This experiment optimized the extraction and detection methods of biosurfactant produced by the strain Dietzia sp. DQ12-45-1b, and studied biosurfactant products of the strain growing on n-hexadecane for different cultural time. Ethyl acetate was the best extraction solvent with the highest metabolite recovery effi cient. The mixture of hexane : ammonia : isopropanol = 2:1:4 (V:V:V) was used as the best solvent system in thin layer chromatography analysis. The strain DQ12-45-1b produced glycolipid biosurfactant, with the biosurfactant concentration of the culture broth positively correlated with the incubation time. The content ratio of crude extracts in emulsifi ed hexadecane layer and aqueous medium was 4.16:1. A glycolipid was detected in the emulsifi ed hexadecane layer but not in aqueous phase. Abstract This study analyzed the microbial community structure, microorganisms and the oil-degrading activities in the dehydrated crude oil from Daqing Oil fi eld. The results showed the existence of many microbial cells in the dehydrated crude oil, and screened a microbial community with oil-emulsifying and oil-dispersing activity. Community analysis showed that the types and rations of the microorganisms in the community changed with incubation time. All together 166 bacteria were isolated and puri fi ed from the crude oil culture system, up to 13.1% of the tested strains having oil-dispersing or oil-emulsifying activities. The results showed that there are microbial communities with oil emulsifying and oil dispersing activities in Daqing oil reservoir, that the harsh conditions and the lack of nutrition leads to smaller community density, and that the oil degrading strains occupy a high proportion.
