In this study, the closely linked fluorescence molecular marker PM-Xa7 of Xa7 gene was developed on the basis of sequence difference between the resistant variety ‘IRBB7’ containing Xa7 and other nine bacterial blight susceptible rice varieties, and combine with PARMS technology (Penta-primer amplification refractory mutation system). Genotypes of Xa7 were identified in 72 rice germplasm and the bacterial blight resistance level of ‘Mei B’ was improved by backcross and screening with this marker. The results showed that two rice germplasms containing Xa7 were screened out from 72 rice germplasm; By introducing the resistant Xa7 gene from ‘IRBB7’ into ‘Mei B’ with PM-Xa7, a stable new maintainer with resistance to bacterial blight was successfully obtained. The bacterial blight resistant fluorescence signal was detected in 289 plants and the bacterial blight susceptible fluorescence signal was detected in 93 plants of BC 2 F 2 population generated from ‘IRBB7’ and ‘Mei B’, and the genotypes were consistent with the phenotypes of Xoo inoculation. These research results indicated that PM-Xa7 can be effectively used for the marker-assisted breeding of bacterial blight resistant rice variety with the resistant Xa7 gene.

Small gourd ( Lagenaria siceraria var . microcarpa (Naud.) Hara), a cultivar of Lagenaria family, is an important medicinal and ornamental plants. However, there are few studies on its chloroplast (cp) genome. In this study, we characterized its complete chloroplast (cp) genome with high throughput sequencing. The circular cp genome is determined to be 157, 121 bp in length, with a long sing-copy and a small sing-copy separated by a pair of inverted repeat regions. A total of 132 genes were annotated, including 86 protein-coding genes, 37 tRNA genes, 8 rRNA genes, and 1 pseudo gene. The overall GC content of cp genome was 37.09%. The maximum likelihood tree suggested that small gourd was closely related to Lagenaria siceraria. These results provided an understanding of small gourd cp genome and might be useful for future genetic studies.

Cucumis melo L. var . agrestis Naud. belong to the genus Cucumis in the Gourd family, it is rich in oil. It can extract high quality edible oil and provide raw materials for medicine and food industry. The chloroplast genome of Cucumis melo L. was sequenced for the first time. The results showed that the chloroplast genome of Cucumis melo L. was similar to that of common melon. This study can provide a theoretical reference for breeding.

Oil plants are not only used as essential nutriments in food but also used broadly as ingredients of industrial products. Cornus wilsoniana Wanger is an important native woody oil plant with high-yield and high-oiliness characters in China. The fruit oil of Cornus wilsoniana not only could be used as equilibrated dietary oil, but also has hypolipidemic function and helps to overcome EFA deficiency. Genomic information is currently not available for Cornus wilsoniana , which will therefore affect its genetic improvement process. In this study, 8713 EST-SSRs were identified from the transcriptome sequencing of Cornus wilsoniana . Most of these EST-SSRs were composed of dimer and trimer repeats. The AG/CT motif is the most common dimeric EST-SSRs motif (53.5%), whereas the CG/CG (0.44%) microsatellites are present only at low abundances. Among the trimeric microsatellites, AAG/CTT (5.53%) and ATC/ATG (4.27%) are the most common motifs. There are no obvious dominant motifs among the tetra-, penta-, and hexa- nucleotide motifs. Fifteen pairs of EST-SSRs primers are developed with polymorphism and used for the genetic diversity analysis of twenty genotypes of Cornus wilsoniana with high fruit output. Allele number per locus ranges from 2 to6 with an average of 2.87, and the PIC value ranges from 0.05 to 0.58 with an average of 0.36. Average genetic diversity overall SSR loci for the 20 genotypes was 0.427, ranging from 0.049 to 0.659. All the loci are polymorphic and clearly distinguish the genotypes. Cluster analyze (NJ tree, UPGMA) identifies a similar pattern of variation. 15developed EST-SSRs are informative, codominant and reliable, and could be applied in the improvement programs of Cornus wilsoniana in the future.

