Microbiological and molecular characterization of E. coli and Salmonella isolated from diarrheic calves 从腹泻犊牛中分离出的大肠杆菌和沙门氏菌的微生物学和分子鉴定

Diarrhea is one of the most common fatal disease of neonatal calves regardless of farm welfare. What makes treatment of these cases challenging is the increase in the incidence of multi-drug resistant bacteria, including E. coli and Salmonella. To isolate, identify, molecular characterize of E. coli and salmonella in diarrheic calves, 50 fecal samples were collected and analyzed aseptically. The preliminary result showed that 62% and 14% samples were positive for E. coli and salmonella respectively. Among the E.coli isolates O86:K59, O128:K, O55:K59 (2), O86:K61, O119:K58, O08:K61, O126:K71 serotypes was detected in eight isolates while one isolate was untypeable. Three salmonella serotypes, Salmonella Typhimurium, Salmonella Anatum and Salmonella Florida was detected in the isolated samples . All of the E. coli strains had eaeA genes while 16.7 and 11.1% of them harbored stx1 and stx2 genes respectively. About 71.4% of salmonella isolates were positive for all five pathogenicity island genes from SPI-1 to SPI-5, while SPI-1, SPI-2 and SPI-5 were detected in 14.3% and SPI-2, SPI-3, SPI-4 and SPI-5 were positive in 14.3%. All E. coli strains were resistant to Ampicillin, Amoxicillin/Clavulanic, Cefazolin and Aztreonam., and all Salmonella serovars were resistant to Cefazolin, Chloramphenicol, Gentamycin, Kanamycin and Aztreonam. The present study identified multidrug resistant E. coli and salmonella as the common pathogen of calf diarrhea in the study area and the pathogens harbored common virulence markers of human diarrheic strains which might cause a food borne outbreak in the future.

Copper oxide nanoparticles (CuO NPs) were green synthesized using the aqueous extract of banana peels. CuO NPs were characterized by X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy, transmission electron microscopy (TEM), Brunauer-Emmett-Teller (BET) surface area analysis. Analyses confirmed the formation of crystalline CuO with 15 nm particle size and 9 m 2 g -1 surface area. The current study is an endeavor to explore the in vivo impact of acute different oral doses of the synthesized CuO NPs. One hundred and fifty adult male albino rats were used, they divided into 5 groups, each group contained 3 replicates (n = 30). Rats were administered suspension of CuO NPs using oral gavage in different concentrations (0, 100, 200,1000, and 2000 mg/kg) as a single dose. Liver and kidney function markers (AST, ALP, ALP, urea and creatinine) as well as antioxidant enzymes (catalase and glutathione reductase) were evaluated 2, 8 and 15 days after administration. The results revealed a high mortality rate with rats receiving 1000 and 2000 mg/kg (16% and 30.7%, respectively). Further, within the same high doses, rats showed marked elevation of serum AST, ALP, ALP, urea and creatinine after 2, 8 and 15 days from administration. However, the activity of catalase enzyme declined significantly at the same timepoints with the former high doses. Moreover, the activity of glutathione reductase enzyme in the liver homogenate showed marked decrease after administration of all CuO NPs doses. In conclusion, the harmful effect of CuO NPs appears with the higher dose so, attention should be considered during use of CuO NPs as nutritive supplements.

Microbiological and molecular characterization of Vibrio cholera and Vibrio parahaemolyticus isolated from Tilapia fish (Oreochromis niloticus 罗非鱼霍乱弧菌和副溶血性弧菌的微生物学和分子特性研究

Vibrio species are zoonotic pathogens that can affect humans by different routes such as ingestion or contact causing gastrointestinal diseases and wound infection. This study aimed to detect V. cholera and V. parahaemolyticus in Nile Tilapia fish collected from Aswan Governorate, Egypt. A total of 52 muscle samples were collected from Tilapia fish and were subjected to microbiological and molecular characterization. Alkaline peptone water media was used for the enrichment of the samples then followed by inoculation onto thiosulfate citrate bile salt sucrose (TCBS) agar media for the isolation of Vibrio species, biochemical tests were performed to identify V. cholera and V. parahaemolyticus , and then they were confirmed by Vibrio genus-specific gene and virulence genes by PCR . Out of 52 fish muscle samples (Nile Tilapia) 32 appeared as yellow colonies, 2 samples showed green colonies and 18 samples showed mixed yellow and green on TCBS agar. Only 40 samples show biochemically positive for Vibrio species. 15 random samples were amplified to 16srRNA gene by PCR technique for more accurate identification resulting in 14 isolates being positive to 16srRNA, 8 isolates positive for sodB gene-specific of V. cholera , and 6 isolates positive for toxR gene-specific of V. parahaemolyticus . Out of 8 V. cholera isolates were found high resistance rate to amoxiclav (87.5%), gentamicin (75%), chloramphenicol (50 %) and ampicillin (50 %). Conversely, (50 %) sensitive to tetracycline, ciprofloxacin and, trimethoprim-sulphamethoxazole, and the average MRA of V. cholera was 0.48. On the other hand, all 6 V. parahaemolyticus isolates were resistant to amoxiclav (100%), tetracycline (100%), gentamicin (100%) and chloramphenicol (100%) However, all isolates sensitive to trimethoprim-sulphamethazole (100%) but half of them sensitive to ciprofloxacin (50%) and ampicillin (50%), and the average MRA of V. parahaemolyticus was 0.59. In conclusion, this study showed that V. cholera is the most dominant pathogenic one then followed by V. parahaemolyticus in Nile tilapia fish resulting in economic losses and causing public health problems.

This study aimed to detect V. cholera and V. parahaemolyticus in Nile Tilapia fish collected from Aswan Governorate, Egypt. A total of 52 muscle samples were collected from Tilapia fish and were subjected to microbiological and molecular characterization. Alkaline peptone water media was used for the enrichment of the samples then followed by inoculation onto thiosulfate citrate bile salt sucrose (TCBS) agar media for the isolation of Vibrio species, biochemical tests were performed to identify V. cholera and V. parahaemolyticus , and then they were confirmed by Vibrio genus-specific gene and virulence genes by PCR . Out of 52 fish muscle samples (Nile Tilapia) 32 appeared as yellow colonies, 2 samples showed green colonies and 18 samples showed mixed yellow and green on TCBS agar. Only 40 samples show biochemically positive for Vibrio species. 15 random samples were amplified to 16srRNA gene by PCR technique for more accurate identification resulting in 14 isolates being positive to 16srRNA, 8 isolates positive for sodB gene-specific of V. cholera , and 6 isolates positive for toxR gene-specific of V. parahaemolyticus . Out of 8 V. cholera isolates were found high resistance rate to amoxiclav (87.5%), gentamicin (75%), chloramphenicol (50 %) and ampicillin (50 %). Conversely, (50 %) sensitive to tetracycline, ciprofloxacin and, trimethoprim-sulphamethoxazole, and the average MRA of V. cholera was 0.48.
