Separation and Identification of Urinary Proteins Following Digital Rectal Examination in Patients with Benign Prostatic Hypertrophy 良性前列腺肥大患者直肠指检后尿蛋白的分离与鉴定

Specimens of urine were obtained before and after digital rectal examination from four patients with benign prostatic hypertrophy (BPH) for comparative analysis of protein components by reversed-phase high performance liquid chromatography. Four characteristic peaks were detected in urine after compared with before the physical examination, and their molecular masses on SDS-polyacrylamide gel electrophoresis were 16, 16, 34 and 46 kDa. After the proteins were reduced, S-pyridylethylated and cleaved with cyanogen bromide, the amino-terminal amino acids were sequenced and a homology search was conducted. The two 16-kDa proteins were both identified as fl-microseminoprotein. The 34-kDa protein was identified as a prostate-specific antigen, and the 46-kDa protein was a Zn-a2-glycoprotein. The present findings provide important information on pre-analytical sampling of urine for the diagnosis of BPH.

Comparison of Recent Methods for Amylase Activity Measurement Using Synthetic Substrates 合成底物测定淀粉酶活性的最新方法比较

Recently, several methods for measuring the amylase activity have been developed using new synthetic substrates. These methods can detect released chromophores such as 4-nitrophenol (PNP), 2-chloro-4-nitrophenol (CNP), and NADH. We compared ten such methods with the blue dye-linked starch polymer (Blue Starch) method. The intra-run precision (CV) of each method, 0.4-3.3%, was acceptable. The influence from coexisting materials was negligible in each method, and the correlation among the methods was good. There was no problem with the stability of any reagents after preparation. Comparing the reactivities of pancreatic and salivary amylases as a ratio (P/S), the differences among methods became obvious.