Analytic hierarchy process (AHP) was used to distribute the weight of 10 evaluation indexes of flue-cured tobacco varieties of regional trials in Sichuan province in 2016 for dynamic technique for order preference by similarity to ideal solution (DTOPSIS), membership function was used for optimizing dimensionless treatment of chemical evaluation indexes of DTOPSIS, and then DTOPSIS was used for comprehensive evaluation of varieties. The weight of yield, proportion of superior tobacco, output value, leaf number, stem circumference, plant height, nicotine content, total sugar content, potassium content and black shank disease index was 0.090 2, 0.235 1, 0.301 4, 0.035 1, 0.021 7, 0.021 7, 0.057 1, 0.035 1, 0.035 1 and 0.167 6, respectively. The comprehensive evaluation order of new varieties in planting areas in Southwest Sichuan was ‘Yunyan87’>‘SCT01’>‘Hongda’>‘K326’>‘09011’>‘X4’>‘X3’, the order in South Sichuan was ‘K326’>‘Yunyan87’>‘09011’>‘SCT01’>‘X3’>‘X4’, and the order in North Sichuan was ‘09011’>‘Yunyan87’>‘X4’>‘SCT01’>‘X3’. The results of this study show that further experiments and demonstrations of ‘SCT01’ could be conducted in plant areas in Southwest Sichuan, '09011' has the potential to be used as a good variety in plant areas in North Sichuan, and the corresponding cultivation measures of ‘X3’ and ‘X4’ should be improved. This research would provide scientific basis for the screening, identification and promotion of new flue-cured tobacco varieties, and enrich the evaluation system of new flue-cured tobacco varieties.

In order to establish the plant regeneration system of Glycyrrhiza mongolicum and Glycyrrhiza Xinjiang, this study used hypocotyls, leaves, cotyledons and stems to inducing callus and regenerated plant, and it screen out the most suitable explants and the best medium for plant regeneration. The experimental results showed that MS+2.0 mg/L NAA+2.0 mg/L 6-BA was the best medium for callus induction and plant regeneration. Among these, hypocotyls and cotyledons are the best explants for callus induction. The tissue induction rate is higher than 90%. The rapid propagation system established in this study can provide the theoretical reference and practical guidance for the breeding and production of fine Glycyrrhiza uralensis Fisch. varieties.

Garden asparagus ( Asparagus officinalis L.) is a kind of nutritious and healthy vegetable with important economic value in the world. China has become the largest production country of garden asparagus in the world. But the seeds mainly depend on import, so we need to innovate breeding system and improve the international competitiveness of domestic asparagus varieties. Genetic analysis demonstrated that the genetic variation of garden asparagus was low and its heterosis was weak among cultivars. Inter-specific hybridization is an important means to utilize heterosis, but the relationship between the classical taxonomy and molecular phylogeny of the genus Asparagus is disordered, which limits the development of interspecific hybridization breeding. In this genus, thirty-one species have been found in China, which is an effective genetic resource for the utilization of heterosis among Asparagus species. In order to provide reference for the next step to clarify the phylogenetic relationship of this genus and to carry out the interspecific hybrid breeding of garden asparagus in the near future, the research progress of its classical and molecular phylogeny of this genus is reviewed in this study.

In order to improve the rooting rate of Qiuzi pear tissue culture seedlings, this study took ‘sorb’ as experimental material, based on Qiuzi pear rooting tissue culture medium (1/2MS+IBA 0.5 mg/L), and added different concentrations of activated carbon (AC): 0.25 g/L, 0.50 g/L, 0.75 g/L, 1.00 g/L. The results showed that the rooting rate, rooting number and root length of tissue culture seedlings cultured with activated carbon were significantly higher than those of the control group. The most suitable medium for rooting Qiuzi pear was 1/2MS+IBA 0.5 mg/L+AC 1.00 g/L, and the rooting rate of the treated ‘sorb’ tissue culture seedlings reached 90%, which could obviously promote the growth of Qiuzi pear tissue culture. The results provide a theoretical basis for improving the rooting rate and survival rate of tissue culture seedlings of pear plants, and provide a reference for the establishment of rapid propagation system of woody plants.
