To stablish sperm fertilization capacity membrane integrity and functionality are important characteristics to determine. The objectives of the present study were: 1) to evaluate the functionality of the plasma membrane over time by the hypoosmotic test (HOS) and 2) combined the HOS test with Coomassie Blue (CB) staining to simultaneously evaluate the functionality of membrane and acrosomal integrity in raw llama sperm. A total of 12 ejaculates from 6 adult llama males were collected using electroejaculation under general anesthesia. Hypoosmotic swelling test was performed with and without fixation in paraformaldehyde and the samples were evaluated over time (0 h without fixation and 0; 2; 4; and 24 h when fixed). To perform the CB stain one aliquot of the fixed samples was used. No significant differences (p>0.05) were observed in the percentage of sperm with membrane function when comparing fixed samples analyzed over time versus the samples not fixed. Also, no statistical differences (p>0.05) were observed on membrane functionality between the two protocols used; HOS test versus HOS combined with the CB stain. It is possible to differ the moment of the evaluation of the membrane functionality of llama spermatozoa using the HOS technique from the semen collection. The use of the dual technique (HOS + CB) allow the evaluation of sperm membrane function and acrosome status in raw llama semen with only one technique that can be applied on the field.

Bovine embryos that rapidly reach the two-cell stage have higher probabilities of becoming viable blastocysts. Our objective was to evaluate the effect of time of the first embryonic division on the development rate of bovine blastocysts produced in vitro and also to determine embryo viability by differential staining of living and dead blastomeres. In vitro embryos divided at 36h (early) and 48h (late) post fertilization were separated and in vitro cultured for the remaining time (up to 7 days). Blastocyst rates from the groups of embryos divided in two-cells at 36h and 48h, corresponding to three repetitions, were 38.7% and 23.2%, respectively, showing statistically significant differences (p=0.0168). However, the proportion of living cells assessed by differential staining of the 36h and 48h groups were 93.7% and 88.2%, respectively, showing no significant differences. Although the results indicate that an early embryo splitting favors a high rate of blastocysts, separation management of the embryos causes stress reducing the total number of blastocysts compared to our control group without separation of embryos two-cell.

The use of reproductive technologies in the reproduction of the domestic animals has different objectives, between the most important uses are the prevention of diseases transmitted via natural mating, the optimization of the reproductive management, the more economic interchange of genetic material, the development of more efficient animal production systems, the generation of offspring with predetermined sex and genetic composition according to market requirements, the conservation of genetic resources and the increment of the rate of genetic gain in the animal populations; being this last application the one that has the most frequent and important usage and the most significant impact in the animal production industry. The importance of the reproductive technologies in animal improvement is because it allows us to achieve a significant increment in the reproductive rate of selected individuals (to get the maximum number of offspring per breeding animal), this makes possible to obtain high intensity and precision of selection and at the earliest age possible (reducing generation interval) resulting in the highest genetic gain per unit of time. From all the reproductive technologies developed, the artificial insemination (AI) in cattle is the most widely used technology globally and at national level in Peru, followed by the embryo transfer (ET) in cattle and by the AI and ET in sheep. From the design and usage of the in vitro embryo production and methodologies for the embryo micromanipulation it has started the development of a series of advanced reproductive technologies from a simple embryo bisection up to cloning of individuals, reproduction using stem cells and production of quimeras with cells genetically modified. In Peru is only in the last decade that the research and development and commercial application of reproductive technologies in the livestock reproduction has started with some advances in in vitro embryo production, embryo transfer, animal cloning, etc. with visible impact in the animal production industry and generating many challenges in order to increase its efficiencies and achieve its commercial use.
