-related genes were significantly reduced by treatment with viridicatol through RT-qPCR analysis. Moreover, Kyoto Encyclopedia of Genes and Genomes enrichment analysis indicated that viridicatol was important in mast cell stabilisation by affecting MAPK and JAK-STAT signalling pathways. Additionally, molecular docking and western blot analysis revealed that the phosphorylated JNK, ERK, P38, and STAT6 proteins were inhibited by viridicatol. Taken together, viridicatol has the potential to be used as a new type of anti-food allergic functional material via controlling MAPK and JAK-STAT signalling pathways of mast cells.

Simultaneous detection of OTA and AFB1 in cereals based on bispecific monoclonal antibody using quantum dot nanobead lateral flow immunoassay 基于双特异性单克隆抗体的量子点纳米球横向流动免疫法同时检测谷物中OTA和AFB1

ABSTRACT Simultaneous contamination of ochratoxin A (OTA) and aflatoxin B1 (AFB1) in cereals are an ongoing global concern. For this reason, a sensitive method for the simultaneous detection of OTA and AFB1 in soybean, corn, and rice is required. Herein, we prepared a bispecific monoclonal antibody (BsMAb) that specifically recognized OTA and AFB1, and subsequently developed a quantum dot nanobead (QB) lateral flow immunoassay (QB-LFIA) based on BsMAb. The half-inhibitory concentrations (IC50) for OTA and AFB1 detection were 0.73 and 0.01 µg/kg in soybean, 1.14 and 0.03 µg/kg in corn, and 1.69 and 0.07 µg/kg in rice, respectively. The average recoveries of OTA and AFB1 were 80.20–130.05% with coefficients of variation below 11%. The performance of the proposed method was compared with that of traditional LFIA, and higher sensitivity was observed. The BsMAb-based QB-LFIA was sensitive and specific for the simultaneous detection of OTA and AFB1 in cereals.

Crayfish shells-derived carbon dots as a fluorescence sensor for the selective detection of 4-nitrophenol 小龙虾壳碳点作为荧光传感器选择性检测4-硝基苯酚

ABSTRACT As a type of environmentally deleterious compound, 4-nitrophenol (4-NP) has become a global concern due to its extreme toxicity to aquatic organisms and humans. Herein, we used naturally available crayfish shells as precursors to prepare a new type of carbon dots (CDs) by the hydrothermal method. The as-synthesized CDs exhibited strong blue fluorescence emission which can be sensitively quenched by 4-NP through the static quenching effect because of the formation of a ground state complex between CDs and 4-NP. Accordingly, a facile and rapid fluorescence sensor was developed to detect 4-NP from 0 μM to 50 μM with a limit of detection (LOD) of 0.16 μM. Furthermore, the proposed sensor was successfully applied for the detection of 4-NP in fresh crayfish meat and aquatic water samples.

Background: The Azores holds the largest population of Cory's shearwater Calonectrisborealis (Cory, 1881) (Aves, Procellariiformes, Procellariidae) in the world. One of the major threats of this species in the Azores is the predation by invasive mammals, which were introduced during European colonisation of the islands.The present study provides a dataset from a camera-trapping survey performed in colonies of Cory's shearwater. The sampling was conducted between 7 April and 23 October 2019, covering the entire breeding season, in three colonies of the Terceira Island (Azores). A total of 32 nests were sampled using motion-triggered cameras. The aims of this study are to provide information about the ecological patterns of the Cory shearwater and to identify potential nest predators.

New information: Our results include a total of 6972 records of 15 species (nine species of birds, five of mammals and one reptile), of which 5414 records are of Cory's shearwater, 478 of potential mammal predators and 1080 of another vertebrate species. Information about the biology of the species is also provided, as species circadian behaviour and habitat description.

ABSTRACT Shenling Baizhu Powder is a traditional Chinese medicine formula that has been used for thousands of years to invigorate spleen, nourish stomach and regulate immune function. The multi-herbal extracts, an improved formular from Shenling Baizhu Powder, were used in our study to investigate its immune-enhancing effects and underling mechanisms. Based on UPLC-Q-TOF-MS and SPME-GC-MS analysis, 106 chemical compounds were identified from the extracts. In vitro assays indicated that the extracts could promote cell proliferation and NO secretion of RAW264.7 cells. RNA-seq and RT-qPCR showed that RAW264.7 cells treated with extracts increased the expression of pro-inflammatory factors IL-1β, iNOS, TNF-α, and TGF-β. Western-Blot revealed that the extracts could significantly enhance ERK phosphorylation level, and slightly increased JNK and p38 phosphorylation levels. These data suggest that multi-herbal extracts have a potential of regulating immune response via the JNK/p38/ERK signalling of MAPK pathways in RAW264.7 cells. GRAPHICAL ABSTRACT

ABSTRACT Tobacco mosaic virus (TMV) causes a significant yield loss in solanaceous crops. Accurate and rapid diagnosis of TMV is an essential pre-requisite for proactive and tailored disease management strategies, filling the gap between more rigorous laws regarding pesticide use and maintaining crop production. Herein, we developed a time-resolved fluorescence immunoassay (TRFIA) to quantitatively detect TMV in tobacco. TMV virions were used as immunogens to develop monoclonal antibodies, which were then conjugated to Eu3+ nanospheres. The detection limit of the TRFIA platform was 0.24 ng/mL, with recovery rates in the range of 90.71–108.11% in spiked tobacco. The specificity was confirmed using ten species of tobacco-infecting phytopathogens. In-field applicability was implemented by screening for TMV in 10,000 tobacco leaf samples collected from five fields in Kunming, China. Overall, the proposed TRFIA platform is rapid, accurate, and end-user friendly, demonstrating its application in TMV onsite testing in tobacco.
