ABSTRACT Zecicindela tekapoensis sp. nov. (Coleoptera: Carabidae: Cicindelinae) is described. It is placed in the endemic genus Zecicindela Larochelle & Larivière, even though it is the only known species in the genus with a tridentate labrum. The specimens were found on a very dry, barren terrace surface near the Tekapo River, in the Mackenzie Basin.

Three weeks after celebrating his 99th birthday in the company of family and friends, Guillermo (Willy) Kuschel completed a manuscript, had a beer and died peacefully in his sleep on the afternoon of 1 August 2017. It was a fitting end for a formidable man who had lived a full life and had gained a reputation as being all but indestructible. Willy is survived by his wife Beverley, their three children and four grandchildren. Born in Frutillar in southern Chile, Willy was the sixth of 11 children to parents of German heritage. He grew up on the family farm, speaking German and Spanish, and left home to attend boarding school. After completing his high-school education, he obtained a Licentiate in Philosophy in 1940 at the University of Chile. He then entered a seminary in Buenos Aires to become a priest, gaining a Licentiate in Theology in 1944. He learned French, classical Greek and Latin as part of his theological training (English came later). Soon after this, he became involved in biological research. Initially strongly interested in botany, he quickly became fascinated by weevils through their associations with plants, and this led to a Licentiate in Biology and Chemistry. He researched the biology and systematics of water weevils in the genus Lissorhoptrus for a PhD degree in biological sciences, conferred in 1953 and being the first PhD degree awarded by the University of Chile (and for all of Chile). From 1950 he was employed by the university as a research entomologist in the Centro de Investigaciones Zoológicas, and in 1957 he became head of the Department of Entomology. During his time at the University of Chile, he was president of the Sociedad Chilena de Entomología

ABSTRACT Type specimens of six species of the genus Folsomia Willem, 1902 described by J. Salmon were examined. Folsomia pusilla Salmon, 1944 syn. nov. is synonymised with Mucrosomia caeca (Wahlgren, 1906). Synonymy of Folsomia lunata Salmon, 1943 with M. caeca is verified. Three species are transferred from the genus Folsomia to other genera, resulting in Mucrosomia novaezealandiae (Salmon, 1943) comb. nov., Cryptopygus miradentatus (Salmon, 1943) comb. nov. and Proisotoma sedecimoculata (Salmon, 1943) comb. nov. Folsomia parasitica Salmon, 1943 syn. nov. is synonymised with Folsomia candida Willem, 1902. Bagnallela is removed from the list of synonyms of the genus Folsomia. Proisotoma dubia (Deharveng, 1981) comb. nov. gets a new generic position.

Use of shelterbelt pine trees as ‘stepping stones’ by Hylastes ater in agricultural landscapes Hylastes

ABSTRACT Hylastes ater and Hylurgus ligniperda, two exotic pine bark beetles in New Zealand, have recently been collected at locations far from pine plantations or other dead pine host material. We investigated if dispersing beetles are reproducing in the roots and root collars of damaged pine trees of shelterbelts (windbreaks) that are common in New Zealand’s agricultural landscapes. The ability of these bark beetles to breed in roots and root collars has been noted previously, primarily on dead trees and stumps. But to our knowledge it has not been empirically studied in New Zealand if roots of standing but physiologically declining (i.e. dying) trees can serve as suitable breeding material. We used a novel and effective root herbivore emergence trap, which is described here, to carry out this study. A total of 37 H. ater were captured across 13 traps and three sites in inland Canterbury; however, no H. ligniperda were found. We confirm that H. ater is able to colonise roots of damaged and declining isolated pine trees and show that shelterbelts can act as ‘stepping stones’ aiding spread and persistence through the landscape.

Adoptive immunotherapy leveraging chimeric antigen receptor-modified T (CAR-T) cells holds great promise for the treatment of cancer. However, tumor-associated antigens often have low expression levels in normal tissues, which can cause on-target, off-tumor toxicity. Recently, we reported that GPC3-targeted CAR-T cells could eradicate hepatocellular carcinoma (HCC) xenografts in mice. However, it remains unknown whether on-target, off-tumor toxicity can occur. Therefore, we proposed that dual-targeted CAR-T cells co-expressing glypican-3 (GPC3) and asialoglycoprotein receptor 1 (ASGR1) (a liver tissue-specific protein)-targeted CARs featuring CD3ζ and 28BB (containing both CD28 and 4-1BB signaling domains), respectively, may have reduced on-target, off-tumor toxicity. Our results demonstrated that dual-targeted CAR-T cells caused no cytotoxicity to ASGR1+GPC3- tumor cells, but they exhibited a similar cytotoxicity against GPC3+ASGR1- and GPC3+ASGR1+ HCC cells in vitro. We found that dual-targeted CAR-T cells showed significantly higher cytokine secretion, proliferation and antiapoptosis ability against tumor cells bearing both antigens than single-targeted CAR-T cells in vitro. Furthermore, the dual-targeted CAR-T cells displayed potent growth suppression activity on GPC3+ASGR1+ HCC tumor xenografts, while no obvious growth suppression was seen with single or double antigen-negative tumor xenografts. Additionally, the dual-targeted T cells exerted superior anticancer activity and persistence against single-targeted T cells in two GPC3+ASGR1+ HCC xenograft models. Together, T cells carrying two complementary CARs against GPC3 and ASGR1 may reduce the risk of on-target, off-tumor toxicity while maintaining relatively potent antitumor activities on GPC3+ASGR1+ HCC.
